Abstract

Hairless, immunodeficient mice on a non-obese diabetic (NOD) background and methods for their production are disclosed herein. The mice are hairless and have multiple immunodeficiencies, including B-cell and T-cell deficiencies, as well as impaired macrophage and complement function. The mice also have a further deficit in natural killer and dendritic cells of the immune system. The mice are useful for biomedical research, for example, in studies involving xenograft transplantation, spontaneous tumors, cancer cell tumorigenesis, tumor angiogenesis, tumor metastatic potential, tumor suppression therapy, carcinogenesis regulation, and tumor imaging.

Claims (8)

1. A method of producing a transgenic, hairless, immunodeficient mouse whose genome comprises a homozygous Prkdc scid mutation and a homozygous Hr hr mutation on a non-obese diabetic NOD.CB17 background, the method comprising:

(a) crossing a homozygous NOD.CB17-Prkdc scid mouse with a homozygous MF-1-hr mouse to produce a progeny that is heterozygous for both the Hr allele and the Prkdc allele;

(b) crossing the heterozygous progeny produced from step (a) with NOD.CB17-Prkdc scid mice;

(c) selecting an offspring of step (b) that has a NOD.CB17 background, is homozygous for the Prkdc allele, and is heterozygous for the Hr allele;

(d) crossing the offspring selected in step (c) with mice homozygous or heterozygous for the Hr allele to obtain a homozygous NOD.CB17-Prkdc scid Hr hr double mutant transgenic mouse,

wherein the NOD.CB17-Prkdc scid Hr hr double mutant transgenic mouse exhibits B-cell, T-cell, NK-cell, macrophage, and complement deficiencies, and a reduction in the number of dendritic cells compared to an immunodeficient mouse of a NOD.CB17-Prkdc scid background that is not hairless.

2. A transgenic, hairless, immunodeficient mouse whose genome comprises a homozygous Prkdc scid mutation and a homozygous Hr hr mutation on a non-obese diabetic NOD.CB17 background,

wherein the mouse exhibits B-cell, T-cell, NK-cell, macrophage, and complement deficiencies and a reduction in the number of dendritic cells compared to an immunodeficient mouse of a NOD.CB17-Prkdc scid background that is not hairless.
