Abstract

Provided are methods for introducing a molecule of interest into a plant cell comprising a cell wall. Methods are provided for genetically or otherwise modifying plants and for treating or preventing disease in plant cells comprising a cell wall.

Claims (31)

1. A method of introducing a molecule of interest into a plant cell having a cell wall, the method comprising:

providing the plant cell having a cell wall;

coating a nanoparticle with a nucleic acid molecule of interest, wherein the nanoparticle is a gold nanoparticle or quantum dot;

further coating the nanoparticle with a second molecule of interest;

placing the cell having a cell wall and the coated nanoparticle in contact with each other; and

allowing uptake of the nanoparticle and the molecule of interest into the cell comprising a cell wall.

2. A method of expressing a gene, the method comprising:

providing a plant cell having a cell wall;

coating a nanoparticle with a gene, wherein the nanoparticle is a gold nanoparticle or quantum dot;

coating the nanoparticle with a subcellular compartment targeting protein;

placing the plant cell having a cell wall and the coated nanoparticle in contact with each other;

allowing uptake of the nanoparticle and the gene into the plant cell comprising a cell wall; and

expressing the gene.

3. The method according to claim 2 , wherein the gene is expressed in the chloroplast.

4. The method according to claim 2 , further comprising selecting for cells expressing the gene.

5. A method for transferring a molecular substance into a plant cell, comprising:

coating a nanoparticle with a plasmid DNA, wherein the nanoparticle is a gold nanoparticle or quantum dot;

coating the nanoparticle with a protein capable of targeting the nanoparticle to a subcellular compartment; and

contacting the coated nanoparticle with an intact, cell wall-bearing plant cell under conditions permitting the uptake of the coated nanoparticles into the plant cell,

wherein the nanoparticle is targeted to the subcellular compartment of the plant cell by the protein capable of targeting the nanoparticle to a subcellular compartment.

6. The method according to claim 5 , wherein the nanoparticle comprises gold.

7. The method according to claim 5 , wherein uptake of the nanoparticle is allowed into a subcellular compartment targeted by the protein capable of targeting the nanoparticle to a subcellular compartment.

8. The method according to claim 5 , wherein the subcellular compartment is a chloroplast.

9. The method according to claim 1 , wherein the second molecule of interest is selected from the group consisting of amino acids, polypeptides, enzymes, peptide hormones, glycol-peptides, signaling peptides, and antibodies.

10. The method according to claim 1 , wherein the second molecule of interest is selected from the group consisting of messengers, second messengers, and cAMP.

11. The method according to claim 1 , wherein the second molecule of interest is a sugar.

12. The method according to claim 1 , wherein the second molecule of interest is a fat.

13. The method according to claim 1 , wherein the second molecule of interest is a vitamin.

14. The method according to claim 1 , wherein the second molecule of interest is a herbicide.

15. The method according to claim 1 , wherein the second molecule of interest is a fungicide.

16. The method according to claim 1 , wherein the second molecule of interest is an antibiotic.
