Abstract

A biologic artificial bone includes an artificial fiber material formed from a synthetic polymer with mechanical properties similar to type I collagen. A biocompatible liquid substance is impregnated in the fiber material that hardens and stiffens the fiber material. A bone substitute is impregnated in the hardened and stiffened fiber material forming an artificial bone composite. Vascular channels are formed in the artificial bone composite to facilitate in-growth of vessels and bone forming cells. The construction and methods achieve an artificial composite structure that is similar to natural bone with comparable properties.

Claims (26)

1. A method of constructing a biologic artificial bone comprising:

(a) forming an artificial fiber material from a synthetic polymer with mechanical properties similar to type I collagen, wherein the artificial fiber material is formed into a hollow cast shaped corresponding to a periphery of a human bone, wherein the center being substantially empty to facilitate population of the biological artificial bone with blood forming cells, and wherein the artificial fiber material comprises fibers formed in a fiber network with fibers arranged in multiple orientations in a single sheet, wherein the artificial fiber material comprises fibers formed in a fiber network with fibers arranged in horizontal, vertical and oblique orientations while maintaining the hollow cast shape corresponding to a periphery of human bone prior to, during and after impregnating with a biocompatible liquid;

(b) impregnating the artificial fiber material with the biocompatible liquid, the biocompatible liquid hardening and stiffening the fiber material;

(c) impregnating the hardened and stiffened fiber material with a bone substitute, thereby forming an artificial bone composite wherein the artificial bone composite has structural characteristics by virtue of the artificial fiber in the hollow cast, the biocompatible liquid and the bone substitute such that the artificial bone composite retains its shape after installation wherein step is practiced by impregnating with liquid ceramics; and

(d) prior to installation, forming vascular channels in the artificial bone composite to facilitate in-growth of vessels and bone forming cells.

2. The method according to claim 1 , wherein step (c) is practiced by immersing the hardened and stiffened fiber material into a mold containing the liquid ceramic.

3. The method according to claim 1 , wherein step (c) is practiced by placing the hardened and stiffened fiber material in a mold and pouring ceramic into the mold and allowing the liquid ceramic to set.

4. The method according to claim 1 , wherein step (c) is practiced by grit-blasting with a ceramic the hardened and stiffened fiber material impregnated with the ceramic.

5. The method according to claim 1 , wherein step (a) comprises rolling the artificial fiber material into a cylinder to produce a desired shape prior to step (b).

6. The method according to claim 1 , further comprising, prior to step (b), wrapping the artificial fiber material with a removable porous sheet of absorbable material having spikes thereon facing the artificial fiber material, wherein the spikes penetrate the artificial fiber material.

7. The method according to claim 6 , further comprising, after step (c) removing the porous sheet of absorbable material by one of chemical dissolution, heating, or dissolution in a fluid prior to installation.

The method according to claim 1 , further comprising, prior to step (b), placing the artificial fiber material in a mold including spikes facing the artificial fiber material, the spikes on the mold penetrating the artificial fiber material, and after step (c), removing the spikes prior to installation.

9. The method according to claim 1 , wherein step (d) is practiced by micro-drilling the channels.

10. A method of constructing a biologic artificial bone comprising:

forming an artificial fiber material from a synthetic polymer with mechanical properties similar to type I collagen, wherein the artificial fiber material is formed into a hollow cast shaped corresponding to a periphery of a human bone, wherein the center being substantially empty to facilitate population of the biological artificial bone with blood forming cells, while maintaining the hollow cast shape corresponding to a periphery human bone prior to, during and after impregnating with a biocompatible liquid;

providing the artificial fiber material with fibers formed in a fiber network including fibers arranged in multiple orientations in a single sheet, and providing the artificial fiber material with fibers formed in a fiber network including fibers arranged in horizontal, vertical and oblique orientations;

impregnating the artificial fiber material with the biocompatible liquid, the biocompatible liquid hardening and stiffening the artificial fiber material wherein step is practiced by impregnating with liquid ceramics;

impregnating a bone substitute in the hardened and stiffened artificial fiber material forming an artificial bone composite, wherein the artificial bone composite has structural characteristics by virtue of a combination of the hollow cast shape, the biocompatible liquid hardening and stiffening and the addition of the bone substitute wherein the artificial bone composite material being formed into the hollow cast shape retains its shape after installation; and

prior to installation, forming vascular channels in the artificial bone composite to facilitate in-growth of vessels and bone forming cells.

11. The method according to claim 10 , wherein the bone substitute comprises ceramics.

12. The method according to claim 10 , further comprising coating the channels with angiogenic factors.

The method according to claim 10 , further comprising wrapping the artificial bone composite with a porous sheet of absorbable material having a surface adjacent the artificial fiber network with dissolvable spikes, the porous sheet being wrapped on the artificial fiber network of the artificial bone composite with the spikes facing inward to penetrate the artificial fiber network of the artificial bone composite, thereby creating the vascular channels for vascular in-growth after implantation of the biologic artificial bone.

14. The method according to claim 10 , further comprising providing a single or multiple stumps of the artificial fiber material in continuity with the artificial bone composite for attachment of tendons after implantation of the biologic artificial bone.

15. The method according to claim 10 , further comprising providing a trabecular network of ceramic plates augmented with a polymer network that confer compressive strength to ends of the artificial bone.

16. The method according to claim 10 , wherein the step of providing the artificial fiber material with fibers formed in a fiber network includes having fibers arranged in vertical, horizontal and spiral orientations and forming a concentric cylindrical arrangement of fibers.

17. The method according to claim 10 , wherein the step of providing the artificial fiber material with fibers formed in a fiber network includes having fibers arranged in vertical, horizontal and spiral orientations and forming a fibrous sheet rolled into a multilayer cylinder.
