Abstract

The present invention provides isolated anti-interferon alpha monoclonal antibodies, particularly human monoclonal antibodies, that inhibit the biological activity of multiple interferon (IFN) alpha subtypes but do not substantially inhibit the biological activity of IFN alpha 21 or the biological activity of either IFN beta or IFN omega. Immunoconjugates, bispecific molecules and pharmaceutical compositions comprising the antibodies of the invention are also provided. The invention also provides methods for inhibiting the biological activity of IFN alpha using the antibodies of the invention, as well as methods of treating disease or disorders mediated by IFN alpha, such as autoimmune diseases, transplant rejection and graft versus host disease, by administering the antibodies of the invention.

Claims (11)

1. A method for preparing an anti-interferon alpha monoclonal antibody, comprising:

(a) providing: a first antibody or antigen-binding portion thereof comprising a heavy chain variable region that comprises CDR1, CDR2, and CDR3 domains and a light chain variable region that comprises CDR1, CDR2, CDR3 domains, wherein the heavy chain variable region CDR1 domain comprises the amino acid sequence set forth in SEQ ID NO: 1, the heavy chain variable region CDR2 domain comprises the amino acid sequence set forth in SEQ ID NO: 4, the heavy chain variable region CDR3 domain comprises the amino acid sequence set forth in SEQ ID NO: 7, the light chain variable region CDR1 domain comprises the amino acid set forth in SEQ ID NO: 10, the light chain variable region CDR2 domain comprises the amino acid sequence set forth in SEQ ID NO: 13, and the light chain variable region CDR3 domain comprises the amino acid sequence set forth in SEQ ID NO: 16;

(b) altering at least one amino acid residue within one or both of the heavy chain variable region and light chain variable region of the first antibody or antigen-binding portion thereof to create an altered antibody comprising no more than five amino acid alterations compared to the heavy and light chain variable regions of the first antibody or antigen-binding portion thereof;

(c) expressing the altered antibody as a protein;

(d) assessing the binding affinity of the altered antibody;

(e) identifying the altered antibody that binds specifically to human interferon alpha; and

(f) obtaining the altered antibody that binds specifically to human interferon alpha.

2. The method of claim 1 , wherein the no more than five amino acid alterations are within the heavy chain variable region of the first antibody or antigen-binding portion thereof.

3. The method of claim 2 , wherein the no more than five amino acid alterations are within the heavy chain variable region CDR1 domain, the heavy chain variable region CDR2 domain, or a combination thereof of the first antibody or antigen-binding portion thereof.

4. The method of claim 1 , wherein the altered antibody has one, two, or five amino acid alterations compared to the heavy and light chain variable regions of the first antibody or antigen-binding portion thereof.

5. The method of claim 1 , wherein the amino acid alterations are selected from amino acid substitutions, amino acid additions, amino acid deletions, and combinations thereof.
