Pharmaceutical compositions with enhanced performance
View Patent ↗Disclosed are polymers of hydroxypropyl methyl cellulose acetate succinate (HPMCAS) and hydroxypropyl methyl cellulose acetate (HPMCA) with unique degrees of substitution of hydroxypropoxy, methoxy, acetyl, and succinoyl groups. When used in making compositions comprising a low-solubility drug and such polymers, the polymers provide enhanced aqueous concentrations and/or improved physical stability.
1. A composition comprising a solid amorphous dispersion of a low-solubility drug having a solubility parameter δ D and polymeric hydroxypropyl methyl cellulose acetate succinate (HPMCAS) having a solubility parameter δ P , wherein the degree of substitution of acetyl groups (DOS Ac ) and the degree of substitution of succinoyl groups (DOSs) of said HPMCAS are as follows:
DOS S ≧0.02,
DOS Ac ≧0.7, and
DOS Ac +DOS S ≧0.9; and
wherein (δ D −δ P ) 2 <2.
2. The composition of claim 1 wherein DOS Ac ≧0.72.
3. The composition of claim 1 wherein the degree of substitution of HPMCAS is as follows:
DOS S ≧0.05,
DOS Ac ≧0.72, and
DOS Ac +DOS S ≧0.9.
4. The composition of claim 1 wherein the degree of substitution of methyl groups (DOS M ) of HPMCAS is equal to or greater than 1.6 and less than or equal to 2.15.
5. The composition of claim 4 wherein DOS M of HPMCAS is equal to or greater than 1.75 and less than or equal to 2.0.
6. The composition of claim 4 wherein DOS Ac +DOS S +DOS M ≧2.7.
7. The composition of claim 6 wherein DOS Ac +DOS S +DOS M ≧2.8.
8. The composition of claim 6 wherein DOS Ac +DOS S +DOS M ≧2.85.
9. The composition of claim 1 wherein (δ D −δ P ) 2 <1.8.
10. The composition of claim 1 wherein (δ D −δ P ) 2 <1.5.
