Abstract

The present invention provides methods of administering Factor IX; methods of administering chimeric and hybrid polypeptides comprising Factor IX; polynucleotides encoding such chimeric and hybrid polypeptides; cells comprising such polynucleotides; and methods of producing such chimeric and hybrid polypeptides using such cells.

Claims (36)

1. A method of controlling the incidence of a bleeding episode in a subject having hemophilia comprising administering to the subject multiple doses of at least about 50 IU/kg of a long-acting Factor IX (“FIX”) polypeptide at a dosing interval of about a week or longer between two doses.

2. The method of claim 1 , wherein the long-acting FIX polypeptide comprises an FIX portion and an Fc portion and exhibits one or more characteristics selected from the group consisting of:

a. an incremental recovery (K-Value) (activity; observed) greater than 0.75 IU/dL per IU/kg;

b. an incremental recovery (K value) (activity; observed) of about 0.62-1.17 IU/dL per IU/kg;

c. a clearance (CL) (activity) of about 1.84-4.58 mL/hour/kg;

d. a clearance (CL) (activity) of about 3.36±0.93 mL/hour/kg;

e. a mean residence time (MRT) (activity) of about 53.1-85.8 hours;

f. a mean residence time (MRT) (activity) of about 45, about 50, about 55, about 60, about 65, about 70, about 75, about 80, about 85, or about 90 hours;

g. a MRT (activity) of at least about 68.05±11.16 hours;

h. a t 1/2beta (activity) of about 40-67.4 hours;

i. a t 1/2beta (activity) of at least about 40, about 45, about 50, about 55, about 60, about 65, about 70, or about 75 hours;

j. a t 1/2beta (activity) of about 3 fold longer than the t 1/2beta (activity) of a polypeptide consisting of FIX;

k. a Vss (activity) of about 145-365 mL/kg;

l. a Vss (activity) of about 226±6736 mL/kg;

m. an AUC/dose of about 21.80-54.30 IU*h/dL per IU/kg; and

n. an AUC/dose of about 32.44±10.75 IU*h/dL per IU/kg.

3. The method of claim 1 , wherein each of said doses is selected from about 50 IU/kg to about 60 IU/kg, about 60 IU/kg to about 70 IU/kg, about 70 IU/kg to about 80 IU/kg, about 80 IU/kg to about 90 IU/kg, or about 90 IU/kg, to about 100 IU/kg.

4. The method of claim 1 , wherein each of said doses is selected from about 50 nag, about 55 IU/kg, about 60 IU/kg, about 65 IU/kg, about 70 IU/kg, about 75 IU/kg, about 80 IU/kg, about 85 IU/kg, about 90 III/kg, about 95 IU/kg, or about 100 IU/kg.

5. The method of claim 1 , wherein each of the multiple doses is about 50 IU/kg and the dosing interval is about a week.

6. The method of claim 1 , wherein each of the multiple doses is about 100 and the dosing interval is 10 days.

7. The method of claim 2 , wherein the FIX portion is at least 90% identical to amino acids 1 to 415 of SEQ ID NO:2.

8. The method of claim 2 , wherein the Fe portion is at least 90% identical to amino acids 1 to 227 of SEQ ID NO:4.

9. The method of claim 2 , wherein the long-acting FIX polypeptide is in the form of a hybrid comprising a second polypeptide, which comprises an additional Fe portion associated with the first Fe portion.

10. The method of claim 1 , wherein less than 25% of FIX in the long-acting FIX polypeptide in said doses are fully phosphorylated and less than 25% of FIX in the long-acting FIX polypeptide in said doses are fully sulfated.

11. The method of claim 1 , wherein said long-acting FIX polypeptide is administered as part of a pharmaceutical composition comprising at least one excipient.

12. The method of claim 1 , wherein said doses are administered intravenously or subcutaneously.

13. The method of claim 1 , wherein the dosing interval is about 10 days or longer.

14. The method of claim 1 , which is for prophylactic treatment of hemophilia B.

15. The method of claim 1 , wherein the dosing interval is about a week to about 10 days.

16. The method of claim 1 , which is for on-demand treatment.

17. The method of claim 16 , wherein each of the multiple doses is about 50 IU/kg to about 100 IU/kg.

18. The method of claim 1 , wherein the plasma level of the long-acting FIX polypeptide reaches an average trough of at least about 1 IU/dl.

A method of controlling a bleeding episode in a hemophilia patient comprising administering to the patient multiple doses of at least about 50 IU/kg of a long-acting FIX polypeptide at a dosing interval of about a week between two doses, wherein the long-acting FIX polypeptide comprises a FIX portion and an FcRn Binding Partner portion.

20. The method of claim 19 , wherein the FcRn Binding Partner portion comprises an Fc portion.

A method of controlling a bleeding episode in a hemophilia patient comprising administering to the patient multiple doses of at least about 100 IU/kg of a long-acting FIX polypeptide at a dosing interval of about 10 days between two doses, wherein the long-acting FIX polypeptide comprises a FIX portion and an FcRn Binding Partner portion.

22. The method of claim 21 , wherein the FcRn Binding Partner portion comprises an Fc portion.
