Abstract

The invention provides isolated polynucleotide molecules that comprise a DNA sequence encoding an infectious RNA sequence encoding a genetically-modified North American PRRS virus, methods to make it and related polypeptides, polynucleotides, and various components. Vaccines comprising the genetically modified virus and polynucleotides and a diagnostic kit to distinguish between naturally infected and vaccinated animals are also provided.

Claims (7)

1. An isolated DNA polynucleotide molecule comprising a DNA sequence encoding an infectious RNA molecule encoding a North American PRRS virus, wherein said DNA sequence is selected from the group consisting of:

(a) SEQ ID NO: 1, SEQ ID NO: 2, SEQ ID NO: 3, SEQ ID NO: 4, and SEQ ID NO: 6, or

(b) the complement of any sequence in (a).

2. The isolated DNA polynucleotide molecule of claim 1 in the form of a plasmid.

3. A host cell transfected with the plasmid of claim 2 .

4. A vaccine for protecting a porcine animal against infection by a PRRS which vaccine comprises: (a) a North American PRRS virus encoded by an infectious RNA molecule according to claim 1 , (b) an isolated DNA polynucleotide molecule according to claim 1 in the form of a plasmid, or (c) a viral vector containing encoding sequence for the North American PRRS virus of claim 1 ; and a carrier suitable for veterinary use, wherein the vaccine is able to elicit an effective immunoprotective response against infection by PRRS virus, in an amount effective to produce immunoprotection against disease.

5. The vaccine of claim 4 , wherein the vaccine has a decreased interferon-α inhibitory effect as compared to wild-type P129 PRRS virus.
