Abstract

The invention provides methods of modifying the lipids produced by microbial organisms through genetic engineering. The invention also provides genetically engineered microbes and methods of fermenting microbes for oil production. Also provided are oils, fuels, oleochemicals, chemical precursors, and other compounds manufactured by such modified microorganisms. Exemplary oil-bearing organisms include organisms containing one or more exogenous genes encoding a fatty acyl-ACP thioesterase, fatty acyl-CoA/aldehyde reductase, fatty acyl-CoA reductase, fatty aldehyde reductase, fatty aldehyde decarbonylase, and/or an acyl carrier protein.

Claims (17)

1. A residual biomass comprising cell material remaining after extracting lipid from heterotrophically cultivated, recombinant oleaginous microalgal cells comprising a nucleic acid operable to express an exogenous invertase.

2. A residual biomass according to claim 1 , wherein the extracting is by lysing the cells and separating a lipid-containing hydrophobic fraction from aqueous or solid cell materials.

3. A residual biomass according to claim 2 , wherein the cells are dried prior to the lysis.

4. A residual biomass according to claim 2 , wherein the cells are lysed using an acid, a base, a cellulase, a protease, ultrasound, osmotic shock, a virus, expression of a lytic gene or mechanical lysis.

5. A residual biomass according to claim 2 , wherein the separating comprises solvent extraction.

6. A residual biomass according to claim 2 , wherein the separating comprises centrifugation.

7. A residual biomass according to claim 1 , wherein the residual biomass comprises protein.

8. A residual biomass according to claim 1 , wherein the residual biomass comprises cell-wall material.

9. A residual biomass according to claim 1 , wherein the cell is from a strain that has been mutated to increase the production of lipids.

10. A residual biomass according to claim 1 , wherein the cells are cultivated in a bioreactor holding at least 40,000 liters.

11. A residual biomass according to claim 1 , wherein the cells are cultivated for a time under conditions of nitrogen limitation.

12. A residual biomass produced by the process of:

(a) heterotrophically cultivating oleaginous microalgal cells comprising a nucleic acid operable to express an exogenous invertase in a bioreactor holding at least 40,000 liters;

(b) lysing the cells to produce a lysed cell material; and

(c) extracting lipid from the lysed cell material, thus producing the residual biomass material.

13. A residual biomass of claim 12 , wherein the cells are cultivated for a time under conditions of limiting nitrogen.

14. A residual biomass of claim 12 , wherein the lysis comprises drying the cells prior to disruption.
