Unnatural reactive amino acid genetic code additions
This invention provides compositions and methods for producing translational components that expand the number of genetically encoded amino acids in eukaryotic cells. The components include orthogonal tRNAs, orthogonal aminoacyl-tRNA synthetases, orthogonal pairs of tRNAs/synthetases and unnatural amino acids. Proteins and methods of producing proteins with unnatural amino acids in eukaryotic cells are also provided.
1. A composition comprising a protein, an antibody or antibody fragment, wherein the protein, antibody or antibody fragment comprises at least one unnatural amino acid comprising a reactive group for the attachment of a molecule by a cycloaddition reaction, wherein the at least one unnatural amino acid is incorporated site-specifically into the protein, antibody or antibody fragment in vivo in a eukaryotic cell using an orthogonal translation system; and
wherein the at least one unnatural amino acid is incorporated site-specifically at any site other than a lysine or cysteine site in the protein, antibody or antibody fragment.
2. The composition of claim 1 , wherein the cycloaddition reaction is a [3+2] cycloaddition reaction.
3. The composition of claim 1 , wherein the molecule is selected from the group consisting of a small organic molecule, a cytotoxic compound, a second protein or polypeptide, and a prodrug.
4. The composition of claim 1 , wherein the molecule comprises a radionuclide.
5. The composition of claim 1 , wherein the at least one unnatural amino acid is a cyclic unnatural amino acid.
6. The composition of claim 1 , wherein the at least one unnatural amino acid is p-azido-L-phenylalanine, p-benzoyl-phenylalanine or p-propargyloxyphenylalanine.
7. The composition of claim 1 , wherein the at least one unnatural amino acid is a photoreactive unnatural amino acid.
8. The composition of claim 7 , wherein the photoreactive unnatural amino acid comprises a p-azido-L-phenylalanine, a p-benzoyl-phenylalanine or a p-propargyloxyphenylalanine.
9. The composition of claim 1 , wherein the at least one unnatural amino acid comprises a p-propargyloxyphenylalanine.
10. The composition of claim 1 , wherein the antibody or antibody fragment is selected from the group consisting of monoclonal antibodies, chimeric antibodies, humanized antibodies, Fab fragments, fragments produced by an Fab expression library, and single chain antibody fragments.
11. The composition of claim 1 , wherein the protein, antibody or antibody fragment specifically binds a tumor-specific antigen, or wherein the antibody or antibody fragment mediates antibody-dependent cell-mediated cytotoxicity (ADCC) or complement-mediated lysis (CML).
