Abstract

The invention provides a non-naturally occurring microbial organism having a 6-aminocaproic acid, caprolactam, hexametheylenediamine or levulinic acid pathway. The microbial organism contains at least one exogenous nucleic acid encoding an enzyme in the respective 6-aminocaproic acid, caprolactam, hexametheylenediamine or levulinic acid pathway. The invention additionally provides a method for producing 6-aminocaproic acid, caprolactam, hexametheylenediamine or levulinic acid. The method can include culturing a 6-aminocaproic acid, caprolactam or hexametheylenediamine producing microbial organism, where the microbial organism expresses at least one exogenous nucleic acid encoding a 6-aminocaproic acid, caprolactam, hexametheylenediamine or levulinic acid pathway enzyme in a sufficient amount to produce the respective product, under conditions and for a sufficient period of time to produce 6-aminocaproic acid, caprolactam, hexametheylenediamine or levulinic acid.

Claims (15)

1. A non-naturally occurring microbial organism having a hexamethylenediamine pathway comprising at least two exogenous nucleic acids each encoding a hexamethylenediamine pathway enzyme expressed in a sufficient amount to produce hexamethylenediamine, said hexamethylenediamine pathway comprising a 6-aminocaproyl-CoA/acyl-CoA transferase or a 6-aminocaproyl-CoA synthase; 6-aminocaproyl-CoA reductase (aldehyde forming); and a hexamethylenediamine transaminase or a hexamethylenediamine dehydrogenase, and wherein the microbial organism is selected from the group consisting of bacteria, yeast and fungus.

2. The non-naturally occurring microbial organism of claim 1 , wherein said microbial organism comprises said hexamethylenediamine pathway (i) and further comprises a 6-aminocaproic acid pathway.

3. The non-naturally occurring microbial organism of claim 1 , wherein said microbial organism comprises two, three, four, five, six, seven or eight exogenous nucleic acids each encoding a hexamethylenediamine pathway enzyme.

4. The non-naturally occurring microbial organism of claim 1 , wherein said at least one exogenous nucleic acid is a heterologous nucleic acid.

5. The non-naturally occurring microbial organism of claim 1 , wherein said non-naturally occurring microbial organism is in a substantially anaerobic culture medium.

6. The non-naturally occurring microbial organism of claim 1 further comprising one or more metabolic modifications of Table 16.

7. The non-naturally occurring microbial organism of claim 6 , wherein said microbial organism comprises a set of metabolic modifications selected from one of design ID 1 to design ID 877, or a combination thereof.

8. The non-naturally occurring microbial organism of claim 1 further having an adipyl-CoA pathway and comprising an exogenous nucleic acid encoding a thiolase, wherein said thiolase selectively produces 3-oxoadipyl-CoA in comparison to acetoacetyl-CoA.

The non-naturally occurring microbial organism of claim 8 , wherein said thiolase is selected from the group consisting of the beta-ketothiolase encoded by the phaD gene of Pseudomonas putida (KT2440), the beta-ketoadipyl-CoA thiolase encoded by the pcaF gene of Pseudomonas aeruginosa PAO1, and the 3-oxoadipyl-CoA thiolase encoded by the bkt gene of Burkholderia ambifaria AMMD, or an ortholog thereof having thiolase activity.

10. The non-naturally occurring microbial organism of claim 9 , wherein said exogenous nucleic acid comprises a nucleic acid sequence selected from SEQ ID NO: 20, 38 and 47.

11. Culture medium comprising hexamethylenediamine, wherein said hexamethylenediamine was produced by the non-naturally occurring microbial organism of claim 1 .

12. A method for producing hexamethylenediamine, comprising culturing the non-naturally occurring microbial organism of claim 1 under conditions and for a sufficient period of time to produce hexamethylenediamine.

13. The method of claim 12 , wherein said method further comprises separating the hexamethylenediamine from other components in the culture.

14. A method for production of hexamethylenediamine by the microbial organism having a hexamethylenediamine pathway in which adipyl-CoA is an intermediate in said pathway, comprising culturing the microbial organism of claim 8 .

15. The method of claim 14 , wherein said method further comprises separating the hexamethylenediamine from other components in the culture.
