Abstract

Provided herein is technology relating to isolating nucleic acids. In particular, the technology relates to methods and kits for extracting nucleic acids from problematic samples such as stool.

Claims (25)

1. A kit for isolating a plurality of different target human DNAs from a human stool sample, the kit comprising:

a) a volume of at least 16 milliliters of a buffer solution for stool homogenization for processing a human stool sample having a mass of at least 4 grams using 4 milliliters of buffer solution per gram of stool;

b) a spin column comprising a spin filter comprising:

i) a hollow body (1);

ii) a bottom end (2); and

iii) an opening (3) at a top end of said hollow body (1), opposite the bottom end (2),

wherein the hollow body (1) and the bottom end (2) of said spin filter are both composed of the same porous filtering material,

c) a plurality of different magnetic target-specific capture particles, wherein different target-specific capture particles comprise covalently-attached oligonucleotides complementary to at least a portion of different target human DNAs, wherein said different target human DNAs comprise NDRG4, BMP3, and and KRAS,

and

d) guanidine thiocyanate.

2. The kit of claim 1 , wherein the porous filtering material is polyethylene.

3. The kit of claim 1 , wherein the bottom end (2) of said spin filter has a shape selected from the group consisting of a hemisphere, a disc, a cone, or a portion of an ellipsoid.

4. The kit of claim 1 , further comprising a magnet.

5. The kit of claim 1 , further comprising polyvinylpyrrolidone.

6. The kit of claim 5 wherein the polyvinylpyrrolidone is a polyvinylpolypyrrolidone.

7. The kit of claim 5 wherein the polyvinylpyrrolidone is in a premeasured form.

8. The kit of claim 7 wherein the polyvinylpyrrolidone is provided as a tablet.

9. The kit of claim 1 , further comprising an elution or wash solution.

10. The kit of claim 1 further comprising a vessel in which to hold isolated target human DNA.

11. The kit of claim 1 , further comprising a shipping container.

12. The kit of claim 1 , further comprising a nucleic acid control reagent.

13. The kit of claim 1 , wherein each of said oligonucleotides are selected from the group consisting of the oligonucleotides comprising SEQ ID NOS:2-3 and 6-7.

14. The kit of claim 1 , further comprising a magnetic target-specific capture particle comprising an oligonucleotide that is complementary to at least a portion of a reference DNA.

15. The kit of claim 14 , wherein said reference DNA is beta-actin.

16. The kit of claim 15 , wherein said oligonucleotide comprises SEQ ID NO:5.
