Abstract

The present invention relates to isolated polypeptides having xylanase activity and polynucleotides encoding the polypeptides. The invention also relates to nucleic acid constructs, vectors, and host cells comprising the polynucleotides as well as methods of producing and using the polypeptides.

Claims (30)

1. A process for degrading a cellulosic material, comprising treating the cellulosic material with a composition comprising a polypeptide having xylanase activity, a cellobiohydrolase I, a cellobiohydrolase II, an endoqlucanase, a beta-qlucosidase, and a GH61 polypeptide, wherein the polypeptide having xylanase activity is selected from the group consisting of:

(a) a polypeptide having at least 95% sequence identity to the sequence of amino acids 18-349 of SEQ ID NO: 2;

(b) a polypeptide encoded by a polynucleotide that hybridizes under high stringency conditions with (i) the mature polypeptide coding sequence of SEQ ID NO: 1, (ii) the cDNA sequence thereof, or (iii) the full-length complement of (i) or (ii), wherein high stringency conditions are defined as prehybridization and hybridization at 42° C. in 5×SSPE, 0.3% SDS, 200 micrograms/ml sheared and denatured salmon sperm DNA, and 50% formamide, and washing three times each for 15 minutes in 2×SSC, 0.2% SDS at 65° C.; and

(c) a fragment of the sequence of amino acids 18-349 of SEQ ID NO: 2 that has xylanase activity.

2. A process for producing a fermentation product, comprising:

(a) saccharifying a cellulosic material with a composition comprising a polypeptide having xylanase activity, a cellobiohydrolase I, a cellobiohydrolase II, an endoqlucanase, a beta-qlucosidase, and a GH61 polypeptide, wherein the polypeptide having xylanase activity is selected from the group consisting of:

(1) a polypeptide having at least 95% sequence identity to the sequence of amino acids 18-349 of SEQ ID NO: 2;

(2) a polypeptide encoded by a polynucleotide that hybridizes under high stringency conditions with (i) the mature polypeptide coding sequence of SEQ ID NO: 1, (ii) the cDNA sequence thereof, or (iii) the full-length complement of (i) or (ii), wherein high stringency conditions are defined as prehybridization and hybridization at 42° C. in 5×SSPE, 0.3% SDS, 200 micrograms/ml sheared and denatured salmon sperm DNA, and 50% formamide, and washing three times each for 15 minutes in 2×SSC, 0.2% SDS at 65° C.; and

(3) a fragment of the sequence of amino acids 18-349 of SEQ ID NO: 2 that has xylanase activity; and

(b) fermenting the saccharified cellulosic material with one or more fermenting microorganisms to produce the fermentation product.

3. The process of claim 2 , further comprising recovering the fermentation product.

A process for degrading a cellulosic material, comprising treating the cellulosic material with a composition comprising a cellobiohydrolase I, a cellobiohydrolase II, an endoglucanase, a beta-glucosidase, a GH61 polypeptide, and a polypeptide comprising a catalytic domain having at least 95% sequence identity to amino acids 18 to 349 of SEQ ID NO: 2, wherein the catalytic domain has xylanase activity.

5. A process for producing a fermentation product, comprising:

(a) saccharifying a cellulosic material with a composition comprising a cellobiohydrolase I, a cellobiohydrolase II, an endoglucanase, a beta-glucosidase, a GH61 polypeptide, and a polypeptide comprising a catalytic domain having at least 95% sequence identity to amino acids 18 to 349 of SEQ ID NO: 2, wherein the catalytic domain has xylanase activity; and

(b) fermenting the saccharified cellulosic material with one or more fermenting microorganisms to produce the fermentation product.

6. The process of claim 5 , further comprising recovering the fermentation product.

7. The process of claim 1 , wherein the polypeptide having xylanase activity has at least 97% sequence identity to the sequence of amino acids 18-349 of SEQ ID NO: 2.

The process of claim 1 , wherein the polypeptide having xylanase activity is a variant of the sequence of amino acids 18-349 of SEQ ID NO: 2 comprising a substitution, deletion, and/or insertion at one or more positions and wherein the variant has at least 95% sequence identity to the sequence of amino acids 18-349 of SEQ ID NO: 2.

9. The process of claim 1 , wherein the polypeptide having xylanase activity comprises the sequence of amino acids 18-349 of SEQ ID NO: 2.

10. The process of claim 1 , further comprising recovering the degraded cellulosic material.

11. The process of claim 2 , wherein the polypeptide having xylanase activity has at least 97% sequence identity to the sequence of amino acids 18-349 of SEQ ID NO: 2.

The process of claim 2 , wherein the polypeptide having xylanase activity is a variant of the sequence of amino acids 18-349 of SEQ ID NO: 2 comprising a substitution, deletion, and/or insertion at one or more positions and wherein the variant has at least 95% sequence identity to the sequence of amino acids 18-349 of SEQ ID NO: 2.

13. The process of claim 2 , wherein the polypeptide having xylanase activity comprises the sequence of amino acids 18-349 of SEQ ID NO: 2.

14. The process of claim 2 , wherein steps (a) and (b) are performed simultaneously.

15. The process of claim 4 , further comprising recovering the degraded cellulosic material.

16. The process of claim 4 , wherein the polypeptide comprising a catalytic domain comprises amino acids 18 to 349 of SEQ ID NO: 2, wherein the catalytic domain has xylanase activity.

17. The process of claim 4 , wherein the polypeptide comprising a catalytic domain further comprises a cellulose binding domain.

18. The process of claim 5 , wherein steps (a) and (b) are performed simultaneously.

19. The process of claim 5 , wherein the polypeptide comprising a catalytic domain comprises amino acids 18 to 349 of SEQ ID NO: 2, wherein the catalytic domain has xylanase activity.

20. The process of claim 5 , wherein the polypeptide comprising a catalytic domain further comprises a cellulose binding domain.
