Abstract

Newly identified mammalian taste-cell-specific G protein-coupled receptors, and the genes and cDNA encoding said receptors are described. Specifically, T1R G protein-coupled receptors active in taste signaling, and the genes and cDNA encoding the same, are described, along with methods for isolating such genes and for isolating and expressing such receptors. Methods for representing taste perception of a particular taste stimulus in a mammal are also described, as are methods for generating novel molecules or combinations of molecules that elicit a predetermined taste perception in a mammal, and methods for simulating one or more tastes. Further, methods for stimulating or blocking taste perception in a mammal are also disclosed.

Claims (11)

1. A method of identifying a cell comprising a T1R polypeptide, said method comprising:

(i) providing at least one cell that potentially comprises a polypeptide which is at least 90% identical to the polypeptide in SEQ ID NO:4;

(ii) contacting said at least one cell with a probe that specifically binds to the T1R polypeptide of SEQ ID NO:4; and

(iii) identifying said at least one cell as a cell comprising said T1R polypeptide if said at least one cell specifically binds to said probe.

2. The method of claim 1 wherein the polypeptide of step (i) is at least 95% identical to SEQ ID NO: 4.

3. The method of claim 1 wherein the polypeptide of step (i) is at least 98% identical to SEQ ID NO: 4.

4. The method of claim 1 wherein the polypeptide of step (i) is at least 99% identical to SEQ ID NO: 4.

5. The method of claim 1 , which further includes contacting said cell comprising said T1R polypeptide with at least one putative T1R modulatory compound and detecting whether said compound specifically binds to, or modulates the activity of the T1R polypeptide of step (i), thereby determining if the compound putatively is a T1R taste modulatory compound.

6. The method of claim 1 , wherein said cell comprising said T1R polypeptide is a mammalian cell.

7. The method of claim 6 , wherein said mammalian cell is a human cell.

8. The method of claim 1 wherein said polypeptide of step (i) was expressed from a gene endogenous to said cell.
