Abstract

The invention provides heterodimeric antibodies comprising a first heavy chain comprising a first Fc domain and a single chain Fv region that binds a first antigen. The heterodimeric antibodies also comprise a second heavy chain comprising a second Fc domain, a first variable heavy chain and a first variable light chain, wherein the first and second Fc domains are different.

Claims (30)

1. A heterodimeric antibody comprising:

a) a first heavy chain comprising:

i) a first variant Fc domain; and

ii) a single chain Fv (scFv) region that binds a first antigen; and

b) a second heavy chain comprising a VH-CH1-hinge-CH2-CH3 region, wherein said CH2-CH3 is a second variant Fc domain; and

c) a first light chain comprising:

i) a first variable light chain; and

ii) a first constant light chain;

wherein said first and second variant Fc domains comprise amino acid variant set L368E/K370S and S364K according to the EU index as in Kabat, and wherein said first and second variant Fc domains are variant human IgG Fc domains.

2. A heterodimeric antibody according to claim 1 wherein said scFv region binds to CD3.

3. A composition comprising a heterodimeric antibody according to claim 1 and a pharmaceutical carrier.

4. A method of purifying a heterodimeric antibody comprising:

a) providing a heterodimeric antibody according to claim 1 ;

b) loading said heterodimeric antibody onto an ion exchange column; and

c) collecting the heterodimeric fraction.

5. The heterodimeric antibody of claim 1 , wherein said first Fc domain comprises amino acid substitutions L368E and K370S and said second Fc domain comprises amino acid substitution S364K.

6. The heterodimeric antibody of claim 1 , wherein said first Fc domain comprises amino acid substitution S364K and said second Fc domain comprises amino acid substitutions L368E and K370S, according to the EU index as in Kabat.

7. The heterodimeric antibody of claim 1 wherein said first variant and second variant Fc domains are selected from the group consisting of human IgG1, IgG2 and IgG4 variant Fc domains.

8. The heterodimeric antibody of claim 2 wherein said first variant and second variant Fc domains are selected from the group consisting of human IgG1, IgG2 and IgG4 variant Fc domains.

9. The heterodimeric antibody of claim 6 wherein said first variant and second variant Fc domains are selected from the group consisting of human IgG1, IgG2 and IgG4 variant Fc domains.

10. A composition comprising a heterodimeric antibody according to claim 2 and a pharmaceutical carrier.

11. A composition comprising a heterodimeric antibody according to claim 6 and a pharmaceutical carrier.

12. A method of purifying a heterodimeric antibody comprising:

a) providing a heterodimeric antibody according to claim 2 ;

b) loading said heterodimeric antibody onto an ion exchange column; and

c) collecting the heterodimeric fraction.

13. A method of purifying a heterodimeric antibody comprising:

a) providing a heterodimeric antibody according to claim 6 ;

b) loading said heterodimeric antibody onto an ion exchange column; and

c) collecting the heterodimeric fraction.
