Abstract

During an assay, optical measurements are made at a sensing surface ( 112 ), and at least one “homogeneity-image” of the sensing surface ( 112 ) is generated. From this image, an “homogeneity-indicator” is determined for at least one region of interest, and the optical measurements are then evaluated in dependence on said indicator. The homogeneity-indicator may for example be a binary value which indicates if an inhomogeneity was detected or not. If an inhomogeneity was detected, all optical measurements may be rejected, only measurements for the involved region of interest may be rejected, or measurements for a selected sub-area of the involved region of interest (ROI) may be rejected.

Claims (30)

1. A method for evaluating an assay of a sample on a sensing surface performed with an optical analyzer including at least an image sensor and a computer, said method comprising:

generating at least one homogeneity-image of the sensing surface by imaging the sensing surface with the image sensor, the homogeneity-image including a plurality of pixels, each pixel having an image value;

using a computer executing software:

determining whether the image values of pixels in sub-areas of at least one region of interest (ROI) deviate from a given target range;

in response to the image values of the pixels in one or more sub-areas deviating from the given target range, controlling the optical analyzer to one of:

rejecting the assay with respect to a respective ROI in which the image values of the pixels in one or more sub-areas deviate from the target range, or

rejecting the assay with respect to one or more sub-areas of the respective ROI in which the image values of the pixels deviate from the target range; and

in response to the image values of the pixels in one or more sub-areas deviating from the given target range, assaying the sample based only on sub-areas of the respective ROI in which the pixels are within the given target range.

2. The method according to claim 1 wherein the assay comprises the specific binding of target components of the sample to at least one binding spot on the sensing surface.

3. The method according to claim 1 , further including:

generating a plurality of images of the sensing surface.

4. The method according to claim 1 , wherein the determining of whether the image values of pixels in sub-areas of at least one region of interest (ROI) deviate from the given target range is determined by a feature detection procedure.

5. The method according to claim 1 , wherein the determining of whether the image values of pixels in sub-areas of at least one region of interest (ROI) deviate from a given target range includes:

generating a histogram of the image values of the pixels within the at least one region of interest (ROI); and

determining whether the image values of the pixels in the sub-areas of at least one ROI deviate from the given target range based on whether a width of a main peak of the histogram falls within a given range.

6. The method according to claim 1 , wherein the homogeneity image is generated at the start of the assay, an end of the assay, and during the assay.

7. The method according to claim 1 , wherein determining whether the image values of pixels in sub-areas of at least one region of interest (ROI) deviate from a given target range includes:

determining a shape and a location of an inhomogeneity present on the sensor surface, wherein the shape and location is stored as data on a data carrier, and wherein the shape and location data is readable by the data analyzer for subsequent determination of the deviation from the target range.

8. The method according to claim 1 , wherein the optical analyzer further includes a light source, and the generating of the image includes emitting an input light beam toward the sensing surface using the light source.

9. The method according to claim 1 , wherein the optical analyzer further includes magnets, the method further includes disposing magnetic beads at the sensing surface, and the generating of the image includes applying a magnetic field to the magnetic beads using the magnets.

10. The method according to claim 9 , wherein the assaying comprises performing an assay to detect cardiac troponin.

11. The method according to claim 9 , wherein the assaying comprises performing an assay to detect parathyroid hormone (PTH).

12. An assay method for evaluating an assay of a sample on a sensing surface performed with an optical analyzer including at least an image sensor and a computer, said method comprising:

using the image sensor, generating at least one homogeneity-image of the sensing surface, the homogeneity-image including a plurality of pixels, each pixel having an image value; and

using a computer executing software:

determining whether the image values of pixels in sub-areas of at least one region of interest (ROI) deviate from a given target range, wherein the determining includes subtractively combining the homogeneity-image with a reference image of the ROI to generate a difference image and comparing pixels of the difference image to a given target image value; and

in response to the image values of the pixels in one or more sub-areas deviating from the given target range controlling the optical analyzer to one of:

reject the assay with respect to a respective ROI in which the image values of the pixels in one or more sub-areas deviate from the target range, or

reject the assay with respect to one or more sub-areas of the respective ROI in which the image values of the pixels deviate from the target range; and

in response to the image values of the pixels in one or more sub-areas deviating from the given target range, assaying the sample based only on sub-areas of the respective ROI in which the pixels are within the given target range.
