Abstract

An osmotic delivery system flow modulator includes an outer shell constructed and arranged for positioning in an opening, an inner core inserted in the outer shell, and a fluid channel having a spiral shape defined between the outer shell and the inner core. The fluid channel is adapted for delivery of an active agent formulation from the reservoir of the osmotic delivery system.

Claims (55)

1. An implantable delivery system for an active agent formulation, comprising:

a reservoir made of an impermeable, metallic material;

a partition within the reservoir, the partition defining within the reservoir a first chamber and a second chamber;

the first chamber in the reservoir containing an osmotic engine;

the second chamber in the reservoir containing the active agent formulation, the active agent formulation comprising an active agent;

a semipermeable plug disposed at a first end of the reservoir adjacent the first chamber; and

a flow modulator comprising (i) two pieces, an outer shell and an inner core, wherein the outer shell and the inner core are made of a non-metallic, non-reactive polymer material, and (ii) an internal fluid channel, wherein the flow modulator is disposed at a second end of the reservoir adjacent the second chamber, the flow modulator comprising

the outer shell positioned in an opening at the second end of the reservoir, the outer shell comprising an inner surface,

the inner core inserted in the outer shell, the inner core comprising an outer surface, and

the internal fluid channel having a spiral shape defined between the outer shell and the inner core, wherein the fluid channel is (i) formed on the outer surface of the inner core that mates with the inner surface of the outer shell, or (ii) formed on the inner surface of the outer shell that mates with the outer surface of the inner core.

2. The implantable delivery system of claim 1 , wherein the inner core further comprises an outer shoulder and the outer shell further comprises an inner shoulder, wherein the outer shoulder and the inner shoulder engage to prevent expulsion of the inner core from the outer shell.

3. The implantable delivery system of claim 1 , wherein a length of the fluid channel ranges from 10 mm to 50 mm.

4. The implantable delivery system of claim 1 , wherein an effective cross-sectional diameter of the fluid channel ranges from 0.1 mm to 0.5 mm.

5. The implantable delivery system of claim 1 , wherein the active agent comprises a protein or a peptide.

6. The implantable delivery system of claim 1 , wherein the flow modulator is press-fitted into the opening at the second end of the reservoir.

The implantable delivery system of claim 1 , wherein the non-metallic, non-reactive polymer material of the outer shell and the inner core comprises a polyaryletherketone, an ultra-high molecular weight polyethylene, a fluorinated ethylene-propylene, a polymethylpentene, or a liquid crystal polymer.

8. The implantable delivery system of claim 7 , wherein the polyaryletherketone is polyetheretherketone or polyaryletheretherketone.

9. The implantable delivery system of claim 1 , wherein the metallic material of the reservoir comprises gold-plated ferrous alloys, platinum-plated ferrous alloys, cobalt-chromium alloys, or titanium nitride coated stainless steel.

10. The implantable delivery system of claim 1 , wherein the metallic material of the reservoir comprises stainless steel, titanium, platinum, tantalum, gold, or alloys thereof.

11. The implantable delivery system of claim 1 , wherein the metallic material of the reservoir comprises titanium or a titanium alloy.

12. The implantable delivery system of claim 1 , wherein the partition comprises a slidable piston.

13. The implantable delivery system of claim 1 , wherein the semipermeable plug comprises a material selected from the group consisting of plasticized cellulosic materials, enhanced polymethyl methacrylates, elastomeric materials, polyether-polyamide copolymers, and thermoplastic copolyesters.

14. The implantable delivery system of claim 1 , wherein the semipermeable plug comprises a polyurethane.

15. An implantable delivery system for an active agent formulation, comprising:

a reservoir made of an impermeable, metallic material;

a partition within the reservoir, the partition defining within the reservoir a first chamber and a second chamber;

the first chamber in the reservoir containing an osmotic engine;

the second chamber in the reservoir containing the active agent formulation, the active agent formulation comprising an active agent;

a semipermeable plug disposed at a first end of the reservoir adjacent the first chamber; and

a flow modulator comprising (i) two pieces, an outer shell and an inner core, wherein the outer shell and the inner core are made of a non-metallic polymer material, and (ii) an internal fluid channel having an effective cross-sectional diameter of from 0.1 mm to 0.5 mm, wherein the flow modulator is disposed at a second end of the reservoir adjacent the second chamber, the flow modulator comprising

the outer shell positioned in an opening at the second end of the reservoir, the outer shell comprising an inner surface,

the inner core inserted in the outer shell, the inner core comprising an outer surface, and

the internal fluid channel having a spiral shape defined between the outer shell and the inner core, wherein the fluid channel is (i) formed on the outer surface of the inner core that mates with the inner surface of the outer shell, or (ii) formed on the inner surface of the outer shell that mates with the outer surface of the inner core.

16. The implantable delivery system of claim 15 , wherein the inner core further comprises an outer shoulder and the outer shell further comprises an inner shoulder, wherein the outer shoulder and the inner shoulder engage to prevent expulsion of the inner core from the outer shell.

17. The implantable delivery system of claim 15 , wherein a length of the fluid channel ranges from 10 mm to 50 mm.

18. The implantable delivery system of claim 15 , wherein the active agent comprises a protein or a peptide.

19. The implantable delivery system of claim 15 , wherein the flow modulator is press-fitted into the opening at the second end of the reservoir.

20. The implantable delivery system of claim 15 , wherein the non-metallic polymer material of the outer shell and the inner core comprises a polyaryletherketone, an ultra-high molecular weight polyethylene, a fluorinated ethylene-propylene, a polymethylpentene, or a liquid crystal polymer.

21. The implantable delivery system of claim 20 , wherein the polyaryletherketone is polyetheretherketone or polyaryletheretherketone.

22. The implantable delivery system of claim 15 , wherein the metallic material of the reservoir comprises gold-plated ferrous alloys, platinum-plated ferrous alloys, cobalt-chromium alloys, or titanium nitride coated stainless steel.

23. The implantable delivery system of claim 15 , wherein the metallic material of the reservoir comprises stainless steel, titanium, platinum, tantalum, gold, or alloys thereof.

24. The implantable delivery system of claim 15 , wherein the metallic material of the reservoir comprises titanium or a titanium alloy.

25. The implantable delivery system of claim 15 , wherein the partition comprises a slidable piston.

26. The implantable delivery system of claim 15 , wherein the semipermeable plug comprises a material selected from the group consisting of plasticized cellulosic materials, enhanced polymethyl methacrylates, elastomeric materials, polyether-polyamide copolymers, and thermoplastic copolyesters.

27. The implantable delivery system of claim 15 , wherein the semipermeable plug comprises a polyurethane.

28. An implantable delivery system for an active agent formulation, comprising:

a reservoir made of titanium or a titanium alloy;

a slidable piston within the reservoir, the piston defining within the reservoir a first chamber and a second chamber;

the first chamber in the reservoir containing an osmotic engine;

the second chamber in the reservoir containing the active agent formulation, wherein the active agent formulation comprises a protein or a peptide;

a semipermeable plug disposed at a first end of the reservoir adjacent the first chamber, wherein the semipermeable plug comprises a polyurethane; and

a flow modulator comprising (i) two pieces, an outer shell and an inner core, wherein the outer shell and the inner core are made of a polyetheretherketone or a polyaryletheretherketone, and (ii) an internal fluid channel having an effective cross-sectional diameter of from 0.1 mm to 0.5 mm, wherein the flow modulator is disposed at a second end of the reservoir adjacent the second chamber, the flow modulator comprising

the outer shell positioned in an opening at the second end of the reservoir, the outer shell comprising an inner surface,

the inner core inserted in the outer shell, the inner core comprising an outer surface, and

the internal fluid channel having a spiral shape defined between the outer shell and the inner core, wherein the fluid channel is (i) formed on the outer surface of the inner core that mates with the inner surface of the outer shell, or (ii) formed on the inner surface of the outer shell that mates with the outer surface of the inner core.
