Abstract

A eukaryotic expression vector capable of displaying a multi-chain polypeptide on the surface of a host cell is provided, such that the biological activity of the multi-chain polypeptide is exhibited at the surface of the host cell. Such a vector allows for the display of complex biologically active polypeptides, e.g., biologically active multi-chain polypeptides such as immunoglobulin Fab fragments. The present invention describes and enables the successful display of a multi-chain polypeptide on the surface of a eukaryotic host cell. Preferred vectors are described for expressing the chains of a multi-chain polypeptide in a host cell separately and independently (e.g., under separate vector control elements, and/or on separate expression vectors, thus forming a matched vector set). The use of such matched vector sets provides flexibility and versatility in the generation of eukaryotic display libraries, for example the ability to generate and to display multi-chain polypeptides by combining and recombining vectors that express variegations of the individual chains of a multi-chain polypeptide. Entire repertoires of novel chain combinations can be devised using such vector sets.

Claims (30)

1. A multi-chain protein display library, comprising a plurality of multi-chain proteins each comprising at least two polypeptide chains, wherein the multi-chain proteins are displayed on a surface of yeast host cells, and wherein the multi-chain proteins are multi-chain T cell receptors, multi-chain MHC class I molecules, multi-chain MHC class II molecules, multi-chain immunoglobulin molecules, or multi-chain biologically active fragments thereof, wherein the multi-chain immunoglobulin molecules each comprise a heavy chain variable region (V H ) and a light chain variable region (V L ), which are on separate chains.

2. The multi-chain protein display library of claim 1 , wherein the multi-chain proteins are linked to a surface anchor protein of the yeast host cells.

3. The multi-chain protein library of claim 2 , wherein the multi-chain proteins are linked to the surface anchor protein indirectly via an intermediate associate.

4. The multi-chain protein library of claim 3 , wherein the surface anchor protein is an Aga1p-Aga2p pair.

5. The multi-chain protein library of claim 1 , wherein the multi-chain proteins are immunoglobulins or functional fragments thereof.

6. The multi-chain protein library of claim 1 , wherein the library comprises at least 10 4 multi-chain proteins.

7. The multi-chain protein library of claim 6 , wherein the library comprises at least 10 7 multi-chain proteins.

8. The multi-chain protein library of claim 7 , wherein the library comprises at least 10 8 multi-chain proteins.

9. The multi-chain protein library of claim 1 , wherein the yeast host cells are diploid cells.

10. A method for preparing the multi-chain protein library of claim 1 , comprising:

inserting nucleotide sequences encoding the polypeptide chains of the multi-chain proteins into yeast expression vectors; and

introducing the yeast expression vectors to yeast cells to produce the multi-chain protein library, in which multi-chain proteins are displayed on surfaces of the resultant yeast host cells.

11. The method of claim 10 , wherein the nucleotide sequences encoding the polypeptide chains of the multi-chain proteins are inserted into the yeast expression vectors by homologous recombination.

12. The method of claim 11 , wherein the yeast host cells are diploid yeast cells.

13. The method of claim 12 , wherein the yeast expression vectors are introduced into haploid yeast cells of opposite mating types, and the haploid yeast cells of opposite mating types are fused to produce the diploid yeast cells.

14. The method of claim 10 , wherein the multi-chain proteins are immunoglobulins or functional fragments thereof.

15. The method of claim 10 , wherein the multi-chain proteins are linked to a surface anchor protein of the yeast host cells.

16. The method of claim 15 , wherein the multi-chain proteins are linked indirectly to the surface anchor protein via an intermediate association.

17. The method of claim 16 , wherein the surface anchor protein is an Aga1p-Aga2p pair.

18. A method for screening a multi-chain protein display library, the method comprising:

providing the multi-chain protein display library of claim 1 , in which multi-chain proteins are displayed on surfaces of yeast host cells,

interacting a target protein with the library, and

selecting the multi-chain proteins that bind to the target protein from the library.

19. The method of claim 18 , wherein the multi-chain proteins in the library are immunoglobulins or functional fragments thereof.

20. The method of claim 18 , wherein the multi-chain proteins in the library are linked to a surface anchor protein of the yeast host cells.

21. The method of claim 20 , wherein the multi-chain proteins in the library are linked indirectly to the surface anchor protein via an intermediate association.

22. The method of claim 21 , wherein the surface anchor protein is an Aga1p-Aga2p pair.

23. The multi-chain protein library of claim 5 , wherein the multi-chain proteins are Fab fragments.

24. The method of claim 14 , wherein the multi-chain proteins are Fab fragments.

25. The method of claim 19 , wherein the multi-chain proteins are Fab fragments.
