Abstract

Targeted therapeutics that localize to a specific subcellular compartment such as the lysosome are provided. The targeted therapeutics include a therapeutic agent and a targeting moiety that binds a receptor on an exterior surface of the cell, permitting proper subcellular localization of the targeted therapeutic upon internalization of the receptor. Nucleic acids, cells, and methods relating to the practice of the invention are also provided.

Claims (17)

1. An underglycosylated targeted therapeutic fusion protein comprising: a lysosomal enzyme, wherein the lysosomal enzyme is selected from the group consisting of: acid-α1,4-glucosidase; β-galactosidase; β-hexosaminidase A; β-hexosaminidase B; α-galactosidase A; glucocerebrosidase; arylsulfatase A; galactosylceramidase; acid sphingomyelinase; acid ceramidase; acid lipase; α-L-iduronidase; iduronate sulfatase; heparan N-sulfatase; α-N-acetylglucosaminidase; acetyl-CoA-glucosaminide acetyltransferase; N-acetylglucosamine-6-sulfatase; galactosamine-6-sulfatase; β-galactosidase; arylsulfatase B; β-glucuronidase; α-mannosidase; β-mannosidase; α-L-fucosidase; N-aspartyl-β-glucosaminidase; α-neuraminidase; lysosomal protective protein; α-N-acetyl-galactosaminidase; N-acetylglucosamine-l-phosphotransferase; and palmitoyl-protein thioesterase; and a lysosomal targeting domain that binds an extracellular domain of human cation-independent mannose-6-phosphate receptor in a mannose-6-phosphate-independent manner, wherein the lysosomal targeting domain is mature human IGF-II or a mutein of mature human IGF-II having an amino acid sequence at least 70% identical to mature human IGF-II.

2. The targeted therapeutic fusion protein of claim 1 , wherein the targeted therapeutic fusion protein is deglycosylated.

3. The targeted therapeutic fusion protein of claim 2 , wherein the targeted therapeutic fusion protein is deglycosylated by treatment with periodic acid.

4. The targeted therapeutic fusion protein of claim 1 , wherein the lysosomal targeting domain is mature human IGF-II.

5. The targeted therapeutic fusion protein of claim 1 , wherein the lysosomal targeting domain is a mutein of mature human IGF-II having an amino acid sequence at least 70% identical to mature human IGF-II.

6. The targeted therapeutic fusion protein of claim 1 , wherein the lysosomal enzyme is β-hexosaminidase A.

7. The targeted therapeutic fusion protein of claim 1 , wherein the lysosomal enzyme is α-galactosidase A.

8. The targeted therapeutic fusion protein of claim 1 , wherein the lysosomal enzyme is arylsulfatase A.

9. The targeted therapeutic fusion protein of claim 1 , wherein the lysosomal enzyme is galactosylceramidase.

10. The targeted therapeutic fusion protein of claim 1 , wherein the lysosomal enzyme is acid sphingomyelinase.

11. The targeted therapeutic fusion protein of claim 1 , wherein the lysosomal enzyme is acid lipase.

12. The targeted therapeutic fusion protein of claim 1 , wherein the lysosomal enzyme is α-L-iduronidase.

13. The targeted therapeutic fusion protein of claim 1 , wherein the lysosomal enzyme is iduronate sulfatase.

14. The targeted therapeutic fusion protein of claim 1 , wherein the lysosomal enzyme is heparan N-sulfatase.

15. The targeted therapeutic fusion protein of claim 1 , wherein the lysosomal enzyme is α-N-acetylglucosaminidase.

16. The targeted therapeutic fusion protein of claim 1 , wherein the lysosomal enzyme is β-glucuronidase.

17. The targeted therapeutic fusion protein of claim 1 , wherein the lysosomal enzyme is acid-α1,4-glucosidase.
