Abstract

The present invention relates to crystalline polymorphic forms of monosodium N-[8-(2-hydroxybenzoyl)amino]caprylate (“SNAC”), including two hydrates, a methanol solvate, and an ethanol solvate, of SNAC. More specifically, the present invention provide six polymorphic forms of SNAC (hereafter referred to as Forms I-VI). The present invention also provides an amorphous form of SNAC.

Claims (34)

1. A method of preparing Form I of monosodium N-[8-(2-hydroxybenzoyl)amino]caprylate comprising

(a) heating Form III, V, or VI of monosodium N-[8-(2-hydroxybenzoyl)amino]caprylate or a mixture thereof to at least 50° C. for a time sufficient to form Form I of monosodium N-[8-(2-hydroxybenzoyl)amino]caprylate;

(b) heating amorphous monosodium N-[8-(2-hydroxybenzoyl)amino]caprylate at from about 30 to about 90° C. for a time sufficient to form Form I of monosodium N-[8-(2-hydroxybenzoyl)amino]caprylate; or

(c) lyophilizing any form of monosodium N-[8-(2-hydroxybenzoyl)amino]caprylate other than Form I to yield Form I of monosodium N-[8-(2-hydroxybenzoyl)amino]caprylate.

2. The method of claim 1 , wherein in step (a) the SNAC monosodium N-[8-(2-hydroxybenzoyl)amino]caprylate is heated to from about 50 to about 110° C. for a time sufficient to form Form I of monosodium N-[8-(2-hydroxybenzoyl)amino]caprylate.

3. The method of claim 1 , wherein in step (b) the amorphous monosodium N-[8-(2-hydroxybenzoyl)amino]caprylate is heated at from about 40 to about 80° C.

4. The method of claim 1 , wherein step (c) comprises lyophilizing one or more of Forms II-VI of monosodium N-[8-(2-hydroxybenzoyl)amino]caprylate and/or amorphous monosodium N-[8-(2-hydroxybenzoyl)amino]caprylate to yield Form I of monosodium N-[8-(2-hydroxybenzoyl)amino]caprylate.

A method of preparing Form II of monosodium N-[8-(2-hydroxybenzoyl)amino]caprylate comprising the step of drying a solvate of monosodium N-[8-(2-hydroxybenzoyl)amino]caprylate without agitation and exposing the dried monosodium N-[8-(2-hydroxybenzoyl)amino]caprylate to moisture for a sufficient time to yield Form II of SNAC monosodium N-[8-(2-hydroxybenzoyl)amino]caprylate.

6. The method of claim 5 , wherein the solvate is an ethanol solvate or a methanol solvate.

7. A method of preparing Form III of monosodium N-[8-(2-hydroxybenzoyl)amino]caprylate comprising

(a) exposing Form I, II, or IV of monosodium N-[8-(2-hydroxybenzoyl)amino]caprylate or amorphous monosodium N-[8-(2-hydroxybenzoyl)amino]caprylate or a mixture thereof to an environment having a relative humidity of 75% or greater for a sufficient time to yield Form III of monosodium N-[8-(2-hydroxybenzoyl)amino]caprylate;

(b) wet granulating Form I of monosodium N-[8-(2-hydroxybenzoyl)amino]caprylate for a sufficient time to yield Form III of monosodium N-[8-(2-hydroxybenzoyl)amino]caprylate;

(c) exposing Form V or VI of monosodium N-[8-(2-hydroxybenzoyl)amino]caprylate or a mixture thereof to an environment having a relative humidity of 30% or greater for a sufficient time to yield Form III of monosodium N-[8-(2-hydroxybenzoyl)amino]caprylate;

(d) exposing Form VI of monosodium N-[8-(2-hydroxybenzoyl)amino]caprylate to an environment having a relative humidity of 10% or greater for a sufficient time to yield Form III of monosodium N-[8-(2-hydroxybenzoyl)amino]caprylate;

(e) exposing amorphous monosodium N-[8-(2-hydroxybenzoyl)amino]caprylate to moisture for a sufficient time to yield Form III of monosodium N-[8-(2-hydroxybenzoyl)amino]caprylate; or

(f) crystallizing monosodium N-[8-(2-hydroxybenzoyl)amino]caprylate from water to yield Form III of monosodium N-[8-(2-hydroxybenzoyl)amino]caprylate.

A method of preparing Form IV of monosodium N-[8-(2-hydroxybenzoyl)amino]caprylate comprising the step of heating Form I, II, III, V, or VI of monosodium N-[8-(2-hydroxybenzoyl)amino]caprylate or a mixture thereof to a temperature between about 110° C. and the melting point of monosodium N-[8-(2-hydroxybenzoyl)amino]caprylate for a sufficient time to yield Form IV of monosodium N-[8-(2-hydroxybenzoyl)amino]caprylate.

The method of claim 8 , wherein the Form II of monosodium N-[8-(2-hydroxybenzoyl)amino]caprylate is heated to between about 150° C. and the melting point of monosodium N-[8-(2-hydroxybenzoyl)amino]caprylate for a sufficient time to yield Form IV of monosodium N-[8-(2-hydroxybenzoyl)amino]caprylate.

10. A method of preparing Form V of monosodium N-[8-(2-hydroxybenzoyl)amino]caprylate comprising

(a) crystallizing monosodium N-[8-(2-hydroxybenzoyl)amino]caprylate from a methanol solution at a relative humidity of at least about 30% to yield Form V of monosodium N-[8-(2-hydroxybenzoyl)amino]caprylate;

(b) equilibration of Forms I-IV or VI of monosodium N-[8-(2-hydroxybenzoyl)amino]caprylate with methanol to yield Form V of monosodium N-[8-(2-hydroxybenzoyl)amino]caprylate; or

(c) slurrying any of Forms I-IV or VI of monosodium N-[8-(2-hydroxybenzoyl)amino]caprylate or a mixture thereof in methanol at a relative humidity of at least 30%, and maintaining the slurried mixture at ambient temperatures for a sufficient time to form Form V of monosodium N-[8-(2-hydroxybenzoyl)amino]caprylate.

11. The method of claim 10 , wherein the methanol solution is substantially free of water.

12. The method of claim 10 , wherein the equilibration in step (b) is performed in the absence of water.

13. A method of preparing Form VI of monosodium N-[8-(2-hydroxybenzoyl)amino]caprylate comprising

(a) crystallizing monosodium N-[8-(2-hydroxybenzoyl)amino]caprylate from an ethanol solution at a relative humidity of at least about 30% to yield Form VI of monosodium N-[8-(2-hydroxybenzoyl)amino]caprylate;

(b) preparing a saturated solution of monosodium N-[8-(2-hydroxybenzoyl)amino]caprylate in ethanol at a relative humidity of at least about 30% and cooling the resulting solution to room temperature or lower to yield Form VI of monosodium N-[8-(2-hydroxybenzoyl)amino]caprylate; or

(c) slurrying any of Forms I-IV of monosodium N-[8-(2-hydroxybenzoyl)amino]caprylate in ethanol at a relative humidity of at least about 30% to yield Form VI of monosodium N-[8-(2-hydroxybenzoyl)amino]caprylate.

14. The method of claim 13 , wherein the ethanol solution in step (a) is substantially free of water.

15. The method of claim 13 , wherein step (c) is performed in the absence of water.

16. A method of preparing amorphous monosodium N-[8-(2-hydroxybenzoyl)amino]caprylate comprising the step of dehydrating Form III, V, or VI of monosodium N-[8-(2-hydroxybenzoyl)amino]caprylate for a sufficient time to form amorphous monosodium N-[8-(2-hydroxybenzoyl)amino]caprylate.

17. The method of claim 16 , wherein Form III, V, or VI of monosodium N-[8-(2-hydroxybenzoyl)amino]caprylate is dehydrated in a vacuum to form amorphous monosodium N-[8-(2-hydroxybenzoyl)amino]caprylate.

18. The method of claim 16 , wherein Form III is dehydrated to form amorphous monosodium N-[8-(2-hydroxybenzoyl)amino]caprylate.

19. The method of claim 1 , wherein the method comprises (a) heating Form III of monosodium N-[8-(2-hydroxybenzoyl) amino]caprylate or a mixture thereof to at least 50° C. for a time sufficient to form Form I of monosodium N-[8-(2-hydroxybenzoyl) amino]caprylate.
