Abstract

A kinase is used in a biological indicator for validation of treatment processes designed to reduce the amount or activity of a biological agent in a sample. The indication can be used for validation of sterilization treatment. The formation of ATP from a substrate comprising ADP is measured via the liciferin/luciferate system in a luminameter. Thermostable adenylate kinase from sulfolobus acidocaldarius is especially suitable for the validation of procedures to inactivate transmissable spongiform encephalopathy agents.

Claims (30)

1. A biological process indicator, comprising:

(i) a thermostable kinase, wherein the thermostable kinase retains at least 95% of its kinase activity after exposure to 70° C. for 30 minutes; and

(ii) a matrix, wherein the matrix is a non-infectious biofilm, and wherein the thermostable kinase is dispersed within the matrix; or

(iii) a solid support, wherein the solid support is:

(a) a plastic, ceramic, or metallic surface, or an indicator strip, dip stick or bead, wherein the thermostable kinase is immobilised inside the solid support or wherein the thermostable kinase is immobilised directly onto the solid support, via non-specific adsorption or via chemical cross-linking; or

(b) a tube, wherein the thermostable kinase is attached directly to an internal face of the tube via non-specific adsorption or via chemical cross-linking.

2. The biological process indicator according to claim 1 , wherein the thermostable kinase catalyses formation of ATP from a substrate comprising ADP.

3. The biological process indicator according to claim 1 , wherein the thermostable kinase retains at least 95% of its kinase activity after exposure to 80° C. for 10 minutes.

4. The biological process indicator according to claim 1 , wherein the thermostable kinase is an adenylate kinase, acetate kinase or pyruvate kinase.

5. The biological process indicator according to claim 1 , wherein the thermostable kinase is a trimeric adenylate kinase or a monomeric adenylate kinase.

6. The biological process indicator according to claim 1 , wherein the thermostable kinase is a trimeric adenylate kinase.

7. The biological process indicator according to claim 1 , wherein the thermostable kinase is a trimeric adenylate kinase from a Sulfolobus species or a monomeric adenylate kinase from a Thermotoga species.

The biological process indicator according to claim 1 , wherein the thermostable kinase is selected from the group consisting of an adenylate kinase from Alicyclobacillus acidocaldarius, Archaeoglobus fulgidus, Aeropyrum pernix, Aquifex pyrophilus, Bacillus caldotenax BT1, Bacillus species PS3, Bacillus stearothermophilus 11057, Bacillus stearothermophilus 12001, Bacillus thermocatenulatus, Clostridium stercocorarium, Methanococcus species, Meiothermus ruber, Pyrococcus abyssi, Pyrococcus furiosus, Pyroccoccus horikoshii, Pyrococcus woesei, Rhodothermus marinus, Sulfolobus acidocaldarius, Sulfolobus shibatae, Sulfolobus solfataricus, Thermoanaerobacter ethanolicus, Thermoanaerobacter thermosulfurogenes, Thermobrachium celere, Thermococcus litoralis, Thermus aquaticus YT1, Thermus caldophilus GK24, Thermus thermophilus HB8, Thermatoga maritima and Thermatoga neapolitana.

9. The biological process indicator according to claim 1 , wherein the thermostable kinase has an amino acid sequence selected from the group consisting of SEQ ID NOs: 1-25 or is encoded by a nucleic acid sequence selected from the group consisting of SEQ ID NOs: 26-30.

10. The biological process indicator according to claim 9 , wherein the thermostable kinase has an amino acid sequence selected from the group consisting of SEQ ID NOs: 17-19.

11. The biological process indicator according to claim 1 wherein the solid support is (a) the matrix.

12. The biological process indicator according to claim 1 , wherein the solid support is (b) the indicator strip, the dip stick or the bead.

13. The biological process indicator according to claim 1 , further comprising an agent to stabilise the thermostable kinase.

14. The biological process indicator according to claim 12 , wherein the agent is selected from the group consisting of metal ions, sugars, sugar alcohols and gel-forming agents.

15. The biological process indicator according to claim 1 , further comprising means for attaching the biological process indicator to a surface.

16. The biological process indicator according to claim 1 , wherein the means for attaching the biological process indicator to a surface comprises a projection, recess or aperture for attachment of the indicator to the surface by a screw, a nut and bolt, or a clamp.

17. A kit for validating a treatment process for reducing the amount or activity of a biological agent in a sample comprising:

(i) the biological process indicator according to claim 1 ; and

(ii) a substrate for the thermostable kinase.

18. The kit according to claim 17 , further comprising means for detecting ATP.

19. The kit according to claim 17 , further comprising luciferin and luciferase.

20. The kit according to claim 17 , further comprising a luminometer.

21. The kit according to claim 17 , further comprising a look-up table correlating activity of the thermostable kinase of the biological process indicator with a reduction in an amount or activity of the biological agent.

22. The kit according to claim 17 , wherein the biological agent is selected from the group consisting of bacteria, viruses, spores, proteins, peptides and a TSE agent.

23. The kit according to claim 17 , wherein the kit is portable.
