Abstract

A recombinant yeast that makes an isoprenoid compound is provided. The yeast comprises an endogenous mevalonate pathway comprising (i) an enzyme that converts acetyl-CoA to acetoacetyl-CoA, (ii) an enzyme that converts acetoacetyl-CoA to hydroxymethylglutaryl-CoA, (iii) an enzyme that converts hydroxymethylglutaryl-CoA to mevalonic acid, (iv) an enzyme that converts mevalonic acid to phosphomevalonic acid, (v) an enzyme that converts phosphomevalonic acid to phosphomevalonate, and (vi) an enzyme that converts phosphomevalonate to isopentenyl pyrophosphate. The yeast further comprises heterologous nucleic acid sequences encoding an enzyme that converts hydroxymethylglutaryl-CoA to mevalonic acid, an enzyme that converts mevalonic acid to phosphomevalonic acid, an enzyme that converts phosphomevalonic acid to phosphomevalonate, and an enzyme that converts phosphomevalonate to isopentenyl pyrophosphate. Carbon flow through the mevalonate pathway is increased in the recombinant yeast compared to a yeast that does not comprise the heterologous nucleic acid molecule encoding an enzyme that converts hydroxymethylglutaryl-CoA to mevalonic acid.

Claims (29)

1. A recombinant yeast that is capable of making an isoprenoid compound, the recombinant yeast comprising:

an endogenous mevalonate pathway comprising (i) an enzyme that converts acetyl-CoA to acetoacetyl-CoA, (ii) an enzyme that converts acetoacetyl-CoA to hydroxymethylglutaryl-CoA, (iii) an enzyme that converts hydroxymethylglutaryl-CoA to mevalonic acid, (iv) an enzyme that converts mevalonic acid to phosphomevalonic acid, (v) an enzyme that converts phosphomevalonic acid to phosphomevalonate, and (vi) an enzyme that converts phosphomevalonate to isopentenyl pyrophosphate; and

at least one heterologous nucleic acid sequence encoding an enzyme that converts hydroxymethylglutaryl-CoA to mevalonic acid;

at least one heterologous nucleic acid sequence encoding an enzyme that converts mevalonic acid to phosphomevalonic acid;

at least one heterologous nucleic acid sequence encoding an enzyme that converts phosphomevalonic acid to phosphomevalonate; and

at least one heterologous nucleic acid sequence encoding an enzyme that converts phosphomevalonate to isopentenyl pyrophosphate;

wherein carbon flow through the mevalonate pathway from mevalonate to isopentenyl pyrophosphate is increased in the recombinant yeast compared to a yeast that does not comprise the at least one heterologous nucleic acid sequence encoding the enzyme that converts mevalonic acid to phosphomevalonic acid, the at least one heterologous nucleic acid sequence encoding the enzyme that converts phosphomevalonic acid to phosphomevalonate, and the at least one heterologous nucleic acid sequence encoding the enzyme that converts phosphomevalonate to isopentenyl pyrophosphate.

2. The recombinant yeast of claim 1 , wherein the enzyme that converts acetyl-CoA to acetoacetyl-CoA is a thiolase.

3. The recombinant yeast of claim 1 , wherein the enzyme that converts acetoacetyl-CoA to hydroxymethylglutaryl-CoA is an HMG Co A synthase (HMGS).

4. The recombinant yeast of claim 1 , wherein the enzyme that converts hydroxymethylglutaryl-CoA to mevalonic acid is an HMG Co A reductase (HMGR).

5. The recombinant yeast of claim 1 , wherein the enzyme that converts mevalonic acid to phosphomevalonic acid is a mevalonate kinase (MK).

6. The recombinant yeast of claim 1 , wherein the enzyme that converts phosphomevalonic acid to phosphomevalonate is a phosphomevalonate kinase (PMK).

7. The recombinant yeast of claim 1 , wherein the enzyme that converts phosphomevalonate to isopentenyl pyrophosphate is a phosphomevalonate decarboxylase (PMD).

The recombinant yeast of claim 1 , wherein at least one of (i) the enzyme that converts hydroxymethylglutaryl-CoA to mevalonic acid, (ii) the enzyme that converts mevalonic acid to phosphomevalonic acid, (iii) the enzyme that converts phosphomevalonic acid to phosphomevalonate, and (iv) the enzyme that converts phosphomevalonate to isopentenyl pyrophosphate is under inducible control.

The recombinant yeast of claim 1 , wherein (i) the enzyme that converts hydroxymethylglutaryl-CoA to mevalonic acid, (ii) the enzyme that converts mevalonic acid to phosphomevalonic acid, (iii) the enzyme that converts phosphomevalonic acid to phosphomevalonate, and (iv) the enzyme that converts phosphomevalonate to isopentenyl pyrophosphate is under inducible control.

The recombinant yeast of claim 1 , wherein at least one of the heterologous nucleic acid sequences selected from the group consisting of the at least one heterologous nucleic acid sequence encoding the enzyme that converts hydroxymethylglutaryl-CoA to mevalonic acid; the at least one heterologous nucleic acid sequence encoding the enzyme that converts mevalonic acid to phosphomevalonic acid; the at least one heterologous nucleic acid sequence encoding an enzyme that converts phosphomevalonic acid to phosphomevalonate; and the at least one heterologous nucleic acid sequence encoding the enzyme that converts phosphomevalonate to isopentenyl pyrophosphate; is integrated into the genome.

The recombinant yeast of claim 1 , wherein each of the heterologous nucleic acid sequences in the group consisting of the at least one heterologous nucleic acid sequence encoding the enzyme that converts hydroxymethylglutaryl-CoA to mevalonic acid; the at least one heterologous nucleic acid sequence encoding the enzyme that converts mevalonic acid to phosphomevalonic acid; the at least one heterologous nucleic acid sequence encoding the enzyme that converts phosphomevalonic acid to phosphomevalonate; and the at least one heterologous nucleic acid sequence encoding the enzyme that converts phosphomevalonate to isopentenyl pyrophosphate; are integrated into the genome.

12. The recombinant yeast of claim 1 , wherein the yeast comprises more than one heterologous nucleic acid sequence encoding an enzyme that converts hydroxymethylglutaryl-CoA to mevalonic acid.

13. The recombinant yeast of claim 1 , wherein the yeast further comprises at least one heterologous nucleic acid sequence encoding an enzyme that converts acetoacetyl-CoA to hydroxymethylglutaryl-CoA.

14. The recombinant yeast of claim 13 , wherein the enzyme that converts acetoacetyl-CoA to hydroxymethylglutaryl-CoA is an HMGS.

15. The recombinant yeast of claim 1 , wherein the yeast is Saccharomyces cerevisiae.

16. The recombinant yeast of claim 1 , wherein the enzyme that converts hydroxymethylglutaryl-CoA to mevalonic acid is a bacterial enzyme.

17. The recombinant yeast of claim 1 , wherein the enzyme that converts hydroxymethylglutaryl-CoA to mevalonic acid is a yeast enzyme.

The recombinant yeast of claim 1 , wherein the enzyme that converts hydroxymethylglutaryl-CoA to mevalonic acid, the enzyme that converts mevalonic acid to phosphomevalonic acid, the enzyme that converts phosphomevalonic acid to phosphomevalonate, and the enzyme that converts phosphomevalonate to isopentenyl pyrophosphate are each eukaryotic.

The recombinant yeast of claim 1 , wherein the enzyme that converts hydroxymethylglutaryl-CoA to mevalonic acid, the enzyme that converts mevalonic acid to phosphomevalonic acid, the enzyme that converts phosphomevalonic acid to phosphomevalonate, and the enzyme that converts phosphomevalonate to isopentenyl pyrophosphate are each yeast enzymes.

The recombinant yeast of claim 1 , wherein each enzyme in the group consisting of: the enzyme that converts hydroxymethylglutaryl-CoA to mevalonic acid, the enzyme that converts mevalonic acid to phosphomevalonic acid, the enzyme that converts phosphomevalonic acid to phosphomevalonate, and the enzyme that converts phosphomevalonate to isopentenyl pyrophosphate; is encoded on a different heterologous nucleic acid molecule.

The recombinant yeast of claim 1 , wherein at least two enzymes selected from the group consisting of: the enzyme that converts hydroxymethylglutaryl-CoA to mevalonic acid, the enzyme that converts mevalonic acid to phosphomevalonic acid, the enzyme that converts phosphomevalonic acid to phosphomevalonate, and the enzyme that converts phosphomevalonate to isopentenyl pyrophosphate; are encoded on the same heterologous nucleic acid molecule.

The recombinant yeast of claim 1 , wherein at least three enzymes selected from the group consisting of: the enzyme that converts hydroxymethylglutaryl-CoA to mevalonic acid, the enzyme that converts mevalonic acid to phosphomevalonic acid, the enzyme that converts phosphomevalonic acid to phosphomevalonate, and the enzyme that converts phosphomevalonate to isopentenyl pyrophosphate; are encoded on the same heterologous nucleic acid molecule.

The recombinant yeast of claim 1 , wherein the enzyme that converts hydroxymethylglutaryl-CoA to mevalonic acid, the enzyme that converts mevalonic acid to phosphomevalonic acid, the enzyme that converts phosphomevalonic acid to phosphomevalonate, and the enzyme that converts phosphomevalonate to isopentenyl pyrophosphate are encoded on the same heterologous nucleic acid molecule.
