Abstract

The present invention is directed to antibodies and fragments thereof (especially chimeric and humanized) having binding specificity for HGF and their use in therapy and diagnosis. These antibodies inhibit or block HGF-associated activities including HGF's effects on cell proliferation, invasion, angiogenesis, metastasis and fibrosis. Particularly the antibodies may be used as a monotherapy or in combination therapies in treating cancer, other proliferative disorders and other conditions wherein inhibition of HGF and/or the HGF/HGF-R (c-met) interaction is desired.

Claims (17)

1. A humanized antibody or humanized antibody fragment that specifically binds hepatocyte growth factor (“HGF”) and said antibody or antibody fragment inhibits or neutralizes at least one HGF associated biological activity, wherein said humanized antibody or humanized antibody fragment comprises a variable heavy chain comprising CDR1, CDR2 and CDR3 polypeptides respectively having the amino acid sequences of SEQ ID NO: 4; SEQ ID NO: 6; and SEQ ID NO: 8 and a variable light chain comprising CDR1, CDR2 and CDR3 polypeptides respectively having the amino acid sequences of SEQ ID NO: 24; SEQ ID NO: 26; and SEQ ID NO: 28.

2. The human zed antibody of claim 1 which comprises human IgG1, IgG2, IgG3 or IgG4 constant regions.

3. A method of treating a cancer condition associated with the overexpression of HGF comprising administering a therapeutically effective amount of a humanized antibody according to claim 2 , wherein proliferation of cancer cells is inhibited.

4. The method of claim 3 wherein the antibody binds to HGF with a Kd of 5×10 −8 M or less.

5. The humanized antibody of claim 1 which comprises human IgG1 constant regions.

6. A method of treating a cancer condition associated with the overexpression of HGF comprising administering a therapeutically effective amount of a humanized antibody according to claim 5 , wherein proliferation of cancer cells is inhibited.

7. The method of claim 6 wherein the antibody binds to HGF with a Kd of 5×10 −8 M or less.

8. The humanized antibody of claim 1 which comprises a full length humanized antibody wherein the variable heavy chain comprises an amino acid sequence at least 90% identical to SEQ ID NO:2 and the variable light chain comprises an amino acid sequence at least 90% identical to SEQ ID NO:22.

9. A method of treating a cancer condition associated with the overexpression of HGF comprising administering a therapeutically effective amount of a humanized antibody according to claim 8 , wherein proliferation of cancer cells is inhibited.

10. The method of claim 9 wherein the antibody binds to HGF with a Kd of 5×10 −8 M or less.

11. The humanized antibody of claim 1 which comprises a full length humanized antibody wherein the variable heavy chain comprises an amino acid sequence at least 95% identical to SEQ ID NO:2 and the variable light chain comprises an amino acid sequence at least 95% identical to SEQ ID NO:22.

12. A method of treating a cancer condition associated with the overexpression of HGF comprising administering a therapeutically effective amount of a humanized antibody according to claim 11 , wherein proliferation of cancer cells is inhibited.

13. The method of claim 12 wherein the antibody binds to HGF with a Kd of 5×10 −8 M or less.

14. The humanized antibody of claim 1 which comprises a humanized antibody fragment selected from a Fab fragment, Fab′ fragment, and a F(ab′) 2 fragment.

15. A method of treating a cancer condition associated with the overexpression of HGF comprising administering a therapeutically effective amount of a humanized antibody fragment according to claim 14 , wherein proliferation of cancer cells is inhibited.

16. The method of claim 15 wherein the antibody binds to HGF with a Kd of 5×10 −8 M or less.

17. A method of treating a cancer condition associated with the overexpression of HGF comprising administering a therapeutically effective amount of a humanized antibody according to claim 1 , wherein proliferation of cancer cells is inhibited.
