3,5-disubstituted-3H-imidazo[4,5-B]pyridine and 3,5-disubstituted-3H-[1,2,3]triazolo[4,5-B] pyridine compounds as modulators of protein kinases
The present invention provides, inter alia, compounds of formula I as protein kinase modulators, methods of preparing them, pharmaceutical compositions containing them and methods of treatment, prevention and/or amelioration of kinase mediated diseases or disorders with them.
1. A compound of the formula
or a tautomer, stereoisomer, enantiomer, diastereomer, or salt thereof, wherein
X is CR 1 or N;
B is selected from Cy 1 , —NR c R d or —U═V—W—R 5 ;
Cy 1 is selected from substituted or unsubstituted cycloalkyl, substituted or unsubstituted heterocyclic group, substituted or unsubstituted aryl, and substituted or unsubstituted heteroaryl;
wherein when X is N, Cy 1 is other than phenyl substituted with one to five substituents selected from halogen, substituted or unsubstituted alkyl, and —CONR x R y ;
R 1 is selected from hydrogen, nitro, hydroxy, cyano, halogen, —OR a , —S(═O) q —R a , —NR a R b , —C(═Y)—R a , —CR a R b —C(═Y)—R a , —CR a R b —Y—CR a R b , —C(═Y)—NR a R b , —NR a —C(═Y)—NR a R b , —S(═O) q —NR a R b , —NR a —S(═O) q —NR a R b , —NR a —NR a R b , substituted or unsubstituted C 1-6 alkyl, substituted or unsubstituted C 2-6 alkenyl, substituted or unsubstituted C 2-6 alkynyl, substituted or unsubstituted C 3-6 cycloalkyl, substituted or unsubstituted C 3-6 cycloalkylalkyl, substituted or unsubstituted C 3-6 cycloalkenyl;
R 2 is hydrogen, nitro, hydroxy, cyano, halogen, —OR a , —S(═O) q —R a , —NR a R b , —C(═O)—R a , or —C(═O)—R a , wherein R a and R b in the R 2 group are independently hydrogen, hydroxy, or substituted or unsubstituted C 1-6 alkyl;
each occurrence of R a and R b may be same or different and are independently selected from hydrogen, halogen, or substituted or unsubstituted (C 1-6 ) alkyl, or both together with the carbon atom to which they are attached form a saturated 3 to 6 member cyclic ring which may optionally include heteroatoms which may be same or different and are selected from O, NR e or S;
R c and R d together with the nitrogen to which they are attached form a substituted or unsubstituted, saturated or unsaturated 3-10 member ring, which may optionally include one or more heteroatoms which may be same or different and are selected from O, NR e or S;

Z is selected from CR c , S, O, NR c , R c C═CR c , —N═CR c —, —R c C═N—;
Z 1 is selected from N, NR c or CR c ;
each occurrence of Y is independently selected from O, S, and NR a ;
each occurrence of n independently represents 0, 1, 2, 3 or 4;
each occurrence of q independently represents 0, 1 or 2 and
R c is absent or selected from hydrogen, hydroxy or halogen.
2. The compound of claim 1 , wherein X is N.
3. The compound of claim 1 , wherein each occurrence of R 2 is H.
4. The compound of claim 1 , wherein each of R a and R b are same or different and selected from hydrogen, methyl or fluoro.
5. The compound of claim 4 , wherein each of R a and R b is selected from hydrogen or methyl.
6. The compound of claim 1 , wherein Z is CR c , N, S, O, HC═CH—, —N═CH—.
7. The compound of claim 1 , wherein Z 1 is CH or N.
8. The compound of claim 6 , wherein Z is —HC═CH—, —S— or —O— and Z 1 is CH.
9. The compound of claim 8 , wherein Z is —HC═CH— and Z 1 is CH.
10. The compound of claim 6 , wherein Z is —S— and Z 1 is CH.
11. The compound of claim 1 , wherein each occurrence of R c is hydrogen or fluoro.
12. The compound of claim 1 , wherein B is Cy 1 .
13. The compound of claim 12 , wherein Cy 1 is selected from
14. The compound of claim 1 , wherein Cy 1 is substituted or unsubstituted monocyclic aryl or substituted or unsubstituted monocyclic heteroaryl.
15. The compound of claim 14 , wherein Cy 1 is substituted monocyclic aryl.

24. A pharmaceutical composition, comprising a compound of claim 1 and a pharmaceutically acceptable carrier.
25. The pharmaceutical composition of claim 24 , further comprising one or more additional therapeutic agents and mixtures thereof.
26. The pharmaceutical composition of claim 24 , wherein the one or more additional therapeutic agent is an anti-cancer agent, anti-inflammatory agent, immunosuppressive agent, steroid, non-steroidal anti-inflammatory agent, antihistamine, analgesic, or a mixture thereof.
27. A method of inhibiting the HGF/c-Met kinase signaling pathway in a cell comprising contacting said cell with a compound of claim 1 .
28. A method of inhibiting the proliferative activity of a cell comprising contacting said cell with a compound of claim 1 .
