Abstract

In one aspect, the invention is directed to polypeptides having an amylase activity, polynucleotides encoding the polypeptides, and methods for making and using these polynucleotides and polypeptides. In one aspect, the polypeptides of the invention can be used as amylases, for example, alpha amylases, to catalyze the hydrolysis of starch into sugars. In one aspect, the invention provides delayed release compositions comprising a desired ingredient coated by a latex polymer coating.

Claims (66)

1 . An isolated, synthetic, or recombinant nucleic acid comprising:

(c) the amino acid sequence of (a) or (b) having amylase activity but lacking a signal sequence or a carbohydrate binding module; or

(d) the amino acid sequence of (a), (b), or (c) further comprising a heterologous sequence.

3 . A method of producing a polypeptide comprising the steps of introducing a nucleic acid encoding the polypeptide into a host cell under conditions that allows expression of the polypeptide, and recovering the polypeptide, wherein the polypeptide is selected from the group consisting of:

(a) a polypeptide as set forth in claim 2 ; and,

(b) a polypeptide encoded by a nucleic acid as set forth in claim 1 .

4 . A method of hydrolyzing a polysaccharide or oligosaccharide linkage comprising contacting a substance containing the polysaccharide or oligosaccharide with a polypeptide comprising:

(a) a polypeptide encoded by a nucleic acid as set forth in claim 1 , or

(b) a polypeptide as set forth in claim 2 ,

under conditions which facilitate the hydrolysis of the polysaccharide or oligosaccharide linkage; and,

wherein optionally, the method further comprises use of a second polypeptide comprising a second amylase, an alpha amylase, a beta amylase, or another enzyme.

5 . A method of catalyzing the breakdown of a starch, comprising the step of contacting a sample containing starch with a polypeptide comprising:

(a) a polypeptide encoded by a nucleic acid as set forth in claim 1 , or,

(b) a polypeptide as set forth in claim 2 ,

under conditions which facilitate the breakdown of the starch,

wherein optionally, the starch is isolated or derived from rice, germinated rice, corn, barley, wheat, legumes, sweet potato, mild, sorghum, rye, bulgher, or a combination thereof; and,

wherein optionally, the method further comprises use of a second polypeptide comprising a second amylase, an alpha amylase, a beta amylase, or another enzyme.

6 . A composition comprising a polypeptide selected from the group consisting of:

(a) a polypeptide encoded by a nucleic acid as set forth in claim 1 ; and,

(b) a polypeptide as set forth in claim 2 .

7 . A cloning vector or an expression vector comprising a sequence selected from the group consisting of:

(a) a sequence as set forth in claim 1 ; and,

(b) a sequence encoding a polypeptide as set forth in claim 2 .

8 . A host cell comprising a sequence selected from the group consisting of:

(a) a nucleic acid sequence as set forth in claim 1 ; and,

(b) a nucleic acid sequence encoding a polypeptide set forth in claim 2 ; and,

wherein optionally, the host is a prokaryote, eukaryote, fungus, yeast, plant, or a metabolically rich host cell.

9 . A method for liquefying a starch containing composition comprising contacting the starch with a polypeptide of claim 2 or a polypeptide encoded by a nucleic acid as set forth in claim 1 , wherein the polypeptide has amylase activity; and,

wherein optionally, the method further comprises use of a second polypeptide comprising a second amylase, an alpha amylase, a beta amylase, or another enzyme.

10 . A method for washing an object comprising contacting said object with a polypeptide of claim 2 or a polypeptide encoded by a nucleic acid as set forth in claim 1 , wherein the polypeptide has amylase activity, under conditions sufficient for said washing; and,

wherein optionally, the method further comprises use of a second polypeptide comprising a second amylase, an amylase, a beta amylase, or another enzyme.

11 . A method for starch liquefaction comprising contacting said starch with a polypeptide of claim 2 or a polypeptide encoded by a nucleic acid as set forth in claim 1 , wherein the polypeptide has amylase activity, under conditions sufficient for said liquefaction; and,

wherein optionally, the method further comprises use of a second polypeptide comprising a second amylase, an alpha amylase, a beta amylase, or another enzyme.

12 . A detergent additive comprising a polypeptide of claim 2 or a polypeptide encoded by a nucleic acid as set forth in claim 1 wherein the polypeptide has amylase activity.

13 . A method for producing a high-maltose or a high-glucose syrup or a mixed syrup comprising:

liquefying starch using an effective amount of a polypeptide of claim 2 or a polypeptide encoded by a nucleic acid as set forth in claim 1 , wherein the polypeptide has alpha amylase activity, to obtain a soluble starch hydrolysate; and,

saccharifying the soluble starch hydrolysate, thereby resulting in a syrup; and,

wherein optionally, the starch is from a material selected from the group consisting of rice, germinated rice, coin, barley, wheat, legumes, and sweet potato; and,

wherein optionally, the method further comprises use of a second polypeptide comprising a second amylase, an alpha amylase, a beta amylase, or another enzyme.

14 . A method of increasing the flow of production fluids from a subterranean formation by removing a viscous, starch-containing; fluid formed during production operations and found within the subterranean formation which surrounds a completed well bore comprising:

allowing production fluids to flow from the well bore; reducing the flow of production fluids from the formation below expected flow rates;

formulating an enzyme treatment by blending together an aqueous fluid and a polypeptide of claim 2 or a polypeptide encoded by a nucleic acid as sot forth in claim 1 , wherein the polypeptide has alpha amylase activity;

pumping the enzyme treatment to a desired location within the well bore; and allowing the enzyme treatment to degrade the viscous, starch-containing fluid; wherein the enzyme treatment is effective to attack glucosidic linkages in the starch-containing fluid; and,

wherein optionally, the method further comprises use of a second polypeptide comprising a second amylase, an alpha amylase, a beta amylase, or another enzyme.

15 . A method for making an alcohol comprising use of a polypeptide of claim 2 or a polypeptide encoded by a nucleic acid as set forth in claim 1 ; and,

wherein optionally, the method comprises contacting the polypeptide with a starch-comprising composition; and,

wherein optionally, the method further comprises a second polypeptide comprising a second amylase, an alpha amylase, a beta amylase, or another enzyme; and,

wherein optionally, the alcohol comprises a fuel ethanol.

16 . A baking process comprising use of a polypeptide of claim 2 or a polypeptide encoded by a nucleic acid as set forth in claim 1 ; and

wherein optionally, the baking process further comprises use of a second polypeptide comprising a second amylase, an alpha amylase, a beta amylase, or another enzyme.

17 . A drilling process comprising use of a polypeptide of claim 2 or a polypeptide encoded by a nucleic acid as set forth in claim 1 , and;

wherein optionally, the drilling process further comprises use of a second polypeptide comprising a second amylase, an alpha amylase, a beta amylase, or another enzyme.

18 . A brewing process comprising use of a polypeptide of claim 2 or a polypeptide encoded by the nucleic acid as set forth in claim 1 ; and,

wherein optionally, the process further comprises use of a second polypeptide comprising a second amylase, an alpha amylase, a beta amylase, or another enzyme.

19 . A method for making a beverage comprising use of a polypeptide of claim 2 or a polypeptide encoded by a nucleic acid as set forth in claim 1 ; and,

wherein optionally, the method further comprises use of a second polypeptide comprising a second amylase, an alpha amylase, a beta amylase, or another enzyme.

20 . A dough, a bakery product, a baking premix, a baking additive, or a baked product comprising a polypeptide of claim 2 or a polypeptide encoded by a nucleic acid as set forth in claim 1 , and;

wherein optionally, the dough, bakery product, baking premix, baking additive, or baked product further comprises a second polypeptide comprising a second amylase, an alpha amylase, a beta amylase, or another enzyme.
