Abstract

The invention relates generally to compositions and methods for purifying the desired species from a mixture of desired heterodimer and contaminating homodimer immunoglobulin variants by modifying the isoelectric point(s) of the individual chains.

Claims (6)

1. A composition comprising a heterodimeric polypeptide comprising:

a) a first variant human IgG1 heavy chain constant region comprising amino acid modifications as compared to a human IgG1 heavy chain constant region according to SEQ ID NO: 2, wherein said modifications comprise Q196K, 1199T, P217R, P228R, and N276K, according to the EU index as in Kabat; and

b) a second variant human IgG1 heavy chain constant region comprising amino acid modifications as compared to a human IgG1 heavy chain constant region according to SEQ ID NO: 2, wherein said modifications comprise N203D, K274Q, R355Q, Q419E, and K447del, according to the EU index as in Kabat,

and wherein said first variant human IgG1 heavy chain constant region and said second variant human IgG1 heavy chain constant region each possess at least 95% sequence identity to SEQ ID NO: 2.

2. A nucleic acid composition comprising a first nucleic acid encoding said first variant human IgG1 heavy chain constant region of claim 1 , and a second nucleic acid encoding said second variant human IgG1 heavy chain constant region of claim 1 .

3. An isolated host cell comprising the nucleic acid composition of claim 2 .
