Abstract

Methods for identifying a compound capable of interacting with a Receptor Tyrosine Kinase (RTK) including providing a device capable of measuring cell-substrate impedance, adding test cells expressing a RTK to the device, measuring first impedances, adding a compound to at least one compound well and adding a vehicle control to at least one control well, measuring second impedances of the compound well and the control well, determining the change in the impedance for the compound well and the one control well, comparing the change in impedance between the compound well and the control well, and identifying the compound interacts with the RTK if the comparison demonstrates a significant difference between the change in impedance.

Claims (36)

1. A method of identifying a factor capable of interacting with a receptor tyrosine kinase (RTK) comprising:

a) providing a device capable of measuring cell-substrate impedance, wherein said device comprises at least two wells, further wherein said device is operably connected to an impedance analyzer;

b) adding test cells to at least two wells, wherein said test cells are cells that express a RTK;

c) measuring first impedances of said at least two wells immediately preceding step d) and optionally determining first cell indices from said first impedances;

d) adding a compound suspected of being a factor capable of interacting with a RTK to at least one well containing said test cells to form at least one compound well and adding a vehicle control to at least another well containing test cells to form at least one control well;

e) measuring second impedances of said at least one compound well and of said at least one control well after step d) and optionally determining second cell indices from said second impedance;

f) determining a change in the impedance or cell index for said at least one compound well by comparing said second impedance or said second cell index of said at least one compound well to said first impedance or said first cell index of said at least one compound well, and determining a change in the impedance or cell index for said at least one control well by comparing said second impedance or said second cell index of said at least one control well to said first impedance or said first cell index of the said at least one control well;

g) comparing said changes in impedances or in cell indices between said at least one compound well and said at least one control well; and

h) identifying said compound is a factor that interacts with said RTK and affects cell morphology if said comparison demonstrates a more than 10% difference between said change in impedance or cell index for said at least one compound well and said change in impedance or cell index for said at least one control well.

The method according to claim 1 , wherein said cell index is a normalized cell index wherein normalization is at a time point a short time before adding said compound or said vehicle control, wherein the said short time is selected from the group consisting of less than 1 minute, less than 2 minutes, less than 5 minutes, less than 10 minutes, less than 30 minutes, less than 1 hour, less than 2 hours, less than 5hours, less than 10 hours and less than 24 hours.

3. The method according to claim 1 , wherein said test cells are in a serum free medium.

4. A method of identifying a factor capable of interacting with a receptor tyrosine kinase (RTK) comprising:

(a) providing a device capable of measuring cell-substrate impedance, wherein said device comprises at least two wells, further wherein said device is operably connected to an impedance analyzer;

(b) adding test cells to at least one well to form at least one test well, wherein said test cells are cells that express a RTK, and adding control cells to at least another well to form at least one control well, wherein said control cells do not express said RTK or express said RTK at a significantly lesser level that said test cells;

(c) measuring first impedances from said at least one test well and from said at least one control well immediately preceding step d) and optionally determining first cell indices from said first impedances;

(d) adding a compound to said at least one test well and to said at least one control well;

(e) measuring second impedances from said at least one test well and from said at least one control well after step d) and optionally determining second cell indices from said second impedances;

(f) determining the change in the impedance or cell index for said at least one test well by comparing said second impedance or said second cell index of said at least one test well to said first impedance or said first cell index of said at least one test well, and determining the change in the impedance or cell index for said at least one control well by comparing said second impedance or said second cell index of said at least one control well to said first impedance or said first cell index of the said at least one control well;

(g) comparing said changes in impedance or in cell index between said at least one test well and said at least one control well; and

(h) identifying said compound interacts with said RTK if said comparison demonstrates a more than 10% difference between said change in impedance or cell index for said at least one test well and said change in impedance or cell index for said at least one control well.

5. The method according to claim 1 , wherein said compound is a peptide or a protein.

6. The method according to claim 1 , wherein said compound is selected from the group consisting of a ligand, an agonist, an inverse agonist, an antagonist, an inhibitor, and a RTK activating factor.

7. The method according to claim 1 , wherein two compounds are added to said compound well at a same time or at different time points prior to measuring the second impedance of said compound well.

8. The method according to claim 7 , wherein said two compounds are added at different time points.

9. The method according to claim 8 , wherein said first compound is an antagonist and the second compound is an agonist.

10. The method according to claim 9 , wherein said antagonist is added to compound wells at different concentrations and said agonist is added to said compound wells at a same concentration.

11. The method according to claim 10 , further comprising determining a dose dependent response curve for said antagonist and determining for each well, a maximum change in area under the curve.

12. The method according to claim 11 , further comprising determining an IC50 of said antagonist.

13. The method according to claim 9 , further comprising determining a dose dependent response curve for said agonist and determining for each well, a maximum change in area under the curve.

14. The method according to claim 13 , further comprising determining an EC50 of said agonist.

15. The method according to claim 4 , wherein said compound is a peptide or a protein.

16. The method according to claim 4 , wherein said compound is selected from the group consisting of a ligand, an agonist, an inverse agonist, an antagonist, an inhibitor and a RTK activating factor.

17. The method according to claim 1 , wherein said compound is an inorganic molecule.

18. The method according to claim 4 , wherein said compound is an inorganic molecule.

19. The method according to claim 1 , wherein said first cell indices are determined, said second cell indices are determined, and said changes in cell indices are compared to identify whether said compound is a factor that interacts with said RTK and affects cell morphology.

20. The method according to claim 4 , wherein said first cell indices are determined, said second cell indices are determined, and said changes in cell indices are compared to identify whether said compound is a factor that interacts with said RTK.
