Polycyclic heteroaryl substituted triazoles useful as axl inhibitors
View Patent ↗Polycyclic heteroaryl substituted triazoles and pharmaceutical compositions containing the compounds are disclosed as being useful in inhibiting the activity of the receptor protein tyrosine kinase Axl. Methods of using the compounds in treating diseases or conditions associated with Axl activity are also disclosed.
1. A compound having the structure:
as a free base, a pharmaceutically acceptable salt thereof or an N-oxide thereof.
2. The compound of claim 1 which is the free base.
3. The compound of claim 1 which is a pharmaceutically acceptable salt thereof.
4. The compound of claim 1 wherein the pharmaceutically acceptable salt is a pharmaceutically acceptable acid addition salt.
5. The compound of claim 4 wherein the acid is formic acid, trifluoroacetic acid, hydrochloric acid, benzenesulfonic acid or acetic acid.
6. A pharmaceutical composition comprising a pharmaceutically acceptable excipient and a compound of claim 1 , as a free base, a pharmaceutically acceptable salt thereof, or an N-oxide thereof.

A method for stabilizing a disease, inhibiting a disease, arresting the development of a disease, or relieving a disease in a mammal, wherein the disease is associated with Axl activity and is selected from rheumatoid arthritis, vascular disease, vascular injury, psoriasis, visual impairment due to macular degeneration, diabetic retinopathy, retinopathy of prematurity, kidney disease, osteoporosis, osteoarthritis, cataracts, a solid tumor comprising breast carcinoma, renal carcinoma, endometrial carcinoma, ovarian carcinoma, thyroid carcinoma, non-small cell lung carcinoma, melanoma, prostate carcinoma, sarcoma, gastric cancer or uveal melanoma, a liquid tumor comprising myeloid leukemia or lymphoma, and endometriosis, wherein the method comprises administering to the mammal a therapeutically effective amount of a compound of claim 1 , as a free base, a pharmaceutically acceptable salt thereof, or an N-oxide thereof.
8. The method of claim 7 wherein the disease is alleviated by the modulation of Axl activity.
9. The method of claim 7 wherein the disease is alleviated by a increase in Axl activity.
10. The method of claim 7 wherein the disease is alleviated by a decrease in Axl activity.
11. The method of claim 10 wherein the disease is rheumatoid arthritis, vascular disease, vascular injury, psoriasis, visual impairment due to macular degeneration, diabetic retinopathy, retinopathy of prematurity, kidney disease, osteoporosis, osteoarthritis or cataracts.
12. The method of claim 10 , wherein the disease is a solid tumor comprising breast carcinoma, renal carcinoma, endometrial carcinoma, ovarian carcinoma, thyroid carcinoma, non-small cell lung carcinoma, melanoma, prostate carcinoma, sarcoma, gastric cancer or uveal melanoma.
13. The method of claim 10 , wherein the disease is a liquid tumor comprising myeloid leukemia or lymphoma.
14. The method of claim 10 wherein the disease is endometriosis.
