View Patent ↗The present invention relates to an improved process for the preparation of Letrozole (I) and its synthetic intermediate 4-[(1-(1,2,4-triazoly)methyl]benzonitrile (III). In particular, it relates to a process to prepare Letrozole and its intermediate (III) substantially free from regioisomeric impurities. The present invention further relates to acid addition salts of 4-[(1-(1,2,4-triazoly)methyl]benzonitrile (III) such as the oxalate salt, and also to Letrozole (I), the intermediate (III) and salts thereof preparable by the processes of the present invention.
1. An acid addition salt of 4-[1-(1,2,4-triazolyl)methyl]benzonitrile (III), wherein the acid is butyric, formic or oxalic acid.
2. A process for preparing a salt according to claim 1 , comprising mixing 4-[1-(1,2,4-triazolyl)methyl]benzonitrile (III) with the acid in a solvent system.
3. A process according to claim 2 , wherein,
the solvent system comprises one or more solvents selected from straight chain or branched aliphatic ketones and aliphatic C 1 to C 4 alcohols, or mixtures thereof.
4. An acid addition salt of 4-[1-(1,2,4-triazolyl)methyl]benzonitrile (III), wherein the acid is butyric, formic or oxalic acid with HPLC purity of equal to or more than 99.8%.
5. An acid addition salt of 4-[1-(1,2,4-triazolyl)methyl]benzonitrile (III), wherein the acid is butyric, formic or oxalic acid substantially free of the regioisomeric impurity 4-[1-(1,3,4-triazolyl)-methyl]benzonitrile (V) or salts thereof.
6. The acid addition salt of 4-[1-(1,2,4-triazolyl)methyl]benzonitrile (III) according to claim 5 , with HPLC purity of equal to or more than 99.8%.

The process of claim 2 , wherein the solvent system comprises one or more solvents selected from straight chain or branched aliphatic ketones and aliphatic C1 to C4 alcohols, or mixtures thereof, wherein the ketone is selected from acetone, methyl ethyl ketone and methyl isopropyl ketone, or mixtures thereof.

The process of claim 2 , wherein the solvent system comprises one or more solvents selected from straight chain or branched aliphatic ketones and aliphatic C1 to C4 alcohols, or mixtures thereof, wherein the alcohol is selected from methanol, ethanol, 1-propanol, 2-propanol, 1-butanol and tertiary butanol, or mixtures thereof.
9. The process of claim 2 , wherein the solvent system comprises acetone and methanol.
10. The process of claim 2 , wherein the solvent system comprises acetone and methanol, wherein the ratio of acetone:methanol in the solvent system is about 95:5, about 80:20, about 60:40, or about 50:50 by volume.
11. A method for preparing 4-[1-(1,2,4-triazolyl)methyl]benzonitrile (III), said method comprising purifying the acid addition salt of claim 1 and subsequently liberating the 4-[1-(1,2,4-triazolyl)methyl]benzonitrile (III) from the purified salt.
12. A process according to claim 11 , wherein the salt is purified by crystallisation from a solvent system.
13. The process according to claim 12 , wherein the solvent system comprises one or more solvents selected from straight chain or branched aliphatic ketones and aliphatic C1 to C4 alcohols, or mixtures thereof, wherein the ketone is selected from acetone, methyl ethyl ketone and methyl isopropyl ketone, or mixtures thereof.

The process according to claim 12 , wherein the solvent system comprises one or more solvents selected from straight chain or branched aliphatic ketones and aliphatic C1 to C4 alcohols, or mixtures thereof, wherein the alcohol is selected from methanol, ethanol, 1-propanol, 2-propanol, 1-butanol and tertiary butanol, or mixtures thereof.
15. The process according to claim 12 , wherein the solvent system comprises acetone and methanol.
16. The process according to claim 12 , wherein the solvent system comprises acetone and methanol, wherein the ratio of acetone:methanol in the solvent system is about 95:5, about 80:20, about 60:40, or about 50:50 by volume.
