Compositions and methods for increasing bone mineralization
View Patent ↗Described herein are pharmaceutical compositions comprising an anti-sclerostin antibody and an inhibitor of bone resorption.
1. A pharmaceutical composition comprising (a) an antibody, or an antibody fragment thereof, wherein the antibody or antibody fragment binds to a polypeptide encoded by SEQ ID NO: 1 with an affinity K a of greater than or equal to 10 7 M −1 , and (b) an inhibitor of bone resorption.
2. The pharmaceutical composition of claim 1 , wherein the inhibitor of bone resorption is selected from the group consisting of calcitonin, estrogen, a bisphosphonate, a growth factor having anti-resorptive activity and tamoxifen.
3. The pharmaceutical composition according to claim 1 , wherein the inhibitor of bone resorption is a bisphosphonate.
4. The pharmaceutical composition according to claim 1 , wherein the inhibitor of bone resorption is estrogen.
5. The pharmaceutical composition according to claim 1 , wherein the antibody fragment is selected from the group consisting of F(ab′) 2 , F(ab) 2 , Fab′, Fab, and Fv.
6. The pharmaceutical composition according to claim 1 , wherein the antibody is selected from the group consisting of a murine monoclonal antibody, a human monoclonal antibody, a humanized monoclonal antibody, and an antibody fragment of any of the foregoing.
7. The pharmaceutical composition according to any one of claims 1 - 6 , further comprising a pharmaceutically acceptable carrier, excipient or diluent.

A method of increasing bone mineral content in a human comprising administering to the human (a) an antibody, or an antibody fragment thereof, wherein the antibody binds to a polypeptide encoded by SEQ ID NO: 1 with an affinity K a of greater than or equal to 10 7 M −1 , and (b) an inhibitor of bone resorption, in amounts effective to increase bone mineral content.
9. The method of claim 8 , wherein the inhibitor of bone resorption is selected from the group consisting of calcitonin, estrogen, a bisphosphonate, a growth factor having anti-resorptive activity and tamoxifen.
10. The method of claim 8 , wherein the inhibitor of bone resorption is a bisphosphonate.
11. The method of claim 8 , wherein the inhibitor of bone resorption is estrogen.
