Chimeric adenoviruses for use in cancer treatment
View Patent ↗The present invention relates to oncolytic adenoviruses having therapeutic applications. Recombinant chimeric adenoviruses, and methods to produce them are provided. The chimeric adenoviruses of the invention comprise nucleic acid sequences derived from adenoviral serotypes classified within the subgroups B through F and demonstrate an enhanced therapeutic index.
1. A replication competent oncolytic type B adenovirus comprising the genome of Ad11 characterised in that said genome defines a deletion in the base pairs selected from the group consisting of:
(1) an E3 region deletion comprising base pairs 27979 to 30423, numbered according to SEQ ID NO:2;
(2) an E4orf4 gene deletion comprising base pairs 33164 to 33189, numbered according to SEQ ID NO:2, and
(3) a combination of (1) and (2),
wherein the adenovirus has an enhanced therapeutic index.
2. A replication competent oncolytic type B adenovirus according to claim 1 , wherein the adenovirus comprises the deletion in the E3 region.
3. A replication competent oncolytic type B adenovirus according to claim 2 , wherein the adenovirus comprises the deletion in the E4orf4 gene.
4. A replication competent oncolytic type B adenovirus according to claim 1 , wherein the adenovirus comprises the deletion in the E4orf4 gene.
5. A pharmaceutical composition, comprising a replication competent oncolytic type B adenovirus as defined in claim 1 .
