Abstract

The present invention provides azetidinyl phenyl, pyridyl, or pyrazinyl carboxamide derivatives, as well as their compositions and methods of use, that modulate the activity of Janus kinase (JAKs) and are useful in the treatment of diseases related to the activity of JAK including, for example, inflammatory disorders, autoimmune disorders, cancer, and other diseases.

Claims (44)

1. A compound, which is 4-{3-(cyanomethyl)-3-[4-(7H-pyrrolo[2,3-d]pyrimidin-4-yl)-1H-pyrazol-1-yl]azetidin-1-yl}-2,5-difluoro-N-[(1S)-2,2,2-trifluoro-1-methylethyl]benzamide, or a pharmaceutically acceptable salt thereof.

2. A composition comprising a compound according to claim 1 , or a pharmaceutically acceptable salt thereof, and a pharmaceutically acceptable carrier.

3. A compound, which is 4-{3-(cyanomethyl)-3-[4-(7H-pyrrolo[2,3-d]pyrimidin-4-yl)-1H-pyrazol-1-yl]azetidin-1-yl}-2,5 -difluoro-N-[(1S)-2,2,2-trifluoro-1-methylethyl]benzamide.

4. A method of treating multiple sclerosis in a patient in need thereof, comprising administering to said patient a therapeutically effective amount of the compound of claim 1 , or a pharmaceutically acceptable salt thereof.

5. A method of treating type I diabetes in a patient in need thereof, comprising administering to said patient a therapeutically effective amount of the compound of claim 1 , or a pharmaceutically acceptable salt thereof.

6. A method of treating lupus in a patient in need thereof, comprising administering to said patient a therapeutically effective amount of the compound of claim 1 , or a pharmaceutically acceptable salt thereof.

7. A method of treating myasthenia gravis in a patient in need thereof, comprising administering to said patient a therapeutically effective amount of the compound of claim 1 , or a pharmaceutically acceptable salt thereof.

8. A method of treating myocarditis in a patient in need thereof, comprising administering to said patient a therapeutically effective amount of the compound of claim 1 , or a pharmaceutically acceptable salt thereof.

9. A method of treating contact dermatitis in a patient in need thereof, comprising administering to said patient a therapeutically effective amount of the compound of claim 1 , or a pharmaceutically acceptable salt thereof.

10. A method of treating psoriasis in a patient in need thereof, comprising administering to said patient a therapeutically effective amount of the compound of claim 1 , or a pharmaceutically acceptable salt thereof.

11. A method of treating osteoporosis in a patient in need thereof, comprising administering to said patient a therapeutically effective amount of the compound of claim 1 , or a pharmaceutically acceptable salt thereof.

12. A method of treating bone resorption in a patient in need thereof, comprising administering to said patient a therapeutically effective amount of the compound of claim 1 , or a pharmaceutically acceptable salt thereof.

13. A composition comprising a compound according to claim 3 and a pharmaceutically acceptable carrier.

14. A method of treating multiple sclerosis in a patient in need thereof, comprising administering to said patient a therapeutically effective amount of the compound of claim 3 .

15. A method of treating type I diabetes in a patient in need thereof, comprising administering to said patient a therapeutically effective amount of the compound of claim 3 .

16. A method of treating lupus in a patient in need thereof, comprising administering to said patient a therapeutically effective amount of the compound of claim 3 .

17. A method of treating myasthenia gravis in a patient in need thereof, comprising administering to said patient a therapeutically effective amount of the compound of claim 3 .

18. A method of treating myocarditis in a patient in need thereof, comprising administering to said patient a therapeutically effective amount of the compound of claim 3 .

19. A method of treating contact dermatitis in a patient in need thereof, comprising administering to said patient a therapeutically effective amount of the compound of claim 3 .

20. A method of treating psoriasis in a patient in need thereof, comprising administering to said patient a therapeutically effective amount of the compound of claim 3 .

21. A method of treating osteoporosis in a patient in need thereof, comprising administering to said patient a therapeutically effective amount of the compound of claim 3 .

22. A method of treating bone resorption in a patient in need thereof, comprising administering to said patient a therapeutically effective amount of the compound of claim 3 .

23. A compound, which is 4-{3-(cyanomethyl)-3-[4-(7H-pyrrolo[2,3-d]pyrimidin-4-yl)-1H-pyrazol-1-yl]azetidin-1-yl}-2,5 -difluoro-N-[(1R)-2,2,2-trifluoro-1-methylethyl]benzamide, or a pharmaceutically acceptable salt thereof.

24. A compound, which is 4-{3-(cyanomethyl)-3-[4-(7H-pyrrolo[2,3-d]pyrimidin-4-yl)-1H-pyrazol-1-yl]azetidin-1-yl}-2,5 -difluoro-N-[(1R)-2,2,2-trifluoro--methylethyl]benzamide.

25. A composition comprising a compound according to claim 23 , or a pharmaceutically acceptable salt thereof, and a pharmaceutically acceptable carrier.

26. A composition comprising a compound according to claim 24 and a pharmaceutically acceptable carrier.

27. A method of treating multiple sclerosis in a patient in need thereof, comprising administering to said patient a therapeutically effective amount of the compound of claim 23 , or a pharmaceutically acceptable salt thereof.

28. A method of treating type I diabetes in a patient in need thereof, comprising administering to said patient a therapeutically effective amount of the compound of claim 23 , or a pharmaceutically acceptable salt thereof.

29. A method of treating lupus in a patient in need thereof, comprising administering to said patient a therapeutically effective amount of the compound of claim 23 , or a pharmaceutically acceptable salt thereof.

30. A method of treating myasthenia gravis in a patient in need thereof, comprising administering to said patient a therapeutically effective amount of the compound of claim 23 , or a pharmaceutically acceptable salt thereof.

31. A method of treating myocarditis in a patient in need thereof, comprising administering to said patient a therapeutically effective amount of the compound of claim 23 , or a pharmaceutically acceptable salt thereof.

32. A method of treating contact dermatitis in a patient in need thereof, comprising administering to said patient a therapeutically effective amount of the compound of claim 23 , or a pharmaceutically acceptable salt thereof.

33. A method of treating psoriasis in a patient in need thereof, comprising administering to said patient a therapeutically effective amount of the compound of claim 23 , or a pharmaceutically acceptable salt thereof.

34. A method of treating osteoporosis in a patient in need thereof, comprising administering to said patient a therapeutically effective amount of the compound of claim 23 , or a pharmaceutically acceptable salt thereof.

35. A method of treating bone resorption in a patient in need thereof, comprising administering to said patient a therapeutically effective amount of the compound of claim 23 , or a pharmaceutically acceptable salt thereof.

36. A method of treating multiple sclerosis in a patient in need thereof, comprising administering to said patient a therapeutically effective amount of the compound of claim 24 .

37. A method of treating type I diabetes in a patient in need thereof, comprising administering to said patient a therapeutically effective amount of the compound of claim 24 .

38. A method of treating lupus in a patient in need thereof, comprising administering to said patient a therapeutically effective amount of the compound of claim 24 .

39. A method of treating myasthenia gravis in a patient in need thereof, comprising administering to said patient a therapeutically effective amount of the compound of claim 24 .

40. A method of treating myocarditis in a patient in need thereof, comprising administering to said patient a therapeutically effective amount of the compound of claim 24 .

41. A method of treating contact dermatitis in a patient in need thereof, comprising administering to said patient a therapeutically effective amount of the compound of claim 24 .

42. A method of treating psoriasis in a patient in need thereof, comprising administering to said patient a therapeutically effective amount of the compound of claim 24 .

43. A method of treating osteoporosis in a patient in need thereof, comprising administering to said patient a therapeutically effective amount of the compound of claim 24 .

44. A method of treating bone resorption in a patient in need thereof, comprising administering to said patient a therapeutically effective amount of the compound of claim 24 .
