Optimized strains of
View Patent ↗Engineered strains of the oleaginous yeast Yarrowia lipolytica capable of producing greater than 50 weight percent of eicosapentaenoic acid in the total oil fraction are described. These strains over-express heterologous Δ9 elongases, Δ8 desaturases, Δ5 desaturases, Δ17 desaturases, Δ12 desaturases and C 16/18 elongases, and optionally over-express diacylglycerol cholinephosphotransferases. Gene knockouts are also described, as are methods for producing EPA within host cells, and products comprising EPA from the optimized Yarrowia lipolytica strains.
1. A microbial oil comprising:
a) at least 50.0 weight percent eicosapentaenoic acid as a weight percent of the total fatty acids; and
b) less than 6.2 weight percent of eicosadienoic acid as a weight percent of the total fatty acids, provided that the percentage of eicosadienoic acid is not equal to zero.
2. The oil of claim 1 , wherein the oil has less than 1 weight percent of gamma-linolenic acid as a weight percent of the total fatty acids.
3. The oil of claim 1 , wherein the fungal oil further comprises less than 20.9 weight percent of linoleic acid as a weight percent of the total fatty acids, provided that the percentage of linoleic acid is not equal to zero.
4. The oil of claim 3 , wherein the oil further comprises less than 9.4 weight percent of oleic acid as a weight percent of the total fatty acids.
5. The oil of claim 4 , wherein the oil comprises less than 6.9 weight percent of oleic acid as a weight percent of the total fatty acids.
6. An oil concentrate derived from the oil of claim 4 .
7. The oil concentrate of claim 6 , wherein the oil comprises at least 60 weight percent of eicosapentaenoic acid as a weight percent of the total fatty acids.
8. The oil concentrate of claim 6 , wherein the oil comprises at least 70 weight percent of eicosapentaenoic acid as a weight percent of the total fatty acids.
9. The oil of claim 1 , wherein the oil comprises less than 4.2 weight percent of eicosadienoic acid as a weight percent of the total fatty acids, provided that the percentage of eicosadienoic acid is not equal to zero.
10. The oil of claim 1 , wherein the oil comprises less than 3.1 weight percent of eicosadienoic acid as a weight percent of the total fatty acids, provided that the percentage of eicosadienoic acid is not equal to zero.
11. The oil of claim 3 , wherein the oil comprises less than 4.2 weight percent of eicosadienoic acid as a weight percent of the total fatty acids, provided that the percentage of eicosadienoic acid is not equal to zero.
12. The oil of claim 4 , wherein the oil comprises less than 4.2 weight percent of eicosadienoic acid as a weight percent of the total fatty acids, provided that the percentage of eicosadienoic acid is not equal to zero.
13. The oil of claim 1 , wherein the oil has less than 0.1 weight percent of docosahexaenoic acid as a weight percent of the total fatty acids.
14. The oil of claim 3 , wherein the oil has less than 0.1 weight percent of docosahexaenoic acid as a weight percent of the total fatty acids.
15. The oil of claim 4 , wherein the oil has less than 0.1 weight percent of docosahexaenoic acid as a weight percent of the total fatty acids.
16. The oil of claim 9 , wherein the oil has less than 0.1 weight percent of docosahexaenoic acid as a weight percent of the total fatty acids.
17. The oil of claim 10 , wherein the oil has less than 0.1 weight percent of docosahexaenoic acid as a weight percent of the total fatty acids.
18. The oil of claim 11 , wherein the oil has less than 0.1 weight percent of docosahexaenoic acid as a weight percent of the total fatty acids.
19. The oil of claim 12 , wherein the oil has less than 0.1 weight percent of docosahexaenoic acid as a weight percent of the total fatty acids.
