Abstract

Described are methods for modifying a substrate by exposing the substrate to a densified fluid. The substrate may be a polymer or a metal alloy, and the densified fluid may be carbon dioxide. Uses of such substrate modification include impregnation of the substrate with one or more drugs, impregnation of microcellular particles, and formation of microcellular compositions.

Claims (31)

1. A method of impregnating a substrate with a drug compound,

comprising:

preconditioning the substrate with an organic liquid solvent penetrating agent to provide a conditioned substrate;

positioning the conditioned substrate in a reactor;

introducing the drug compound into the reactor; and

introducing densified carbon dioxide into the reactor, the densified carbon dioxide introduced into the reactor under conditions such that the drug compound is initially dispersed in the densified carbon dioxide, and thereafter transfers from the fluid and impregnates a matrix of the substrate.

2. The method of claim 1 , wherein the drug compound comprises a plurality of biologically active components.

3. The method of claim 1 , wherein the densified fluid is introduced into the reactor under subcritical conditions.

4. The method of claim 3 , wherein the subcritical conditions are maintained in the reactor for a controlled period of time, whereupon the conditions in the reactor are adjusted to supercritical conditions.

5. The method of claim 1 , wherein the densified fluid is introduced into the reactor under supercritical conditions.

6. The method of claim 1 , wherein the drug compound comprises microencapsulated drug particles.

7. The method of claim 6 , wherein the microencapsulated drug particles are impregnated in the substrate in a manner such that the particles are capable of delivering a fixed amount of drug per a selected time period.

8. The method of claim 1 , wherein the organic solvent penetrating agent comprises an alcohol, a ketone, an ether, or an aldehyde.

9. The method of claim 1 , also comprising removing at least part of the organic liquid solvent from the substrate prior to said introducing densified carbon dioxide.

10. The method of claim 1 , wherein said densified carbon dioxide drives at least some of said organic liquid solvent out of the substrate.

11. A method of impregnating a starting substrate with at least one active compound, comprising:

preconditioning the starting substrate with an organic liquid solvent penetrating agent to provide a conditioned substrate;

positioning the conditioned substrate in a reactor;

introducing the at least one active compound in particulate form into the reactor;

introducing densified carbon dioxide into the reactor under conditions wherein amounts of the at least one active compound are dissolved in the densified carbon dioxide to form an active compound-containing densified carbon dioxide and wherein the active compound-containing densified carbon dioxide penetrates into a matrix of the substrate;

removing the densified fluid from the matrix of the substrate so as to leave amounts of the at least one active compound in the matrix of the substrate; and

wherein said conditioned substrate is more susceptible to impregnation by the at least one active compound during said introducing densified carbon dioxide step than said starting substrate.

12. The method of claim 11 , wherein the matrix of the substrate comprises a polymeric matrix.

13. The method of claim 12 , wherein the polymeric matrix comprises a thermoplastic polyurethane elastomer or silicone elastomer.

14. The method of claim 12 , also comprising masking a portion of an exterior surface of the substrate sufficiently to resist penetration of the active compound-containing densified fluid into polymeric material located under said masked portion.

15. The method of claim 13 , wherein the substrate comprises catheter tubing.

16. The method of claim 15 , wherein said one or more active compounds comprise rifampin or minocycline.

17. The method of claim 11 , wherein the substrate comprises catheter tubing.

18. The method of claim 17 , wherein said one or more active compounds comprise rifampin and minocycline.

19. The method of claim 11 , wherein the one or more active compounds comprise one or more antimicrobial agents.

20. The method of claim 11 , wherein said organic solvent penetrating agent comprises an alcohol, a ketone, an ether, or an aldehyde.
