Split dose administration
View Patent ↗The present disclosure provides, inter alia, formulation compositions comprising modified nucleic acid molecules which may encode a protein, a protein precursor, or a partially or fully processed form of the protein or a protein precursor. The formulation composition may further include a modified nucleic acid molecule and a delivery agent. The present invention further provides nucleic acids useful for encoding polypeptides capable of modulating a cell's function and/or activity.
1. A method for producing an increased level of erythropoietin (EPO) in a mammalian subject, comprising
(a) administering to said subject a total daily dose of an mRNA encoding erythropoietin (EPO) in two or more equal or unequal split doses within a 24 hour period, wherein said mRNA comprises SEQ ID NO: 9; and
(b) determining that the level of erythropoietin (EPO) in a bodily fluid of said subject is greater than the level produced by administering the same total daily dose as a single administration.
2. The method of claim 1 , wherein the mammalian subject is a human patient in need of an increased level of erythropoietin (EPO).

The method of claim 1 , wherein the bodily fluid is serum and wherein the method further comprises the step of calculating the ratio of the amount of polypeptide detected in the serum (in pmol) divided by the total dose of mRNA administered (in ug), wherein a ratio of greater than 1 indicates an increased level of erythropoietin (EPO).

The method of claim 3 , further comprising the step of calculating a ratio of the amount of polypeptide detected in the serum (in pmol) divided by the total dose of mRNA administered as a split dose (in ug) divided by the ratio of the amount of polypeptide detected in the serum (in pmol) divided by the total dose of mRNA administered as a single dose (in ug) and wherein where the total dose administered in the split dose administration is the same as the total dose in the single dose administration, the calculated ratio is greater than 4.
5. The method of claim 1 , wherein the administration is transdermal.
6. The method of claim 5 , wherein the distance between the site of injection of said first dose and the site of injection of said second dose is from about 1 mm to about 10 cm.
7. The method of claim 1 , wherein the mRNA comprises at least one nucleoside modification.
8. The method of claim 1 , wherein the amount of mRNA administered in said first and said second dose is substantially equal.
9. The method of claim 1 , wherein said two or more equal or unequal split doses comprise a first dose and a second dose and wherein said first dose and said second dose are administered at substantially the same time.
