Method and compositions involving microRNA
The present invention concerns methods and compositions for isolating, enriching, and/or labeling miRNA molecules and for preparing and using arrays or other detection techniques for miRNA analysis. Moreover, the present invention concerns methods and compositions for generating miRNA profiles and employing such profiles for therapeutic, diagnostic, and prognostic applications.
1. A method for evaluating a thyroid sample in a patient comprising:
a) measuring the expression level of miR-21 in the sample;
b) comparing the expression level to the expression level of the same miRNAs in a noncancerous thyroid sample; and,
c) identifying the patient as having a marker for thyroid cancer if the sample is determined to have increased expression of miR-21.
2. The method of claim 1 , wherein the sample is determined to have increased expression of miR-21.
3. The method of claim 1 , wherein the sample is determined not to have increased expression of miR-21 and further comprising identifying the patient as not having increased expression of miR-21.
4. The method of claim 1 , further comprising obtaining the sample from the patient.
5. The method of claim 1 , wherein the noncancerous sample is a benign thyroid sample.
6. The method of claim 1 , further comprising measuring the expression level of miR-222.
7. The method of claim 6 , wherein the sample is determined to have increased expression of miR-222.
8. The method of claim 1 , further comprising measuring the expression level of one or more of the following miRNAs: miR-10a, miR-15A, miR-22, miR-24, miR-29b, miR-30a, miR-30A-as, miR-31, miR-125, miR-126, miR-135A, miR-138, miR-145, miR-152, miR-183, miR-197, miR-199a, miR-199A-as, miR-200b, miR-203, miR-221, miR-223, miR-224, or miR-331.
9. The method of claim 7 , further comprising measuring the expression level of one or more of the following miRNAs: miR-10a, miR-15A, miR-22, miR-24, miR-29b, miR-30a, miR-30A-as, miR-31, miR-125, miR-126, miR-135A, miR-138, miR-145, miR-152, miR-183, miR-197, miR-199a, miR-199A-as, miR-200b, miR-203, miR-221, miR-223, miR-224, or miR-331.
10. A method for evaluating a thyroid sample in a patient comprising:
a) measuring an increased expression level of miR-21 in the sample compared to the expression level of miR-21 in a noncancerous thyroid sample; and,
b) identifying the patient as having a marker for thyroid cancer.
11. The method of claim 10 , further comprising measuring the expression level of miR-222 in the sample.
12. The method of claim 11 , wherein the sample is determined to have increased expression of miR-222.
13. The method of claim 10 , wherein the noncancerous thyroid sample comprises benign cells.
14. The method of claim 10 , further comprising obtaining the sample from the thyroid of the patient.
15. The method of claim 10 , further comprising measuring the expression level of one or more of the following miRNAs: miR-10a, miR-15A, miR-22, miR-24, miR-29b, miR-30a, miR-30A-as, miR-31, miR-125, miR-126, miR-135A, miR-138, miR-145, miR-152, miR-183, miR-197, miR-199a, miR-199A-as, miR-200b, miR-203, miR-221, miR-223, miR-224, or miR-331.
16. The method of claim 11 , further comprising measuring the expression level of one or more of the following miRNAs: miR-10a, miR-15A, miR-22, miR-24, miR-29b, miR-30a, miR-30A-as, miR-31, miR-125, miR-126, miR-135A, miR-138, miR-145, miR-152, miR-183, miR-197, miR-199a, miR-199A-as, miR-200b, miR-203, miR-221, miR-223, miR-224, or miR-331.
