View Patent ↗The present disclosure provides a binding protein comprising: a polypeptide heavy chain comprising: VH 1 -(X 1 )n-VH 2 —CH (X 2 )y wherein VH 1 is a first variable domain, VH 2 is a second variable domain, CH is a constant domain, X 1 represents an amino acid or peptide, X 2 represents an Fc region, n is 0 or 1 and y is independently 1 or 2, and a polypeptide light chain comprising: VL 1 -(X 1 )n-VL 2 -C wherein VL1 is a first variable domain, VL 2 is a second variable domain, C is a constant domain, X 1 represents an amino acid or peptide and n is 0 or 1, wherein the heavy chain and light chain are aligned such that VH 1 and VL 1 form a first binding domain, and VH 2 and VL 2 form a second binding domain and wherein: there is a disulfide bond between VH 1 and VL 1 , and/or there is a disulfide bond between VH 2 and VL 2 , and use thereof in treatment.
1. A binding protein, comprising:
a first polypeptide comprising:
VH 1 -X 1 -VH 2 -CH-X 2
wherein VH 1 is a first heavy chain variable domain, VH 2 is a second heavy chain variable domain, CH is a constant domain, X 1 represents a flexible peptide linker that is one of SEQ ID NOs: 10 to 79, X 2 represents an Fc region, and
a second polypeptide comprising:
VL 1 -X 1 -VL 2 -C
wherein VL 1 is a first light chain variable domain, VL 2 is a second light chain variable domain, C is a constant domain, and X 1 represents a flexible peptide linker that is one of SEQ ID NOs: 10 to 79,
wherein VH 1 and VL 1 form a first binding domain and VH 2 and VL 2 form a second binding domain; and
wherein there is a disulfide bond between VH 1 and VL 1 .
2. The binding protein of claim 1 , wherein the CH of the first polypeptide is linked to the C of the second polypeptide by a disulfide bond.
3. A pharmaceutically acceptable composition comprising the binding protein of claim 1 .
4. The binding protein of claim 1 or a pharmaceutically acceptable composition comprising the binding protein of claim 1 for use in treatment.
5. The binding protein of claim 1 , wherein the flexible peptide linker has a sequence corresponding to SEQ ID NO: 72.
6. The binding protein of claim 1 , wherein the disulfide bond is between VH37 and VL95, VH44 and VL100, VH44 and VL105, VH45 and VL87, VH100 and VL50, VH98 and VL46, VH105 and VL43 and VH106 and VL57.
