Abstract

The present disclosure provides compositions and methods relating to the evaluation of BAFF in a biological sample from a subject.

Claims (34)

1. A method of identifying a subject who is a candidate for a BAFF antagonist therapy, the method comprising:

(a) contacting a biological sample from the subject with an anti-BAFF antibody that binds BAFF independent of its physiological state;

(b) evaluating the level of total BAFF bound by the antibody; and

(c) selecting a subject who has a greater level of total BAFF bound by the antibody, compared to a control,

wherein a greater level of total BAFF bound by the antibody, compared to a control, indicates that the subject is a candidate for a BAFF antagonist therapy.

2. The method of claim 1 , wherein the anti-BAFF antibody is selected from the group consisting of:

(a) antibody B4H7,

(b) an antibody that competitively inhibits antibody B4H7,

(c) an antibody that is competitively inhibited from binding to BAFF by antibody B4H7, and

(d) an antibody that binds to the same epitope as antibody B4H7.

3. The method of claim 1 , wherein the BAFF antagonist therapy is selected from the group consisting of: an inhibitory anti-BAFF antibody, a soluble BAFF receptor or Fc fusion protein thereof, and an anti-BAFF receptor antibody.

The method of claim 1 , wherein the subject has an autoimmune disease selected from the group consisting of: autoimmune rheumatologic disorders, autoimmune gastrointestinal and liver disorders, vasculitis, autoimmune neurological disorders, autoimmune dermatologic disorders, autoimmune endocrine disorders, renal disorders, and hematologic disorders.

5. The method of claim 1 , wherein the biological sample is selected from the group consisting of: blood, tissue, plasma, synovial fluid, BAL and CSF.

6. The method of claim 1 , wherein the subject has a normal level of free soluble BAFF compared to the control.

7. A method of identifying a subject who is a candidate for a BAFF antagonist therapy, the method comprising:

(a) contacting a biological sample from the subject with a first anti-BAFF antibody, which binds BAFF independent of its physiological state;

(b) contacting the biological sample with a second anti-BAFF antibody, which preferentially binds free BAFF over BAFF bound to receptor; and

(c) comparing the binding of the first antibody and the second antibody, thereby evaluating the level of receptor-bound BAFF,

wherein a greater level of receptor-bound BAFF, compared to a control, indicates that the subject is a candidate for a BAFF antagonist therapy.

The method of claim 7 , wherein the first anti-BAFF antibody is selected from the group consisting of: (a) antibody B4H7, (b) an antibody that competitively inhibits antibody B4H7from binding to BAFF, (c) an antibody that is competitively inhibited from binding to BAFF by antibody B4H7, and (d) an antibody that binds to the same epitope as antibody B4H7.

9. The method of claim 7 , wherein the BAFF antagonist therapy is selected from the group consisting of: an inhibitory anti-BAFF antibody, a soluble BAFF receptor or Fc fusion protein thereof, and an anti-BAFF receptor antibody.

The method of claim 7 , wherein the subject has an autoimmune disease selected from the group consisting of: autoimmune rheumatologic disorders, autoimmune gastrointestinal and liver disorders, vasculitis, autoimmune neurological disorders, autoimmune dermatologic disorders, autoimmune endocrine disorders, renal disorders, and hematologic disorders.

11. The method of claim 7 , wherein the biological sample is selected from the group consisting of: blood, tissue, plasma, synovial fluid, BAL and CSF.

12. The method of claim 7 , wherein the subject has a normal level of free BAFF compared to the control.

13. A method of identifying a subject who is a candidate for a BAFF antagonist therapy, the method comprising:

(a) contacting a first biological sample from the subject with a first anti-BAFF antibody, which binds BAFF independent of whether BAFF is bound to a receptor;

(b) contacting a second biological sample from the subject, of the same type as the first, with a second anti-BAFF antibody, which preferentially binds free BAFF over BAFF bound to receptor; and

(c) comparing the binding of the first antibody and the binding of the second antibody, thereby evaluating the level of receptor-bound BAFF, wherein a greater level of receptor-bound BAFF, compared to a control, indicates that the subject is a candidate for a BAFF antagonist therapy.

The method of claim 13 , wherein the first anti-BAFF antibody is selected from the group consisting of: (a) antibody B4H7, (b) an antibody that competitively inhibits antibody B4H7 from binding to BAFF, (c) an antibody that is competitively inhibited from binding to BAFF by antibody B4H7, and (d) an antibody that binds to the same epitope as antibody B4H7.

15. The method of claim 13 , wherein the BAFF antagonist therapy is selected from the group consisting of: an inhibitory anti-BAFF antibody, a soluble BAFF receptor or Fc fusion protein thereof, and an anti-BAFF receptor antibody.

The method of claim 13 , wherein the subject has an autoimmune disease selected from the group consisting of: autoimmune rheumatologic disorders, autoimmune gastrointestinal and liver disorders, vasculitis, autoimmune neurological disorders, autoimmune dermatologic disorders, autoimmune endocrine disorders, renal disorders, and hematologic disorders.

17. The method of claim 13 , wherein the biological sample type is selected from the group consisting of: blood, tissue, plasma. synovial fluid, BAL and CSF.

18. The method of claim 13 , wherein the subject has a normal level of free BAFF compared to the control.

19. The method of any one of claims 2 , 8 , and 14 , wherein the antibody B4H7 is the antibody expressed by the hybridoma deposited under ATCC No. PTA-7602.
