Abstract

The present invention relates to antibodies and antigen binding fragments thereof which bind to the human CD70 protein with high affinity and display potent inhibition of tumour cell growth.

Claims (42)

1. An isolated polynucleotide that encodes a monoclonal antibody, or a fragment thereof, which specifically binds to an epitope of human CD70, wherein the epitope is within the amino acid sequence HIQVTLAICSS (SEQ ID NO:342), wherein the monoclonal antibody or fragment thereof, when tested as a Fab fragment, has an off-rate (k off ) as measured by surface plasmon resonance for human CD70 of less than 7×10 −4 s −1.

The isolated polynucleotide of claim 1 , wherein the monoclonal antibody or fragment thereof comprises a heavy chain variable (VH) domain and a light chain variable (VL) domain, wherein the VH and VL domains, or one or more complementarity-determining regions (CDRs) thereof, are derived from a camelid species.

An isolated polynucleotide that encodes a monoclonal antibody, or a fragment thereof, which specifically binds to human CD70, said monoclonal antibody or fragment thereof comprising a heavy chain variable (VH) domain comprising CDR3 (HCDR3), CDR2 (HCDR2), and CDR1 (HCDR1), regions, and a light chain variable (VL) domain comprising CDR3 (LCDR3), CDR2 (LCDR2), and CDR1 (LCDR1) regions, wherein the HCDR3, HCDR2, HCDR1, LCDR3, LCDR2, and LCDR1 regions comprise the amino acid sequence set forth in

SEQ ID NO:50, SEQ ID NO:27, SEQ ID NO:11, SEQ ID NO:160, SEQ ID NO:119, and SEQ ID NO:250, respectively.

4. The isolated polynucleotide of claim 3 , wherein the

VH domain comprises the amino acid sequence of SEQ ID NO:223, and the VL domain comprises the amino acid sequence of SEQ ID NO:241.

An isolated polynucleotide that encodes a monoclonal antibody, or a fragment thereof, which specifically binds to human CD70, said monoclonal antibody or fragment thereof comprising a heavy chain variable (VH) domain comprising CDR3 (HCDR3), CDR2 (HCDR2), and CDR1 (HCDR1) regions, and a light chain variable (VL) domain compising CDR3 (LCDR3), CDR2 (LCDR2), and CDR1 (LCDR1) regions, wherein

the HCDR3 region comprises the amino acid sequence of SEQ ID NO:50;

the HCDR2 region comprises an amino acid sequence selected from the group consisting of: SEQ ID NO:27 and SEQ ID NO:249; and

the HCDR1 region comprises an amino acid sequence selected from the group consisting of: SEQ ID NO:11 and SEQ ID NO:248.

6. The isolated polynucleotide of claim 5 , wherein

the HCDR3 region comprises SEQ ID NO:50;

the HCDR2 region comprises SEQ ID NO:27; and

the HCDR1 region comprises SEQ ID NO:11.

7. The isolated polynucleotide of claim 5 , wherein the LCDR3 region comprises SEQ ID NO:160;

the LCDR2 region comprises an amino acid sequence selected from the group consisting of: SEQ ID NO:119 and SEQ ID NO:110; and

the LCDR1 region comprises an amino acid sequence selected from the group consisting of: SEQ ID NO:87, SEQ ID NO:250, SEQ ID NO:251, SEQ ID NO:252, and SEQ ID NO:253.

8. The isolated polynucleotide of claim 7 , wherein the LCDR3 region comprises SEQ ID NO:160;

the LCDR2 region comprises SEQ ID NO:119; and

the LCDR1 region comprises SEQ ID NO:250.

9. The isolated polynucleotide of claim 8 , wherein the HCDR3 region consists of SEQ ID NO:50;

the HCDR2 region consists of SEQ ID NO:27;

the HCDR1 region consists of SEQ ID NO:11;

the LCDR3 region consists of SEQ ID NO:160;

the HCDR2 region consists of SEQ ID NO:119; and

the HCDR1 region consists of SEQ ID NO:250.

10. The isolated polynucleotide of claim 3 , wherein

the VH domain consists of the amino acid sequence shown as SEQ ID NO:223, and the VL domain consist of the amino acid sequence shown as SEQ ID NO:241.

11. An isolated polynucleotide that encodes a monoclonal antibody, or a fragment thereof, which specifically binds to human CD70, said monoclonal antibody or fragment thereof comprising:

a) a heavy chain variable (VH) domain with the amino acid sequence set forth in SEQ ID NO:178 comprising one or more amino acid substitutions at Kabat positions selected from the group consisting of H6, H18, H24, H31, H56, H74, H77, H79, H83, H84, H89, H93, H94, H108, H110, and H112; and

b) a light chain variable (VL) domain with the amino acid sequence set forth in SEQ ID NO:201 comprising one or more amino acid substitutions at Kabat positions selected from the group consisting of L2, L11, L12, L25, L26, L30, L46, L53, L60, L61, L67, L68, L76, L80, L81, L85, and L87, wherein the thermal stability of the antibody, or the fragment thereof, is greater than the thermal stability of an antibody, or antigen binding fragment thereof, comprising a heavy chain variable domain with the amino acid sequence set forth in SEQ ID NO:178 and a light chain variable domain with the amino acid sequence set forth in SEQ ID NO:201.

12. The isolated polynucleotide encoding the monoclonal antibody according to claim 11 which is a llama-human chimeric antibody, wherein the VH and VL domains, or one or more CDRs thereof are derived from llama, and one or more of the constant domains are derived from a human immunoglobulin.

13. The isolated polynucleotide of claim 1 , wherein the monoclonal antibody or fragment thereof, when tested as a Fab fragment, has an off-rate (k off ) as measured by surface plasmon resonance for human CD70 of less than or equal to 4.8×10 −4 s −1 .

14. The isolated polynucleotide of claim 1 , wherein the monoclonal antibody or fragment thereof is a chimeric antibody or a humanized antibody.

15. The isolated polynucleotide of claim 1 , wherein the monoclonal antibody or fragment thereof is a chimeric antibody, wherein the chimeric antibody comprises a constant domain derived from a human immunoglobulin.

16. The isolated polynucleotide according to claim 15 , wherein the constant domain is derived from a human immunoglobulin selected from the group consisting of IgG1, IgG2, IgG3, and IgG4.

17. The isolated polynucleotide according to claim 15 , wherein the constant domain is derived from human IgG1.

18. The isolated polynucleotide of claim 1 , wherein the monoclonal antibody or fragment thereof is a llama-human chimeric antibody comprising VH and VL domains, or one or more CDRs thereof, derived from llama, and one or more constant domains derived from a human immunoglobulin.

The isolated polynucleotide of claim 1 , wherein the monoclonal antibody or fragment thereof is capable of directing immune effector function against a cell expressing human CD70 on its surface, wherein the immune effector function is selected from the group consisting of antibody-dependent cellular cytotoxicity (ADCC), complement-dependent cytotoxicity (CDC), antibody-dependent cellular phagocytosis (ADCP), and any combination thereof.

20. The isolated polynucleotide of claim 1 , wherein the monoclonal antibody or fragment thereof is an scFc, a F(ab′) 2 fragment, a Fab fragment, a Fab′ fragment, an Fd fragment, an Fv fragment, or a single domain antibody (Dab) fragment.

21. The isolated polynucleotide of claim 3 , wherein the VH domain comprises the amino acid sequence of SEQ ID NO:223.

22. The isolated polynucleotide of claim 3 , wherein VL domain comprises the amino acid sequence of SEQ ID NO:241.
