Abstract

Some embodiments of the present inventive concept provide optical coherence tomography (OCT) systems for integration with a microscope. The OCT system includes a sample arm coupled to the imaging path of a microscope. The sample arm includes an input beam zoom assembly including at least two movable lenses configured to provide shape control for an OCT signal beam; a scan assembly including at least one scanning mirror and configured for telecentric scanning of the OCT signal beam; and a beam expander configured to set the OCT signal beam diameter incident on the microscope objective. The shape control includes separable controls for numerical aperture and focal position of the imaged OCT beam.

Claims (27)

1. A method of optical coherence tomography (OCT) imaging in conjunction with a surgical procedure, the method comprising:

acquiring two or more images with a Fourier domain optical coherence tomography (FDOCT) imaging system, wherein the FDOCT imaging system comprises a broadband optical source and a detection module that samples a spectral interferogram at a spectral sampling interval that is greater than zero and less than a total bandwidth of the optical source,

visualizing a first region of interest, wherein the detection module samples the spectral interferogram at a spectral sampling interval v 1 greater than zero and less than the total bandwidth of the optical source and wherein a maximum observable image depth z 1 is inversely proportional to the spectral sampling interval v 1 ; and

visualizing a second region of interest with the FDOCT imaging system, wherein the detection module samples the spectral m at a spectral sampling interval v 2 , wherein v 2 is greater than or equal to 2v 1 , and wherein a maximum observable depth z 2 is inversely proportional to the spectral sampling interval v 2 and less than z 1 .

2. The method of claim 1 :

wherein the FDOCT imaging system comprises at least two movable lenses configured to independently adjust a numerical aperture and a focal position of an OCT imaging beam;

wherein visualizing the first region of interest is performed with a scanning system having a first numerical aperture; and

wherein visualizing the second region of interest is performed with the scanning system having a second numerical aperture, different from the first numerical aperture, the second numerical aperture being greater than the first numerical aperture.

3. A method of optical coherence tomography (OCT) imaging in conjunction with a surgical procedure, the method comprising:

controlling an OCT imaging system, wherein the OCT imaging system comprises at least two movable lenses configured to independently adjust a numerical aperture and a focal position of an OCT imaging beam;

establishing a first setting of a region of interest for OCT imaging;

establishing a first numerical aperture and a first focal position for the OCT imaging;

acquiring at least a first OCT image data set;

computing at least a first clinical parameter from the at least first OCT image data set;

performing a surgical procedure;

acquiring at least a second OCT image data set; and

computing at least a second clinical parameter from the at least a second OCT image data set.

4. A method of optical coherence tomography (OCT) imaging in conjunction with a surgical procedure, the method comprising:

controlling an OCT imaging system, wherein the OCT imaging system comprises at least two movable lenses configured to independently adjust a numerical aperture and a focal position of an OCT imaging beam;

setting a first region of interest within a surgical sample for OCT imaging;

acquiring at least a first OCT image of the first region of interest;

performing a surgical procedure involving the first region of interest;

setting a second region of interest within the surgical sample for OCT imaging, the second region of interest being at least partially different from the first region of interest; and

acquiring at least a second OCT image of the second region of interest.

5. The method of claim 4 :

wherein a first reference arm position, a first numerical aperture, and a first focal position of the OCT imaging system are set for acquiring the OCT image in the first region of interest; and

wherein at least one of a reference arm position, a numerical aperture and a focal position of the OCT imaging system are changed for acquiring the OCT image in the second region of interest.
