Abstract

A modified beetle luciferase protein which is an environmentally sensitive reporter protein is provided.

Claims (21)

1. An isolated polynucleotide encoding a modified beetle luciferase comprising an internal insertion of a heterologous amino acid sequence which interacts with a molecule of interest, wherein the activity of the modified beetle luciferase is altered after the molecule of interest interacts with the insertion sequence relative to the activity before the interaction, wherein the internal insertion is at a residue or region in a beetle luciferase sequence which is tolerant to modification, the isolated polynucleotide further comprising a polynucleotide encoding a tag of at least one amino acid fused to the N-terminus, C-terminus, or both, of the modified beetle luciferase.

2. The polynucleotide of claim 1 , wherein the internal insertion is in a hinge region of the luciferase.

3. The polynucleotide of claim 1 wherein the modified beetle luciferase further comprises a deletion of beetle luciferase residues N-terminal and/or C-terminal to the insertion.

4. The polynucleotide of claim 3 wherein the deletion is no more than 15 residues of beetle luciferase sequence.

5. The polynucleotide of claim 4 wherein the modified beetle luciferase further comprises a deletion of beetle luciferase residues corresponding to the N-terminus and/or C-terminus of the unmodified beetle luciferase.

6. The polynucleotide of claim 5 wherein the deletion is no more than 15 residues of beetle luciferase sequence.

7. The polynucleotide of claim 1 wherein the modified beetle luciferase has at least about 50% the activity of the corresponding unmodified beetle luciferase.

8. The polynucleotide of claim 1 wherein the modified beetle luciferase has at least about 1% the activity of the corresponding unmodified beetle luciferase.

9. The polynucleotide of claim 1 wherein the tag is a PEST sequence, a GST sequence, a polyhistidine sequence or a reporter protein.

10. The polynucleotide of claim 1 wherein the insertion is about 4 to about 50 amino acid residues.

11. The polynucleotide of claim 1 wherein the beetle luciferase is a click beetle luciferase.

12. The polynucleotide of claim 1 wherein the beetle luciferase is a firefly luciferase.

13. A vector comprising the polynucleotide of claim 1 .

14. A host cell comprising the vector of claim 13 .

15. An isolated polynucleotide encoding a circularly permuted modified beetle luciferase comprising an internal insertion of a heterologous amino acid sequence which interacts with a molecule of interest, wherein the activity of the modified beetle luciferase is altered after the molecule of interest interacts with the insertion sequence relative to the activity before the interaction, wherein the circular permutation is at a residue or region in a beetle luciferase sequence which is tolerant to modification.

16. The polynucleotide of claim 15 , wherein the insertion is a protease cleavage site.

17. The polynucleotide of claim 16 wherein the protease cleavage site is inserted at or near residues corresponding to the N- and C-terminus of the corresponding nonpermuted luciferase.

18. The polynucleotide of claim 16 wherein cleavage at the protease cleavage site results in an increase in luminescence.

The polynucleotide of claim 16 where the protease cleavage site is a caspase 3 cleavage site, a caspase 8 cleavage site, an enterokinase cleavage site, a prostate serum antigen cleavage site, a severe acute respiratory syndrome (SARS) viral protease cleavage site, a tobacco etch virus (TEV) protease cleavage site (NLYFQG; SEQ ID NO:118), or a rhinovirus protease cleavage site.

20. A vector comprising the polynucleotide of claim 15 .

21. A host cell comprising the vector of claim 20 .
