Abstract

Crystalline forms of (1r,4r)-6′-fluoro-N,N-dimethyl-4-phenyl-4′,9′-dihydro-3′H-spiro[cyclohexane-1,1′-pyrano[3,4,b]indol]-4-amine, pharmaceutical compositions and medicaments comprising these crystalline modifications, the use of these modifications as well as a process for the enrichment of such crystalline modifications.

Claims (41)

1. A pharmaceutical composition comprising at least one pharmaceutically acceptable additive and/or adjuvant and about 25 μg to about 600 μg of an active ingredient that comprises a crystalline form of (1r,4r)-6′-fluoro-N,N-dimethyl-4-phenyl-4′,9′-dihydro-3′H-spiro-[cyclohexane-1,1′-pyrano-[3,4,b]indol]-4-amine having an X-ray powder diffraction pattern comprising characteristic peaks at 7.8±0.2 degrees 2θ and at 31.6±0.2 degrees 2θ, and wherein the active ingredient comprises (1r,4r)-6′-fluoro-N,N-dimethyl-4-phenyl-4′,9′-dihydro-3′H-spiro-[cyclohexane-1,1′-pyrano-[3,4,b]indol]-4-amine at a diasteromeric excess of about 90% de.

2. The pharmaceutical composition of claim 1 , wherein the crystalline form further comprises a characteristic peak at 11.7±0.2 degrees 2θ.

3. The pharmaceutical composition of claim 1 , wherein the crystalline form further comprises a characteristic peak at 18.3±0.2 degrees 2θ.

4. The pharmaceutical composition of claim 1 , wherein the crystalline form further comprises characteristic peaks at 8.8±0.2 degrees 2θ and/or at 15.8±0.2 degrees 2θ.

5. The pharmaceutical composition of claim 1 , wherein the crystalline form further comprises characteristic peaks at about 20.4±0.2 degrees 2θ and/or at 23.3±0.2 degrees 2θ.

6. The pharmaceutical composition of claim 1 , wherein the crystalline form further comprises characteristic peaks at 11.7±0.2 degrees 2θ, at one or both of 8.8±0.2 degrees 2θ and/or 15.8±0.2 degrees 2θ, and at one or both of 20.4±0.2 degrees 2θ and/or 23.3±0.2 degrees 2θ.

7. The pharmaceutical composition of claim 1 , wherein the crystalline form has an endothermal event with a peak temperature at about 298-308° C., as determined by DSC.

8. The pharmaceutical composition of claim 1 , wherein the crystalline form has a Raman peak at about 1569±2 cm−1 and/or at about 1002±2 cm−1.

9. The pharmaceutical composition of claim 1 , wherein the active ingredient comprises a (1r,4r)-6′-fluoro-N,N-dimethyl-4-phenyl-4′,9′-dihydro-3′H-spiro-[cyclohexane-1,1′-pyrano-[3,4,b]indol]-4-amine at a diasteromeric excess of at least about 95%de.

10. The pharmaceutical composition of claim 1 , wherein the active ingredient comprises a (1r,4r)-6′-fluoro-N,N-dimethyl-4-phenyl-4′,9′-dihydro-3′H-spiro-[cyclohexane-1,1′-pyrano-[3,4,b]indol]-4-amine at a diasteromeric excess of at least about 97%de.

11. The pharmaceutical composition of claim 1 , wherein the active ingredient comprises a (1r,4r)-6′-fluoro-N,N-dimethyl-4-phenyl-4′,9′-dihydro-3′H-spiro-[cyclohexane-1,1′-pyrano-[3,4,b]indol]-4-amine at a diasteromeric excess of at least about 99%de.

The pharmaceutical composition of claim 1 , wherein the crystalline form is present in the active ingredient in an amount of at least about 60 wt. % relative to the total weight of all crystalline and non-crystalline forms of (1r,4r-6′-fluoro-N,N-dimethyl-4-phenyl-4′,9′-dihydro-3′H-spiro-[cyclohexane-1,1′-pyrano-[3,4,b]indol]-4-amine.

The pharmaceutical composition of claim 1 , wherein the crystalline form is present in the active ingredient in an amount of at least about 80 wt. % relative to the total weight of all crystalline and non-crystalline forms of (1r,4r-6′-fluoro-N,N-dimethyl-4-phenyl-4′,9′-dihydro-3′H-spiro-[cyclohexane-1,1′-pyrano-[3,4,b]indol]-4-amine.

The pharmaceutical composition of claim 1 , wherein the crystalline form is present in the active ingredient in an amount of at least about 90 wt. % relative to the total weight of all crystalline and non-crystalline forms of (1r,4r-6′-fluoro-N,N-dimethyl-4-phenyl-4′,9′-dihydro-3′H-spiro-[cyclohexane-1,1′-pyrano-[3,4,b]indol]-4-amine.

The pharmaceutical composition of claim 1 , wherein the crystalline form is present in the active ingredient in an amount of at least about 95 wt. % relative to the total weight of all crystalline and non-crystalline forms of (1r,4r)-6′-fluoro-N,N-dimethyl-4-phenyl-4′,9′-dihydro-3′H-spiro-[cyclohexane-1,1′-pyrano-[3,4,b]indol]-4-amine.

16. The pharmaceutical composition of claim 1 , wherein the pharmaceutical composition comprises 40±20 μg, 100±10 μg or 400±200 μg of the active ingredient.

17. The pharmaceutical composition of claim 13 , wherein the pharmaceutical composition comprises 40±20 μg, 100±10 μg or 400±200 μg of the active ingredient.

18. The pharmaceutical composition of claim 1 , wherein the pharmaceutical composition contains at most about 1.0 wt.-% 4-dimethylamino-4-phenylcyclohexanone, relative to the total content of 6′-fluoro-N,N-dimethyl-4-phenyl-4′,9′-dihydro-3′H-spiro[cyclohexane-1,1′-pyran [3,4b]indol]-4-amine.

19. The pharmaceutical composition of claim 13 , wherein the pharmaceutical composition contains at most about 1.0 wt.-% 4-dimethylamino-4-phenylcyclohexanone, relative to the total content of 6′-fluoro-N,N-dimethyl-4-phenyl-4′,9′-dihydro-3′H-spiro[cyclohexane-1,1′-pyran [3,4b]indol]-4-amine.

A pharmaceutical composition comprising at least one pharmaceutically acceptable additive and/or adjuvant and about 25 μg to about 600 μg of an active ingredient that comprises a crystalline form of (1r,4r)-6′-fluoro-N,N-dimethyl-4-phenyl-4′,9′-dihydro-3′H-spiro-[cyclohexane-1,1′-pyrano-[3,4,b]indol]-4-amine having an X-ray powder diffraction pattern comprising characteristic peaks at 7.8±0.2 degrees 2θ and at 31.6±0.2 degrees 2θ, and wherein the crystalline form is present in the active ingredient in an amount of at least about 60 wt. % relative to the total weight of all crystalline and non-crystalline forms of (1r,4r)-6′-fluoro-N,N-dimethyl-4-phenyl-4′,9′-dihydro-3′H-spiro-[cyclohexane-1,1′-pyrano-[3,4,b]indol]-4-amine.

21. The pharmaceutical composition of claim 20 , wherein the crystalline form further comprises a characteristic peak at 11.7±0.2 degrees 2θ.

22. The pharmaceutical composition of claim 20 , wherein the crystalline form further comprises a characteristic peak at 18.3±0.2 degrees 2θ.

23. The pharmaceutical composition of claim 20 , wherein the crystalline form further comprises characteristic peaks at 8.8±0.2 degrees 2θ and/or at 15.8±0.2 degrees 2θ.

24. The pharmaceutical composition of claim 20 , wherein the crystalline form further comprises characteristic peaks at about 20.4±0.2 degrees 2θ and/or at 23.3±0.2 degrees 2θ.

25. The pharmaceutical composition of claim 20 , wherein the crystalline form further comprises characteristic peaks at 11.7±0.2 degrees 2θ, at one or both of 8.8±0.2 degrees 2θ and/or 15.8±0.2 degrees 2θ, and at one or both of 20.4±0.2 degrees 2θ and/or 23.3±0.2 degrees 2θ.

26. The pharmaceutical composition of claim 20 , wherein the crystalline form has an endothermal event with a peak temperature at about 298-308° C., as determined by DSC.

27. The pharmaceutical composition of claim 20 , wherein the crystalline form has a Raman peak at about 1569±2 cm−1 and/or at about 1002±2 cm−1.

The pharmaceutical composition of claim 20 , wherein the crystalline form is present in the active ingredient in an amount of at least about 70 wt. % relative to the total weight of all crystalline and non-crystalline forms of (1r,4r)-6′-fluoro-N,N-dimethyl-4-phenyl-4′,9′-dihydro-3′H-spiro-[cyclohexane-1,1′-pyrano-[3,4,b]indol]-4-amine.

The pharmaceutical composition of claim 20 , wherein the crystalline form is present in the active ingredient in an amount of at least about 80 wt. % relative to the total weight of all crystalline and non-crystalline forms of (1r,4r)-6′-fluoro-N,N-dimethyl-4-phenyl-4′,9′-dihydro-3′H-spiro-[cyclohexane-1,1′-pyrano-[3,4,b]indol]-4-amine.

30. The pharmaceutical composition of claim 20 , wherein the crystalline form is present in the active ingredient in an amount of at least about 90 wt. % relative to the total weight of all crystalline and non-crystalline forms of (1r,4r)-6′-fluoro-N,N-dimethyl-4-phenyl-4′,9′-dihydro-3′H-spiro-[cyclohexane-1,1′-pyrano-[3,4,b]indol]-4-amine.

The pharmaceutical composition of claim 20 , wherein the crystalline form is present in the active ingredient in an amount of at least about 95 wt. % relative to the total weight of all crystalline and non-crystalline forms of (1r,4r)-6′-fluoro-N,N-dimethyl-4-phenyl-4′,9′-dihydro-3′H-spiro-[cyclohexane-1,1′-pyrano-[3,4,b]indol]-4-amine.

32. The pharmaceutical composition of claim 20 , wherein the pharmaceutical composition contains at most about 1.0 wt.-% 4-dimethylamino-4-phenylcyclohexanone, relative to the total content of 6′-fluoro-N,N-dimethyl-4-phenyl-4′,9′-dihydro-3′H-spiro[cyclohexane-1,1′-pyrano[3,4b]indol]-4-amine.

A pharmaceutical composition comprising at least one pharmaceutically acceptable additive and/or adjuvant and about 25 μg to about 600 μg of an active ingredient that comprises a crystalline form of (1r,4r)-6′-fluoro-N,N-dimethyl-4-phenyl-4′,9′-dihydro-3′H-spiro-[cyclohexane-1,1′-pyrano-[3,4,b]indol]-4-amine having an X-ray powder diffraction pattern comprising characteristic peaks at 7.8±0.2 degrees 2θ and at 31.6±0.2 degrees 2θ, wherein the pharmaceutical composition contains at most about 1.0 wt.-% 4-dimethylamino-4-phenylcyclohexanone, relative to the total content of 6′-fluoro-N,N-dimethyl-4-phenyl-4′,9′-dihydro-3′H-spiro[cyclohexane-1,1′-pyrano[3,4b]indol]-4-amine.

34. The pharmaceutical composition of claim 33 , wherein the crystalline form further comprises a characteristic peak at 11.7±0.2 degrees 2θ.

35. The pharmaceutical composition of claim 33 , wherein the crystalline form further comprises a characteristic peak at 18.3±0.2 degrees 2θ.

36. The pharmaceutical composition of claim 33 , wherein the crystalline form further comprises characteristic peaks at 8.8±0.2 degrees 2θ and/or at 15.8±0.2 degrees 2θ.

37. The pharmaceutical composition of claim 33 , wherein the crystalline form further comprises characteristic peaks at about 20.4±0.2 degrees 2θ and/or at 23.3±0.2 degrees 2θ.

38. The pharmaceutical composition of claim 33 , wherein the crystalline form further comprises characteristic peaks at 11.7±0.2 degrees 2θ, at one or both of 8.8±0.2 degrees 2θ and/or 15.8±0.2 degrees 2θ, and at one or both of 20.4±0.2 degrees 2θ and/or 23±0.2 degrees 2θ.

39. A method for treating chronic pain in a patient, comprising orally administering to a patient diagnosed with a chronic pain disorder the pharmaceutical composition of claim 1 .

40. A method for treating chronic pain in a patient, comprising orally administering to a patient diagnosed with a chronic pain disorder the pharmaceutical composition of claim 20 .

41. A method for treating chronic pain in a patient, comprising orally administering to a patient diagnosed with a chronic pain disorder the pharmaceutical composition of claim 33 .
