Polynucleotides encoding antibodies against sclerostin and uses therefor
View Patent ↗The present invention relates to antibodies against sclerostin and compositions and methods of use for said antibodies to treat a pathological disorder that is mediated by sclerostin or disease related to bone abnormalities such as osteoporosis.
1. An isolated polynucleotide comprising:
a) a nucleic acid sequence having at least 80% identity with the nucleic acid sequence set forth as SEQ ID NO:92 and which encodes a V H polypeptide comprising the amino acid sequence as set forth as SEQ ID NO:70, or the complement thereof;
b) a nucleic acid sequence having at least 80% identity with the nucleic acid sequence set forth as SEQ ID NO:103 and which encodes a V L polypeptide comprising the amino acid sequence as set forth as SEQ ID NO:81, or the complement thereof;
c) both a) and b);
d) a nucleic acid sequence having at least 80% identity with the nucleic acid sequence set forth as SEQ ID NO:136 and which encodes a full length heavy chain polypeptide comprising the amino acid sequence as set forth as SEQ ID NO:114, or the complement thereof;
e) a nucleic acid sequence having at least 80% identity with the nucleic acid sequence set forth as SEQ ID NO:147 and which encodes a full length light chain polypeptide comprising the amino acid sequence as set forth as SEQ ID NO:125, or the complement thereof; or
f) both d) and e).
2. The isolated polynucleotide according to claim 1 , wherein the identity is at least 90%.
3. The isolated polynucleotide according to claim 2 , wherein the identity is at least 95%.
4. A cloning or expression vector comprising the isolated polynucleotide of claim 1 .
5. A host cell comprising the vector according to claim 4 .
6. A process for the production of a protein, comprising culturing a host cell comprising a vector comprising a polynucleotide, wherein the polynucleotide comprises:
a) a nucleic acid sequence having at least 80% identity with the nucleic acid sequence set forth as SEQ ID NO:92 and which encodes a V H polypeptide comprising the amino acid sequence as set forth as SEQ ID NO:70;
b) a nucleic acid sequence having at least 80% identity with the nucleic acid sequence set forth as SEQ ID NO:103 and which encodes a V L polypeptide comprising the amino acid sequence as set forth as SEQ ID NO:81;
c) both a) and b);
d) a nucleic acid sequence having at least 80% identity with the nucleic acid sequence set forth as SEQ ID N0:136 and which encodes a full length heavy chain polypeptide comprising the amino acid sequence as set forth as SEQ ID NO:114;
e) a nucleic acid sequence having at least 80% identity with the nucleic acid sequence set forth as SEQ ID NO:147 and which encodes a full length light chain polypeptide comprising the amino acid sequence as set forth as SEQ ID NO:125; or
f) both d) and e);
under conditions wherein said polynucleotide is translated to said protein.
7. An isolated polynucleotide comprising:
a) the nucleic acid sequence set forth as SEQ ID NO:92 or the complement thereof;
b) the nucleic acid sequence set forth as SEQ ID NO:103 or the complement thereof;
c) both a) and b):
d) the nucleic acid sequence set forth as SEQ ID NO:136 or the complement thereof;
e) the nucleic acid sequence set forth as SEQ ID NO:147 or the complement thereof; or
f) both d) and e).
8. A cloning or expression vector comprising the isolated polynucleotide of claim 7 .
9. A host cell comprising the vector according to claim 8 .
10. A process for the production of a protein, comprising culturing a host cell comprising a vector comprising a polynucleotide, wherein the polynucleotide comprises:
a) the nucleic acid sequence set forth as SEQ ID NO:92;
b) the nucleic acid sequence set forth as SEQ ID NO:103;
c) both a) and b);
d) the nucleic acid sequence set forth as SEQ ID NO:136;
e) the nucleic acid sequence set forth as SEQ ID NO:147; or
f) both d) and e);
under conditions wherein said polynucleotide is translated to said protein.
11. An isolated polynucleotide consisting of:
a) the nucleic acid sequence set forth as SEQ ID NO:92 or the complement thereof;
b) the nucleic acid sequence set forth as SEQ ID NO:103 or the complement thereof;
c) both a) and b);
d) the nucleic acid sequence set forth as SEQ ID NO:138 or the complement thereof;
e) the nucleic acid sequence set forth as SEQ ID NO:147 or the complement thereof; or
f) both d) and e).
12. A cloning or expression vector comprising the isolated polynucleotide of claim 11 .
13. A host cell comprising the vector according to claim 12 .
14. A process for the production of a protein, comprising culturing a host cell comprising a vector comprising a polynucleotide, wherein the polynucleotide consists of:
a) the nucleic acid sequence set forth as SEQ ID NO:92;
b) the nucleic acid sequence set forth as SEQ ID NO:103;
c) bath a) and b);
d) the nucleic acid sequence set forth as SEQ ID NO:136;
e) the nucleic acid sequence set forth as SEQ ID NO:147; or
f) both d and e),
under conditions wherein said polynucleotide is translated to said protein.
15. An isolated polynucleotide comprising a nucleic acid sequence that encodes:
a) a V H polypeptide comprising the amino acid sequence as set forth as SEQ ID NO:70;
b) a V L polypeptide comprising the amino acid sequence as set forth as SEQ ID NO:81; or
c) both a) and b).
16. A cloning or expression vector comprising the isolated polynucleotide of claim 15 .
17. A host cell comprising the vector according to claim 16 .
18. A process for the production of an antibody or antigen binding portion thereof, comprising culturing the host cell of claim 17 under conditions wherein said polynucleotide is translated to said antibody or antigen binding portion thereof.
19. An isolated polynucleotide comprising a nucleic acid sequence that encodes:
a) a V H polypeptide comprising, in sequence,
(i) an amino acid sequence consisting of SEQ ID NO:4;
(ii) an amino acid sequence consisting of SEQ ID NO:15; and
an amino acid sequence consisting of SEQ ID NO:26;
b) a V L polypeptide comprising, in sequence,
(i) an amino acid sequence consisting of SEQ ID NO:37;
(ii) an amino acid sequence consisting of SEQ ID NO:48; and
(iii) an amino acid sequence consisting of SEQ ID NO:59; or
c) both a) and b).
