Enzyme quantification
The invention generally relates to methods for quantifying an amount of enzyme molecules. Systems and methods of the invention are provided for measuring an amount of target by forming a plurality of fluid partitions, a subset of which include the target, performing an enzyme-catalyzed reaction in the subset, and detecting the number of partitions in the subset. The amount of target can be determined based on the detected number.
1. A method for identifying components of a chemical reaction, the method comprising:
(a) forming a plurality of fluid partitions each comprising a particle and a plurality of unbound fluorescently labeled binders specific to a marker having a first concentration;
(b) incubating the plurality of fluid partitions, wherein the unbound fluorescently labeled binders bind to the particle comprising the marker in at least a first fluid partition and wherein the unbound fluorescently labeled binders do not bind to a particle lacking the marker in at least a second fluid partition;
(c) flowing the plurality of fluid partitions through a channel having a narrowed portion adjacent to a detector, causing the fluid partitions to elongate when they pass by the detector;
(d) detecting fluorescence as a function of time as the plurality of fluid partitions pass the detector, wherein the first fluid partition produces a localized spike of fluorescence and the second fluid partition produces a dispersed low-level fluorescence; and
(e) distinguishing, based on the detection step, that the first partition is positive for the marker and the second fluid partition is negative for the marker.
2. The method of claim 1 , wherein the labeled binders are detected by an optical property.
3. The method according to claim 1 , further comprising determining an amount of the bound labeled binders.
4. The method of claim 1 , wherein said fluid partitions are droplets.
5. The method according to claim 4 , wherein the droplets are surrounded by an immiscible carrier fluid.
6. The method of claim 4 , wherein each of the droplets is formed by merging a first droplet comprising the particle with a second droplet comprising a plurality of labeled binders.
7. The method of claim 6 , further comprising flowing the droplets through a narrow channel that causes the droplets to elongate.
8. The method according to claim 1 , further comprising determining an amount of the bound labeled binders based upon a ratio of a localized increase in signal intensity to partition wide decrease in signal intensity in the at least one fluid partition.
9. The method of claim 1 , wherein the detecting is measured by a laser.
10. The method of claim 1 , wherein the particle is a cell or a bead.
11. The method of claim 1 , wherein the labeled binders identify the marker.
12. The method of claim 1 , wherein the labeled binders comprise a plurality of differently colored labels.
13. The method of claim 1 , wherein the partitions comprise multiple copies of a labeled binder that each includes a different fluorescent label.
14. The method of claim 1 , further comprising sorting the first fluid partition from the second fluid partition based on the differentiation in the detecting step.
