Abstract

Disclosed herein are multi-component formulations for enzymatically producing aqueous solutions of peroxycarboxylic acids suitable for use in, e.g., disinfectant and/or bleaching applications. The multi-component peroxycarboxylic acid formulations comprise at least one carbohydrate esterase family 7 enzyme having perhydrolytic activity.

Claims (78)

1. A method of bleaching, destaining, deodorizing, sanitizing or disinfecting a surface, an article of clothing or a textile comprising:

(a) providing a formulation comprising:

(A) a first component comprising:

(i) an enzyme powder comprising:

(1) at least one enzyme catalyst having perhydrolysis activity, wherein said enzyme catalyst comprises an enzyme having a carbohydrate esterase family 7 (CE-7) signature motif that aligns with SEQ ID NO: 1 using CLUSTALW, said signature motif comprising:

 (I) an RGQ motif at amino acid positions aligning with 118-120 of SEQ ID NO:1;

 (II) a GXSQG motif at amino acid positions aligning with 179-183 of SEQ ID NO:1; and

 (III) an HE motif at amino acid positions aligning with 298-299 of SEQ ID NO:1;

 said enzyme comprising at least 30% amino acid identity to SEQ ID NO: 1; and

(2) at least one excipient;

(ii) a carboxylic acid ester substrate selected from the group consisting of

(1) one or more esters having the structure

[X] m R 5

 wherein

 X is an ester group of the formula R 6 C(O)O;

 R 6 is a C1 to C7 linear, branched or cyclic hydrocarbyl moiety, optionally substituted with a hydroxyl group or C1 to C4 alkoxy group, wherein R 6 optionally comprises one or more ether linkages where R 6 is C2 to C7;

 R 5 is a C1 to C6 linear, branched, or cyclic hydrocarbyl moiety optionally substituted with a hydroxyl group, wherein each carbon atom in R 5 individually comprises no more than one hydroxyl group or no more than one ester group, and wherein R 5 optionally comprises one or more ether linkages;

 m is 1 to the number of carbon atoms in R 5 ,

 said one or more esters having solubility in water of at least 5 ppm at 25° C.;

(2) one or more glycerides having the structure

 wherein R 1 is a C1 to C7 straight chain or branched chain alkyl optionally substituted with an hydroxyl or a C1 to C4 alkoxy group and R 3 and R 4 are individually H or R 1 C(O);

(3) one or more esters of the formula

 wherein R 1 is a C1 to C7 straight chain or branched chain alkyl optionally substituted with an hydroxyl or a C1 to C4 alkoxy group and R 2 is a C1 to C10 straight chain or branched chain alkyl, alkenyl, alkynyl, aryl, alkylaryl, alkylheteroaryl, heteroaryl, (CH 2 CH 2 O) n , or (CH 2 CH(CH 3 )—O) n H and n is 1 to 10;

(4) one or more acetylated monosaccharides, acetylated disaccharides, or acetylated polysaccharides; and

(5) any combination of (1) through (4);

the first component is designed to provide a final concentration of 0.5 wt % to 10 wt % in a reaction formulation formed by combining the first and second components;

(iii) a buffer selected from the group consisting of bicarbonate, citrate, acetate, phosphate, pyrophosphate, methylphosphonate, succinate, malate, fumarate, tartrate, and maleate;

(iv) a cosolvent selected from the group consisting of tripropylene glycol methyl ether, dipropylene glycol methyl ether, propylene glycol methyl ether, diethylene glycol butyl ether, dipropylene glycol, triethylene glycol, 1,2-propanediol, N-ethyl-2-pyrroldinone, isopropanol, ethanol, ethyl lactate, 1,3-propanediol, and any combination thereof; and

(v) optionally at least one surfactant; and

(B) a second component comprising water, hydrogen peroxide, and a hydrogen peroxide stabilizer;

(b) combining the first component with the second component whereby a reaction formulation comprising peroxycarboxylic acid is produced; and

(c) applying the reaction formulation to the surface, the article of clothing or the textile for bleaching, stain removal, odor reduction, sanitization, disinfection, or a combination thereof.

2. The method of claim 1 wherein the at least one excipient ranges from about 95 wt % to about 25 wt % of the enzyme powder.

3. The method of claim 1 wherein

(a) the carboxylic acid ester substrate is triacetin; wherein the amount of triacetin in the first component provides a final concentration of 0.5 wt % to 10 wt % triacetin in a reaction formulation formed by combining the first and second components;

(b) the buffer is about 0.1 wt % to about 10% wt of a the first component, wherein said buffer is selected from the group consisting of sodium bicarbonate, potassium bicarbonate, a mixture of sodium bicarbonate and potassium bicarbonate, sodium phosphate, potassium phosphate, and a mixture of sodium phosphate and potassium phosphate;

(d) the cosolvent is tripropylene glycol methyl ether and is in a concentration of up to 80 wt % of the first component;

(e) the surfactant is present and is polysorbate 80; and

(f) the hydrogen peroxide in the second component is present in an amount that provides a final concentration in a reaction formulation formed by combining the first and second components of from 0.33 wt % to about 30 wt %.

4. The method of claim 1 wherein the first component is combined with said second component at a ratio of about 1:1 to about 1:10 by weight.

5. A method of bleaching, destaining, deodorizing, sanitizing or disinfecting a surface, an article of clothing or a textile comprising:

(a) providing a comprising:

(A) a first component comprising:

(i) an enzyme powder; said enzyme powder comprising:

(1) at least one CE-7 enzyme having perhydrolysis activity, wherein said at least one CE-7 enzyme comprises an amino acid sequence selected from the group consisting of SEQ ID NO: 6, SEQ ID NO: 7, SEQ ID NO: 19 and SEQ ID NO: 20 or an amino acid sequence substantially similar to SEQ ID NO: 6, SEQ ID NO: 7, SEQ ID NO: 19 or SEQ ID NO: 20; and

(2) at least one excipient;

(ii) a carboxylic acid ester substrate selected from the group consisting of monoacetin, diacetin, triacetin, and a mixtures thereof; wherein the amount of the carboxylic acid ester substrate in the first component is designed to provide a final concentration of 0.5 wt % to 10 wt % in a reaction formulation formed by combining the first and second components;

(iii) a buffer selected from the group consisting of bicarbonate, citrate, acetate, phosphate, pyrophosphate, methylphosphonate, succinate, malate, fumarate, tartrate, and maleate;

(iv) a cosolvent selected from the group consisting of tripropylene glycol methyl ether, dipropylene glycol methyl ether, propylene glycol methyl ether, diethylene glycol butyl ether, dipropylene glycol, triethylene glycol, 1,2-propanediol, N-ethyl-2-pyrroldinone, isopropanol, ethanol, ethyl lactate, 1,3-propanediol, and any combination thereof; and

(v) optionally at least one surfactant; and

(B) a second component comprising water, hydrogen peroxide, and a hydrogen peroxide stabilizer;

(b) combining the first component with the second component whereby a reaction formulation comprising peroxycarboxylic acid is produced; and

(c) applying the reaction formulation to the surface, the article of clothing or the textile for bleaching, stain removal, odor reduction, sanitization, disinfection, or a combination thereof.

6. The method of claim 5 wherein:

(a) the at least one CE-7 enzyme comprises an amino acid sequence selected from the group consisting of SEQ ID NO: 6, SEQ ID NO: 7, SEQ ID NO: 19 and SEQ ID NO: 20, wherein amino acid residue 277 of SEQ ID NO: 19 or SEQ ID NO: 20 is selected from the group consisting of alanine, valine, serine, and threonine;

(b) the carboxylic acid ester substrate is triacetin; wherein the amount of triacetin in the first component is designed to provide a final concentration of 0.5 wt % to 10 wt % in a reaction formulation formed by combining the first and second components;

(c) the buffer is in a concentration of about 0.1 wt % to about 10% wt of a the first component, wherein the buffer is selected from the group consisting of sodium bicarbonate, potassium bicarbonate, a mixture of sodium bicarbonate and potassium bicarbonate, sodium phosphate, potassium phosphate, and a mixture of sodium phosphate and potassium phosphate;

(d) the cosolvent is tripropylene glycol methyl ether and is in a concentration of up to 80 wt % of the first component;

(e) the surfactant is present and is polysorbate 80; and

(f) the hydrogen peroxide in the second component is present in an amount that provides a final concentration in a reaction formulation formed by combining the first and second components of from 0.33 wt % to about 30 wt %.

7. The method of claim 5 wherein the first component is combined with said second component at a ratio of about 1:1 to about 1:10 by weight to produce peroxycarboxylic acid.

8. A method of bleaching, destaining, deodorizing, sanitizing or disinfecting a surface, an article of clothing or a textile comprising:

(a) providing a formulation comprising:

(A) a first component comprising a formulation of

(i) about 0.1 wt % to about 5 wt % of the first component being an enzyme powder comprising:

(1) about 20 wt % to about 33 wt % of the enzyme powder comprising at least one CE-7 enzyme and having perhydrolysis activity, wherein said at least one CE-7 enzyme comprising an amino acid sequence selected from the group consisting of SEQ ID NO: 6, SEQ ID NO: 7, SEQ ID NO: 19 and SEQ ID NO: 20;

(2) about 67 wt % to about 80 wt % of the enzyme powder comprising at least one oligosaccharide excipient having a number average molecular weight of at least about 1250 and a weight average molecular weight of at least about 9000; wherein the at least one oligosaccharide excipient is selected from the group consisting of maltodextrin, xylan, mannan, fucoidan, galactomannan, chitosan, raffinose, stachyose, pectin, insulin, levan, graminan, amylopectin, and mixtures thereof;

(ii) a carboxylic acid ester substrate selected from the group consisting of monoacetin, diacetin, triacetin, and a mixtures thereof; wherein the amount of the carboxylic acid ester substrate in the first component is designed to provide a final concentration of 0.5 wt % to 10 wt % in a reaction formulation formed by combining the two reaction components;

(iii) about 0.1 wt % to about 10% wt of he first component comprising a buffer selected from the group consisting of sodium bicarbonate, potassium bicarbonate, a mixture of sodium bicarbonate and potassium bicarbonate, sodium phosphate, potassium phosphate, and a mixture of sodium phosphate and potassium phosphate;

(iv) up to 80 wt % of the first component comprising a cosolvent having a log P value of less than 2, said cosolvent selected from the group consisting of tripropylene glycol methyl ether, dipropylene glycol methyl ether, propylene glycol methyl ether, diethylene glycol butyl ether, dipropylene glycol, triethylene glycol, 1,2-propanediol, N-ethyl-2-pyrroldinone, isopropanol, ethanol, ethyl lactate, 1,3-propanediol, and any combination thereof; and

(v) at least one surfactant in a range from about 0.1 wt % to about 5 wt % based on the weight of total protein present in the enzyme powder; and

(B) a second component comprising

(i) at least 70 wt % water;

(ii) hydrogen peroxide in an amount that provides a final concentration of the hydrogen peroxide in the reaction formulation in range from 0.33 wt % to about 30 wt %; and

(iii) about 0.01 wt % to about 1% of a hydrogen peroxide stabilizer;

wherein combining said first and said second reaction components produces an aqueous formulation of peracetic acid;

(b) combining the first component with the second component whereby a reaction formulation comprising peroxycarboxylic acid is produced; and

(c) applying the reaction formulation to the surface, the article of clothing or the textile for bleaching, stain removal, odor reduction, sanitization, disinfection, or a combination thereof.
