Abstract

The invention provides antibodies that specifically bind to OX40 (CD134), referred to as OX40 antibodies, anti-OX40 or anti-OX40 antibodies. Invention antibodies that specifically bind to OX40 include mammalian (human, primate, etc.), humanized and chimeric anti-OX40 antibodies. Invention antibodies and antibody subsequences (fragments) that specifically bind to OX40 include purified and isolated antibodies, as well as pharmaceutical formulations thereof, are useful in various methods including treatment, screening and detection methods.

Claims (27)

1. An isolated or purified monoclonal antibody, or a fragment thereof, that specifically binds to an epitope in an amino acid sequence OX40 extracellular domain, wherein

the antibody comprises amino acid sequences of all CDRs of a heavy chain variable region and a light chain variable region of an antibody which is produced by a hybridoma cell line denoted as 112V8, deposited as ATCC Accession No. PTA-7219.

2. The monoclonal antibody, or the fragment thereof, of claim 1 , wherein the epitope in an amino acid sequence of OX40 extracellular domain is within an amino acid sequence set forth as:

3. The isolated or purified monoclonal antibody, or the fragment thereof, of claim 1 , wherein the antibody comprises the amino acid sequence of VH and VL of the antibody which is produced by a hybridoma cell line denoted as 112V8, deposited as ATCC Accession No. PTA-7219.

The monoclonal antibody, or the fragment thereof, of claim 1 , wherein the antibody comprises: a heavy chain variable region comprising an amino acid sequence from the amino acid at position 20 of SEQ ID NO:9 to the amino acid at position 141 of SEQ ID NO:9, and a light chain variable region comprising an amino acid sequence from the amino acid at position 21 of SEQ ID NO:10 to the amino acid at position 129 of SEQ ID NO:10.

5. The fragment of the monoclonal antibody of any one of claim 1 - 3 and 4 , wherein the fragment is selected from Fab, Fab′, F(ab′) 2 , Fv, Fd, single-chain Fvs (scFv), or disulfide-linked Fvs (sdFv).

6. The monoclonal antibody of any one of claim 1 - 3 and 4 , wherein the OX40 is human OX40.

7. The monoclonal antibody of any one of claim 1 - 3 and 4 , wherein the OX40 comprises a sequence:

8. The monoclonal antibody, or the fragment thereof, of any one of claim 1 - 3 and 4 , wherein the antibody is an IgG1, IgG2, IgG3, IgG4, IgA, IgM, IgE, or IgD isotype.

9. The monoclonal antibody or, the fragment thereof, of any one of claim 1 - 3 and 4 , wherein the antibody or subsequence further comprises a heterologous domain.

10. A pharmaceutical composition comprising the monoclonal antibody or the fragment thereof of any one of claim 1 - 3 and 4 , and a pharmaceutically acceptable carrier or excipient.

11. The monoclonal antibody, or the fragment thereof, any one of claims 1 and 2 , wherein the antibody comprises a human, humanized or chimeric antibody.

12. The monoclonal antibody, or the fragment thereof, any one of claims 1 - 3 and 4 , wherein the antibody has OX40 antagonist activity.
