Abstract

Compounds of formula I: (wherein variables A 1 , m, R 1 , R 2 , R 3 , R 4 and Z are as described herein) which are antagonists of CGRP receptors and which are useful in the treatment or prevention of diseases in which the CGRP is involved, such as migraine. The invention is also directed to pharmaceutical compositions comprising these compounds and the use of these compounds and compositions in the prevention or treatment of such diseases in which CGRP is involved.

or a pharmaceutically acceptable salt or individual diastereomer thereof.

8 . A pharmaceutical composition which comprises an inert carrier and one or more compounds of claim 3 or a salt thereof.

9 . A method for antagonism of CGRP receptor activity in a mammalian patient in need thereof, said method comprising the step of administering to said patient an effective amount of one or more compounds of claim 3 or a salt thereof.

10 . A method for treating headache in a mammalian patient in need thereof, said method comprising the step of administering to said patient a therapeutically effective amount of one or more compounds of claim 3 or a salt thereof.

11 . A method for treating migraine headache or cluster headache in a mammalian patient in need thereof, said method comprising the step of administering to said patient a therapeutically effective amount of one or more compounds of claim 3 or a salt thereof.

12 . A method of treating migraine headaches or cluster headaches in a mammalian patient in need thereof, said method comprising the step of co-administration to said patient a therapeutically effective amount of and one or more compounds of claim 3 or a pharmaceutically acceptable salt thereof, and a therapeutically effective amount of a second agent selected from serotonin agonists, analgesics, anti-inflamatory agents, anti-hypertensives and anticonvulsants.
