Abstract

Protein enriched micro-vesicles and methods of making and using the same are provided. Aspects of the methods include maintaining a cell having a membrane-associated protein comprising a first dimerization domain and a target protein having a second dimerization domain under conditions sufficient to produce a micro-vesicle from the cell, wherein the micro-vesicle includes the target protein. Also provided are cells, reagents and kits that find use in making the micro-vesicles, as well as methods of using the micro-vesicles, e.g., in research and therapeutic applications.

Claims (22)

1. A method of preparing a micro-vesicle containing a target protein, the method comprising:

maintaining a cell comprising:

(a) a membrane-associated protein comprising a first dimerization domain;

(b) a heterologous target protein comprising a second dimerization domain; and

(c) a dimerization mediator;

under conditions sufficient to produce a micro-vesicle from the cell, wherein the micro-vesicle contains a dimerized complex of the membrane-associated protein and the heterologous target protein bound to each other by the dimerization mediator; and

separating the micro-vesicle from the cell.

2. The method according to claim 1 , wherein the dimerization mediator is provided in the cell by contacting the cell with the dimerization mediator.

3. The method according to claim 1 , wherein the dimerization mediator is a modifiable dimerization mediator.

4. The method according to claim 3 , wherein the method comprises modifying the dimerization mediator in the micro-vesicle to release the target protein from the complex.

5. The method according to claim 4 , wherein the modifying comprises applying a stimulus.

6. The method according to claim 1 , wherein the cell further comprises a micro-vesicle inducer.

7. The method according to claim 1 , wherein the cell further comprises:

a first expression cassette comprising a first coding sequence for membrane-associated protein comprising a first dimerization domain; and

a second expression cassette comprising a promoter and a second dimerization domain.

8. The method according to claim 7 , wherein at least one of the first and second expression cassettes is under the control of an inducible promoter.

9. The method according to claim 8 , wherein the method further comprises inducing the promoter.

10. The method according to claim 1 , wherein the method further comprises producing the cell.

11. The method according to claim 1 , wherein the cell further comprises a second target protein comprising a third dimerization domain.

12. The method according to claim 1 , wherein the first dimerization domain contacts the cytosol of the micro-vesicle.

13. The method according to claim 1 , wherein binding of the first and second dimerization domains by the dimerization mediator increases the packaging efficiency of the target protein by about 8-fold compared to a control cell in the absence of the dimerization mediator.

14. The method according to claim 1 , wherein the target protein is selected from the group consisting of a genomic modification protein, a cellular reprogramming protein, a transcription factor and a cytosolic protein.
