Abstract

The present invention provides novel human anti-influenza antibodies and related compositions and methods. These antibodies are used in the diagnosis and treatment of influenza infection.

A recombinant anti-matrix 2 ectodomain (M2e) antibody comprising a variable heavy chain region encoded by a nucleic acid sequence comprising the nucleic acid sequence of SEQ ID NO: 43 and a variable light chain region encoded by a nucleic acid sequence comprising the nucleic acid sequence of SEQ ID NO: 45.

43 . The antibody of claim 42 , wherein the nucleic acid sequence comprising the nucleic acid sequence of SEQ ID NO: 43 is a cDNA and the nucleic acid sequence comprising the nucleic acid sequence of SEQ ID NO: 45 is a cDNA.

44 . The antibody of claim 42 , wherein the nucleic acid sequence of SEQ ID NO: 43 is a cDNA and the nucleic acid sequence of SEQ ID NO: 45 is a cDNA.

45 . A vector comprising a nucleic acid sequence comprising the nucleic acid sequence of SEQ ID NO: 43 and a nucleic acid sequence comprising the nucleic acid sequence of SEQ ID NO: 45.

46 . The vector of claim 45 , wherein the nucleic acid sequence comprising the nucleic acid sequence of SEQ ID NO: 43 is a cDNA or the nucleic acid sequence comprising the nucleic acid sequence of SEQ ID NO: 45 is a cDNA.

47 . The vector of claim 45 , wherein the vector is vector suitable for propagation or replication of the nucleic acid.

48 . The vector of claim 45 , wherein the vector is vector suitable for expressing the nucleic acid.

49 . The vector of claim 47 or 48 , wherein the vector comprises a virus, a plasmid, or a cosmid.

50 . The vector of claim 49 , wherein the virus is a recombinant virus.

51 . The vector of claim 50 , wherein the virus comprises a promoter obtained from a mammalian genome.

52 . The vector of claim 51 , wherein the mammalian genome is a human genome.

53 . The vector of claim 50 , wherein the virus comprises a promoter obtained from a genome of a virus or a retrovirus.

54 . The vector of claim 53 , wherein the virus or retrovirus is a polyoma virus, fowlpox virus, adenovirus, bovine papilloma virus, avian sarcoma virus, cytomegalovirus (CMV), hepatitis-B virus, or a Simian Virus 40 (SV40).

55 . A recombinant anti-matrix 2 ectodomain (M2e) antibody comprising a nucleic acid sequence encoding a variable heavy chain region comprising the amino acid sequence of SEQ ID NO: 44 and a nucleic acid sequence encoding a variable light chain region comprising the amino acid sequence of SEQ ID NO: 46.

56 . The antibody of claim 55 , wherein the nucleic acid sequence encoding a variable heavy chain region comprising the amino acid sequence of SEQ ID NO: 44 is a cDNA and the nucleic acid sequence encoding a variable light chain region comprising the amino acid sequence of SEQ ID NO: 46 is a cDNA.

57 . A vector comprising a nucleic acid sequence encoding a variable heavy chain region comprising the amino acid sequence of SEQ ID NO: 44 and a nucleic acid sequence encoding a variable light chain region comprising the amino acid sequence of SEQ ID NO: 46.

58 . The vector of claim 57 , wherein the nucleic acid sequence encoding a variable heavy chain region comprising the amino acid sequence of SEQ ID NO: 44 is a cDNA or the nucleic acid sequence encoding a variable light chain region comprising the amino acid sequence of SEQ ID NO: 46 is a cDNA.

59 . The vector of claim 57 , wherein the vector is vector suitable for propagation or replication of the nucleic acid.

60 . The vector of claim 57 , wherein the vector is vector suitable for expressing the nucleic acid.

61 . The vector of claim 59 or 60 , wherein the vector comprises a virus, a plasmid, or a cosmid.

62 . The vector of claim 61 , wherein the virus is a recombinant virus.

63 . The vector of claim 62 , wherein the virus comprises a promoter obtained from a mammalian genome.

64 . The vector of claim 63 , wherein the mammalian genome is a human genome.

65 . The vector of claim 62 , wherein the virus comprises a promoter obtained from a genome of a virus or a retrovirus.

66 . The vector of claim 65 , wherein the virus or retrovirus is a polyoma virus, fowlpox virus, adenovirus, bovine papilloma virus, avian sarcoma virus, cytomegalovirus (CMV), hepatitis-B virus, or a Simian Virus 40 (SV40).

A recombinant anti-matrix 2 ectodomain (M2e) antibody comprising a variable heavy chain region encoded by a nucleic acid sequence comprising the nucleic acid sequence of SEQ ID NO: 49 and a variable light chain region encoded by a nucleic acid sequence comprising the nucleic acid sequence of SEQ ID NO: 51.

68 . The antibody of claim 67 , wherein the nucleic acid sequence comprising the nucleic acid sequence of SEQ ID NO: 49 is a cDNA and the nucleic acid sequence comprising the nucleic acid sequence of SEQ ID NO: 51 is a cDNA.

69 . The antibody of claim 67 , wherein the nucleic acid sequence of SEQ ID NO: 49 is a cDNA and the nucleic acid sequence of SEQ ID NO: 51 is a cDNA.

70 . A vector comprising a nucleic acid sequence comprising the nucleic acid sequence of SEQ ID NO: 49 and a nucleic acid sequence comprising the nucleic acid sequence of SEQ ID NO: 51.

71 . The vector of claim 70 , wherein the nucleic acid sequence comprising the nucleic acid sequence of SEQ ID NO: 49 is a cDNA or the nucleic acid sequence comprising the nucleic acid sequence of SEQ ID NO: 51 is a cDNA.

72 . The vector of claim 70 , wherein the vector is vector suitable for propagation or replication of the nucleic acid.

73 . The vector of claim 70 , wherein the vector is vector suitable for expressing the nucleic acid.

74 . The vector of claim 72 or 73 , wherein the vector comprises a virus, a plasmid, or a cosmid.

81 . The antibody of claim 80 , wherein the nucleic acid sequence encoding a variable heavy chain region comprising the amino acid sequence of SEQ ID NO: 50 is a cDNA and the nucleic acid sequence encoding a variable light chain region comprising the amino acid sequence of SEQ ID NO: 52 is a cDNA.

82 . A vector comprising a nucleic acid sequence encoding a variable heavy chain region comprising the amino acid sequence of SEQ ID NO: 50 and a nucleic acid sequence encoding a variable light chain region comprising the amino acid sequence of SEQ ID NO: 52.

83 . The vector of claim 82 , wherein the nucleic acid sequence encoding a variable heavy chain region comprising the amino acid sequence of SEQ ID NO: 50 is a cDNA or the nucleic acid sequence encoding a variable light chain region comprising the amino acid sequence of SEQ ID NO: 52 is a cDNA.

84 . The vector of claim 82 , wherein the vector is vector suitable for propagation or replication of the nucleic acid.

85 . The vector of claim 82 , wherein the vector is vector suitable for expressing the nucleic acid.

86 . The vector of claim 84 or 85 , wherein the vector comprises a virus, a plasmid, or a cosmid.

87 . The vector of claim 86 , wherein the virus is a recombinant virus.

88 . The vector of claim 87 , wherein the virus comprises a promoter obtained from a mammalian genome.

89 . The vector of claim 88 , wherein the mammalian genome is a human genome.

90 . The vector of claim 87 , wherein the virus comprises a promoter obtained from a genome of a virus or a retrovirus.
