Abstract

Compositions and methods are provided for enzymes with altered properties that involve a systematic approach to mutagenesis and a screening assay that permits selection of the desired proteins. Embodiments of the method are particularly suited for modifying specific properties of restriction endonucleases such as star activity. The compositions includes restriction endonucleases with reduced star activity as defined by an overall fidelity index improvement factor.

Claims (10)

1. A composition comprising a variant BamHI restriction endonuclease having reduced star activity, wherein the variant BamHI restriction endonuclease comprises an amino acid sequence that differs from the amino acid sequence of the parent BamHI restriction endonuclease by no more than seven mutations and comprises a mutation at an amino acid position corresponding to position 165 in SEQ ID NO:19.

2. A composition according to claim 1 , wherein the mutation at an amino acid position corresponding to position 165 in SEQ ID NO:19 is Y165F.

3. A composition comprising a variant BamHI restriction endonuclease having reduced star activity, wherein the variant BamHI restriction endonuclease comprises an amino acid sequence that differs from the amino acid sequence of the parent BamHI restriction endonuclease by no more than seven mutations and comprises a mutation at an amino acid position corresponding to position 167 in SEQ ID NO:19.

4. The composition of claim 3 , wherein the mutation at an amino acid position corresponding to position 167 in SEQ ID NO:19 is selected from E167A; E167R; E167K; E167L; E167I; and E167T.

A composition comprising a variant BamHI restriction endonuclease having reduced star activity, wherein the variant BamHI restriction endonuclease comprises an amino acid sequence that differs from the amino acid sequence of the parent BamHI restriction endonuclease by no more than seven mutations and comprises a mutation at an amino acid position corresponding to position 87 in SEQ ID NO:19.

6. The composition of claim 5 , wherein the mutation at an amino acid position corresponding to position 87 in SEQ ID NO:19 is selected from K87A; K87E; K87V; K87N; K87T; K87R; K87P; K87S; K87G; and K87W.

A composition comprising a variant BamHI restriction endonuclease having reduced star activity, wherein the variant BamHI restriction endonuclease comprises an amino acid sequence that differs from the amino acid sequence of the parent BamHI restriction endonuclease by no more than seven mutations and comprises a mutation at an amino acid position corresponding to position 86 in SEQ ID NO:19.

8. The composition of claim 7 , wherein the mutation at an amino acid position corresponding to position 86 in SEQ ID NO:19 is selected from E86A and E86P.

A composition comprising a variant BamHI restriction endonuclease having reduced star activity, wherein the variant BamHI restriction endonuclease comprises an amino acid sequence that differs from the amino acid sequence of the parent BamHI restriction endonuclease by no more than seven mutations and comprises a mutation at an amino acid position corresponding to position 30 in SEQ ID NO:19.

10. The composition of claim 9 , wherein the mutation at an amino acid position corresponding to position 30 in SEQ ID NO:19 is K30A.
