Abstract

The present invention provides novel and improved protein purification processes which incorporate certain types of carbonaceous materials and result in effective and selective removal of certain undesirable impurities without adversely effecting the yield of the desired protein product.

Claims (20)

1. A flow-through process for purifying a target molecule from a Protein A eluate comprising the steps of: (i) contacting the eluate recovered from a Protein A chromatography column with activated carbon, thereby to remove host cell proteins (HCPs); (ii) contacting the flow-through sample from step (i) with an anion exchange chromatography media; (iii) contacting the flow-through sample from step (ii) with a cation exchange chromatography media; and (iv) obtaining the flow-through sample from step (iii) comprising the target molecule, wherein the eluate flows continuously through steps (i)-(iii) and wherein level of HCPs in the flow-through sample after step (i) is less than 30% of the level in the eluate prior to step (i).

2. The flow-through process of claim 1 , further comprising subjecting the flow-through sample from step (iii) to virus filtration.

3. The flow-through process of claim 1 , further comprising use of an in-line static mixer and/or a surge tank between steps (ii) and (iii) to change pH.

4. The flow-through process of claim 1 , wherein the process employs a single skid.

5. The flow-through process of claim 1 , wherein the eluate from the Protein A chromatography column is subjected to virus inactivation prior to contacting with activated carbon.

6. The process of claim 1 , wherein the target molecule is an antibody or a functional fragment thereof.

7. The process of claim 6 , wherein the antibody is a monoclonal antibody.

8. The process of claim 6 , wherein the antibody is a polyclonal antibody.

9. The process of claim 1 , wherein activated carbon comprises activated charcoal.

10. The process of claim 1 , wherein the activated carbon is packed in a column, a sealed disposable device, a cartridge or a capsule.

11. The process of claim 1 , wherein the activated carbon is impregnated into a porous material.

12. The process of claim 11 , wherein the porous material is contained within a column, a sealed disposable device, a cartridge or a capsule.

13. The process of claim 1 , wherein the yield of target protein after step (iv) is greater than 85%.

14. A flow-through purification process for purifying a target molecule from a Protein A eluate comprising contacting the eluate with activated carbon and one or more matrices selected from anion exchange media, cation exchange media and virus filtration media, wherein the flow of the eluate is continuous.

A method of reducing the level of HCPs in a sample containing a protein of interest and HCPs, the method comprising the steps of: (i) contacting a sample comprising a protein of interest and HCPs with a chromatography column containing affinity media; (ii) obtaining a first eluate of the sample; (iii) contacting the first eluate from (ii) in flow-through mode with a carbonaceous material: (iv) obtaining a second eluate of the sample; (v) contacting the second eluate with an anion exchange porous media membrane adsorber; and (vi) obtaining a third eluate of the sample, wherein the third eluate comprises a lower level of HCPs relative to the level of HCPs when the carbonaceous material is not used in the method.

A method of reducing the level of HCPs in a sample containing a protein of interest, the method comprising the steps of: (i) providing a sample comprising a protein of interest and HCPs; (ii) contacting the sample with a suitable media to capture the protein of interest; (iii) obtaining a first eluate of the sample; (iv) contacting the first eluate with a carbonaceous material; (v) obtaining a second eluate of the sample; and (vi) contacting the second eluate with an anion exchange media; and (vii) obtaining a third eluate of the sample, wherein the third eluate comprises a lower level of HCPs relative to the level of one or more impurities when step (iv) is not performed.

17. The method of claim 16 , wherein the protein of interest is an antibody.

18. The method of claim 16 , wherein the antibody is a monoclonal antibody.

19. The method of claim 16 , wherein the anion exchange media is resin or a membrane.

20. The method of claim 16 , wherein the carbonaceous material is activated carbon.
